We have reported that a second rat atrial natriuretic peptide, iso-rANP (1-45), as well as the putative ANP homologue, iso-rANP (17-45), elicited circulatory and renal responses in the rat similar to those found after administration of ANP. Iso-rANP also interacted with ANP to potentiate the observed biological activity in the rat. In the present studies in awake dogs, intravenous infusion of low doses (6.3-50 pmol.kg-1.min-1) of iso-rANP(1-45) and iso-rANP(17-45) increased plasma immunoreactive ANP and suppressed plasma renin activity (PRA) and aldosterone. Iso-rANP, like ring-deleted analogues of ANP, may have displaced ANP from ANP clearance receptors to increase plasma ANP concentration, since factors influencing myocardial ANP release were not changed. The effect of iso-rANP (1-45) and (17-45) in lowering PRA and plasma aldosterone may therefore have been indirect, via ANP stimulation of active guanylate cyclase-linked ANP receptors. However, an additional direct effect of iso-rANP on an active receptor cannot be excluded.  相似文献

The development of automated methods and normative rules, as well as the dependence of some therapeutic approaches on lymphocyte subsets counts, has led to the appearance of calibration reagents. Such reagents are expected to perform equally well in very different settings. We developed a multicenter trial to evaluate the performance of a new quality control reagent, i.e., stabilized blood to be used in immunophenotyping laboratories. Aliquots of the same batch of stabilized blood were shipped to 45 French and Belgian laboratories on a Monday and had to be tested for percentages and absolute counts of at least CD3-, CD4-, and CD8-positive lymphocytes on 4 consecutive days. The percentages and absolute counts obtained on each assay were recorded, as well as the type of lysis used, the trademark of reagents, and the brand of flow cytometer. The mean values collected did not differ significantly from those expected by the manufacturer. Absolute counts generated through one-step techniques displayed lower CVs. This new reagent therefore appears to be a robust product, liable to yield consistent results in the array of different conditions represented by routine laboratories, and it could be useful for quality control procedures.  相似文献

Comparison of the primary structures of pancreatic colipases from man, pig, horse and rat shows a high degree of homology between proteins. Fifty-two out of the 95 residues of the polypeptide are identical. All colipases contain 10 half-cystines which are located at invariant positions. The secondary structure of colipases has been predicted from the sequence using the statistical method of Chou and Fasman and the method of Gibrat, Garnier and Robson based on information theory. Predictions indicate that colipases have a low content of alpha-helix and beta-strand structure. The two segments at positions 7-10 and 56-59, assumed to be part of the lipid binding domain, have predicted beta-sheet conformation and should be in close spatial vicinity to each other in the proteins. Four beta-turns are predicted in all colipases at positions 3-6, 46-49, 61-64, and 81-84. They might contribute, with the five disulfide bridges, to a tight packing of the protein molecule. Surface residues and major sequential antigenic determinants of mammalian colipases have been predicted using methods based either on hydrophilicity/hydropathy scales or amino acid mutability. From these studies, it appears that colipases exhibit large conformational homologies. In the absence of data on the tertiary structure of colipase, predictive methods, together with physico-chemical and immunological studies, provide valuable information on the conformation of the protein in relation to the topology of residues involved in the functional and antigenic sites.  相似文献
