Tissue browning of in vitro cultures of Scots pine: Role of peroxidase and polyphenol oxidase 总被引:7,自引:0,他引:7

Callus cultures from shoot tips of mature Scots pine ( Pinus sylvestris L.) were characterized by rapid browning and an inability to regenerate. The peroxidase (POD) and polyphenol oxidase (PPO) activities and relationship to browning in such cultures were compared with embryogenic and non-embryogenic cultures of Scots pine, started from immature embryos of three different pine clones. The browning in callus cultures derived from pine buds was visible approximately after 2 weeks of culture, and continued thereafter until the callus was dark brown and poorly growing. The non-embryogenic cultures induced from immature embryos showed either light yellow coloring or browning, whereas the embryogenic cultures showed browning. POD activity increased during the first 4 weeks in callus tissue initiated from pine buds, and was significantly higher than in pine buds or cultures derived from immature embryos. The ability of cultures initiated from pine buds to oxidize catechol was notably high compared with cultures initiated from immature embryos, regardless of the time of measurement. Addition of catalase revealed that both POD and PPO were able to use catechol as substrate. An antibody raised against broad bean ( Vicia faba ) chloroplast PPO was used to recognize PPO. One polypeptide with a molecular mass of 50 kDa was detected in all pine samples on SDS-PAGE and non-denaturing PAGE. Another polypeptide with a molecular mass of 70 kDa was shown exclusively in the light-yellow non-embryogenic cultures. The results suggest that especially the high POD activities in callus tissues started from mature trees cause rapid and early browning and possibly subsequent cell death. 相似文献

Res. 1999;7:23–33. Objective : Activity self-reports are a commonly used tool in assessing daily physical activity (PA) and associated energy expenditure (EE). This study examined the effect of relative body fatness (%BF) on differences between self-reported and measured duration and associated EE in healthy adults. Research Methods and Procedures: Men and women (n= 115, age 38±9 years), ranging in %BF from 7.9% to 58.9%, spent two separate days (normal and exercise) in a whole-room indirect calorimeter where EE was measured. While in the room calorimeter, subjects reported the type, intensity, and duration of each performed PA. The Compendium of Physical Activity was used to calculate the energy cost of each reported activity. The EE of all self-reported activities (EEr) was categorized into four intensity levels, synchronized, and compared with EE from the room calorimeter (EEm). Results : With increasing %BF, subjects significantly overestimated duration of more strenuous activities (≥4.5), while underestimating moderate activities (2.5 to 4.4 metabolic equivalents (METs)). Misreporting of duration and/or intensity caused an overestimation or underestimation of PA-associated EE at these levels. Reported EE sleep was lower than measured EE sleep, although both had similar durations. As a result, total EEr was similar to EEm. Discussion : Individual variability of daily total PA and associated EE generated from self-reports in adults is high. Persons with a higher %BF report duration and/or intensity of moderate to high levels of PA with lower accuracy than leaner individuals. We conclude using the Compendium of Physical Activity is not suitable for the accurate estimation of self-reported EE of AA in adults with a higher %BF. 相似文献

Long‐distance migratory passerines may exhibit different post‐fledging rearing times between their early and late broods due to limited time and energy resources. This could affect the migratory fuelling behaviour, condition and fitness of juvenile birds. We analysed long‐term (13‐year) biometric data (body mass, fat score) on Barn Swallows ringed at the nest and later recaptured during their initial stages of autumn migration. The return rates of early‐ and late‐born juveniles after overwintering in Africa were further compared to estimate their fitness. We found that juveniles from late broods started migrating south on average 11 days younger compared with juveniles from early broods. Body masses of nestlings did not differ between early and late broods, but juveniles from late broods were in better condition after leaving the nest, possibly due to a longer period of parental care. Having started migration, juveniles from late broods were, however, less efficient in accumulating energy than juveniles from early broods. A younger departure age, together with less efficient fuelling of late brood juveniles, may partly explain the 39% lower return probability and hence lower fitness value of late‐brood juveniles. Our study is the first to show that juveniles from the early and late broods exhibit different fuelling behaviour and departure strategies (in terms of age and timing), which may be reflected in the fitness value of offspring. 相似文献

We investigated the mechanisms that determine the quality and quantity of organic carbon (C) stocks in boreal forest soils
by analyzing both qualitative and quantitative changes in the organic fractions in the soil organic matter (OM) in a vertical
gradient in the decomposition continuum of the organic horizon [litter layer (L), fermentation layer (F), and humus layer
(H)] in forest soils using a sequential fractionation method at two forest types along a climatic gradient in Finland. We
predicted that the concentrations of water-soluble (WSE) and non-polar (NPE) extractives should decrease and those of the
acid-soluble (AS) fraction and acid-insoluble residue (AIR) should increase from the L to the F, and from the F to the H layers,
but the C/N ratio of soil OM should stay constant after reaching the critical quotient. We also predicted that the AIR concentrations
should be higher in the south than north boreal, and in sub-xeric than mesic forests. Consistent with our hypothesis, the
concentrations of WSE and NPE fractions decreased and concentrations of AIR increased in the vertical soil gradient. The highest
concentrations of the AS fraction were found in the F layer. The C/N ratio was lowest in the F layer, and the highest in the
H layer, indicating that soil OM is depleted in N in relation to C along the vertical soil gradient. Concentrations of WSE
and NPE were lower, and concentrations of AIR were higher in the south than in north boreal forests, which is in agreement
with our hypothesis that higher soil temperatures may enhance accumulation of slowly decomposable OM in the soil. The concentrations
of AIR were higher in the sub-xeric than mesic forests. Contrary to our expectations, however, the differences in the chemical
quality in soil OM between the site types were amplified from the L to the H layer. The size of the C storage was significantly
larger in south than north boreal sites, and larger in the mesic than in the sub-xeric sites. 相似文献

maturity at 3 years old. C. tarichi is an diadrom fish that lives in the lake, but during the reproduction period it immigrates to the surrounding freshwater
rivers returning after the reproduction period of April–July. In the past, pearl mullet was an attractive fresh fish for the
local people and was easily caught during the spawning migration, resulting in over-fishing. The species was one of the highly
endangered animals of Turkey before conservation studies, some 10 years ago, have started. At present, illegal fishing activities

The NAD(P)H dehydrogenase (NDH) complex functions in photosystem I cyclic electron transfer in higher plant chloroplasts and is crucial for plant responses to environmental stress. Chloroplast NDH complex is a close relative to cyanobacterial NDH-1L complex, and all fifteen subunits so far identified in NDH-1L have homologs in the chloroplast NDH complex. Here we report on the identification of two nuclear-encoded proteins NDH48 and NDH45 in higher plant chloroplasts and show their intimate association with the NDH complex. These two membrane proteins are shown to interact with each other and with the NDH complex enriched in stroma thylakoids. Moreover, the deficiency of either the NDH45 protein or the NDH48 protein in respective mutant plants leads to severe defects in both the accumulation and the function of the NDH complex. The NDH48 and NDH45 proteins are not components of the hydrophilic connecting domain of the NDH complex but are strongly attached to the hydrophobic membrane domain. We conclude that NDH48 and NDH45 are novel nuclear-encoded subunits of the chloroplast NDH complex and crucial both for the stable structure and function of the NDH complex. 相似文献
