The general amino acid permease, Gap1, of Saccharomyces cerevisiae is very active in cells grown on proline as the sole nitrogen source. Adding NH4+ to the medium triggers inactivation and degradation of the permease via a regulatory process involving Npi1p/Rsp5p, a ubiquitin–protein ligase. In this study, we describe several mutations affecting the C-terminal region of Gap1p that render the permease resistant to NH4+-induced inactivation. An in vivo isolated mutation ( gap1 pgr  ) causes a single Glu→Lys substitution in an amino acid context similar to the DXKSS sequence involved in ubiquitination and endocytosis of the yeast α-factor receptor, Ste2p. Another replacement, substitution of two alanines for a di-leucine motif, likewise protects the Gap1 permease against NH4+-induced inactivation. In mammalian cells, such a motif is involved in the internalization of several cell-surface proteins. These data provide the first indication that a di-leucine motif influences the function of a plasma membrane protein in yeast. Mutagenesis of a putative phosphorylation site upstream from the di-leucine motif altered neither the activity nor the regulation of the permease. In contrast, deletion of the last eleven amino acids of Gap1p, a region conserved in other amino acid permeases, conferred resistance to NH4+ inactivation. Although the C-terminal region of Gap1p plays an important role in nitrogen control of activity, it was not sufficient to confer this regulation to two NH4+-insensitive permeases, namely the arginine (Can1p) and uracil (Fur4p) permeases.  相似文献

Mechanical injury of the plasmalemma As a possible pre requisite for virus infection of plants
A basic requirement of the infection in case of mechanical inoculation f viruses could be that the plasmalemma of cells is to be injured, for a clearance between cell wall and plasmalemma at the time of inoculation in cells, which are suitable for primary infection prevented the infection.
By means of the hypertonic concentration of 0.7 mol/l of the plasmolytica ethylenglycol, sucrose, glucose, sorbitol and mannitol, which permeate with different speeds, and by modifications of the mannitol treatment, a withdrawal of the plasmalemma to varied distances from the cell wall was induced in cell of the upper side of leaf disk from Nicotiana glutinosa and N tabacum "samsun" This could be observed microscopically in the leaf disks after embedding in liquid paraffin

The discriminative effects of cyclorphan were studied in pigeons trained to discriminate 0.32 mg/kg ethylketazocine, 1.8 mg/kg cyclazocine, or 32 mg/kg naltrexone from saline. A fourth group of pigeons was administered 100 mg/kg/day morphine and trained to discriminate 0.1 mg/kg naltrexone from saline. Cyclorphan produced dose-related ethylketazocine-appropriate responding that reached a maximum of 83% of the total session responses at 0.3 mg/kg. Higher cyclorphan doses produced less ethylketazocine-appropriate responding. In pigeons trained to discriminate cyclazocine from saline, maximum drug-appropriate responding of greater than 90% occured at 5.6–10.0 mg/kg cyclorphan. In narcotic-naive pigeons trained to discriminate 32 mg/kg naltrexone from saline, cyclorphan produced a maximum of less than 50% drug-appropriate responding. In contrast, in pigeons chronically administered morphine and trained to discriminate 0.1 mg/kg naltrexone from saline, 1.0 mg/kg cyclorphan resulted in 100% drug-appropriate responding. In pigeons responding under a multiple fixed-interval, fixed-ratio schedule of food delivery, cyclorphan produced a complete dose-related reversal of the rate-decreasing effects of 10 mg/kg morphine, the maximally effective antagonist doses being 1.0–3.2 mg/kg. Higher cyclorphan doses (10 mg/kg) resulted in response rate decreases that were not reversed by naloxone (1 mg/kg). Thus, cyclorphan has discriminative effects that are similar to those of both ethylketazocine and, at 20-fold higher doses, cyclazocine. In addition, in morphine-treated pigeons, cyclorphan, across the same range of doses that produce ethylketazocine-appropriate responding, has discriminative effects that are similar to those of naltrexone, an effect that is probably related to the antagonist action of the drug.  相似文献

An amino acid, lethal to New Hampshire chickens (LD50, 150 mg/kg) was isolated from dried sclerotia of the fungus Sclerotium rolfsii (Sacc.). Purification of the rather unstable compound was effected on a cation exchange column by means of displacement chromatography and the amino acid was crystallised from 80% methanol. A structure was assigned to the compound on the basis of available chemical and physical data, namely 2(S),3(R)-2- amino-3-hydroxypent-4-ynoic acid. Confirmation of this structure was gained by direct and indirect synthetic procedures.  相似文献

The natural occurrence of sterigmatocystin (S) in in-shell pecans is reported. Aspergillus versicolor was not isolated from contaminated samples. Incidence of A. flavus and A. glaucus, species known to produce sterigmatocystin under laboratory conditions, was high (43 and 35%, respectively). Isolation data suggest sterigmatocystin may have been produced by one or both of these species.  相似文献

The transepithelial resistance of confluent epithelial cell monolayers was monitored to investigate the influence of basic amino acids, Ca2+, protamine and protons on tight junction electrical resistance. In an accompanying paper we investigated the effect of these substances on the lamellar/hexagonal II phase transition in reconstituted phospholipid membranes containing phosphatidylserine and phosphatidylethanolamine. We conclude that the permeability of tight junctions may be described by a lipid phase equilibrium where the lamellar phase corresponds to an open state and the hexagonal lipid phase to the closed state of the cell contact. This dynamic lipid model is well suited to describe the morphological as well as functional properties of the tight junctions.  相似文献

An experiment was designed to compare the "holding power" or "staying power" of absorbable (polyglycolic acid and polyglactin 910) and nonabsorbable (nylon) suture. The aim of this experiment was to determine what provides the lasting strength of the bond between soft tissues that are approximated or plicated. When correcting the rectus diastasis during abdominoplasty, we used nylon sutures in 15 patients and absorbable synthetic sutures in 15 other patients. We then marked the closed folds of the rectus sheath with small metal vascular clips. Two days later and approximately 6 months after operation an upright anteroposterior abdominal x-ray was taken and the position of the metal clips was compared in the test groups. Although there was usually slight separation of the clips after 6 months, no significant difference between the two groups was noted, thereby indicating that holding power is not related to type of suture material but more likely to fibroplasia.  相似文献

Both the hormone dependency and the morphological details of estrogen-dependent ciliogenesis in the shell gland of the chick oviduct were investigated. Ciliogenesis was initiated on day 3 of estrogen treatment, and progressively more cells became differentiated until, on day 10, 55% ciliation occurred with 17-estradiol (1 mg/day) and 75% ciliation occurred with diethylstilbestrol (1 mg/day). Simultaneous administration of progesterone with diethylstilbestrol (1 mg each/day for 10 days) caused a 50% depression in the number of ciliated cells on day 10. The rate of ciliogenesis was found to be affected by progesterone and the type of estrogen administered. The minimum stimulatory dose of estradiol was found to be between 0.01 mg/day and 0.05 mg/day. Ciliogenic cells were first recognized by the appearance of pro-basal bodies in the apical portion of the cell. Pro-basal body maturation and cilium formation were the same as those described for the chick trachea. Ciliogenesis in the chick was found to be homologous to estrogen-dependent ciliogenesis in various mammalian oviducts.  相似文献
