Linoleic acid peroxyl radicals (LOO.) can be viewed as model intermediates occurring during lipid peroxidation processes. Formation and reactions of these species were investigated in aqueous alkaline solution using the technique of pulse radiolysis combined with kinetic spectroscopy. Irradiation of linoleic acid in N2O/O2-saturated solutions leads to a mixture of peroxyl radical isomers, whereas reaction of 13-hydroperoxylinoleic acid (13-LOOH) with azide radicals in N2O-saturated solution produces 13-LOO. radicals specifically. These peroxyl radicals cannot be observed directly, but their reactions with the two flavonols, kaempferol and quercetin, acting as radical-scavenging antioxidants, produced strongly absorbing aroxyl radicals (ArO.). The same aroxyl radicals were generated by .OH and N3. with rate constants exceeding 10(9) dm3 mol-1 s-1. Applying a reaction scheme that includes competing generation and decay reactions of both LOO. and ArO. radicals, we derived individual rate constants for LOO. reactions with the phenols (greater than 10(7) dm3 mol-1 s-1), with the aroxyl radicals to form covalent adducts (greater than 10(8) dm3 mol-1 s-1), as well as for their bimilecular decay (3.0 X 10(8) dm3 mol-1 s-1). These results demonstrate the high reactivity of both fatty acid peroxyl radicals and the flavone antioxidants in aqueous solution.  相似文献

To quantify the demographic effects of food quality, and specificallyof the ‘poorquality’ cyanobecterium Anabaena flos-aquae,we reared individual Boeckella triarticulata (Copepoda, Calanoida)on two diets (monospecific Cryptomom sp. versus mixed Cryptomonas-Anabanmadiets) and quantified individual growth and developmental trajectoriesby examining exuviae produced at each molt, from hatching tomaturity. Size at molting was less variable among individuals,within and between diets, than age. Food quality had significanteffects on male sizes at molting and on stage-specific dailygrowth rates of both sexes; these effects were strongest duringlate naupliar and all copepodite stages Tke med Cryptomonas-Anabaenadiet significantly slowed development, particularly of copepoditestages. As a consequence of these effects, individuals raisedon the mixed diet were smaller and older at maturity. Withina given diet, individual differences explained much, if notmost, of the variation exhibited in growth and development.By following growth and development of a large number of individualsthroughout their Life cycles, we show that individual femalesproduce variable offspring, indicative of a bet-hedging life-historystrategy, and that B.triarticulata (like other calanoids) cangrow, develop and survive on diets that include ‘poorquality’ cyanobacteria. 1Present address: Department of Biological Sciences, Universityof Rhode Island, Kingston, RI 02881, USA  相似文献

A 355 base pair DNA sequence coding for human preproinsulin has been assembled by joining 55 synthetic deoxyoligonucleotide fragments prepared by the modified phosphotriester methodology. Proinsulin was expressed underlac promoter control and truncatedΒ-galactosidase 590 amino acid long sequence. The fusedΒ-galactosidase proinsulin protein was produced in amount to 30 % of the totalEscherichia coli proteins. It was also expressed in M13 bacteriophage and yeast system.  相似文献

Summary Generation of radicals in vivo depends on metabolic activities. The reactions are usually influenced by(i) the presence and concentration of oxygen;(ii) the availability of transition metals (effects of binding and compartimentalization);(iii) the level of reductants and antioxidants (e.g. nutritional effects). The effects of radicals are thought to be due to(i) membrane damage (affecting passive or active transport through altered fluidity/function interrelationships, intercellular messenging through modifications in the synthesis of prostaglandins and leukotrienes);(ii) protein damage (e.g. affecting membrane transporters, channel proteins, receptor or regulatory proteins, immunomodulators);(iii) damage to DNA. Defense mechanisms consist of(i) prevention of the spreading of primary damage by low molecular weight antioxidants (e.g. vitamin E, GSH, vitamin C,-carotene, uric acid);(ii) prevention or limitation of secondary damage by enzymes (e.g. GSH-peroxidase, catalase, superoxide dismutase, DT-diaphorase) and/or chelators;(iii) repair processes, e.g. lipid degradation/membrane repair enzymes (phospholipases, peroxidases, some transferases and reductases), protein disposal or repair enzymes (proteases, GSSG-reductase), DNA degradation repair enzymes (exonuclease III, endonucleases III and IV, glycosylases, polymerases). Recent hypotheses on a messenging function of the superoxide anion O 2 are discussed and possible implications of cross-reactions between O 2 and nitric oxide (endothelium-derived relaxing factor EDRF) are shortly mentioned.Paper given at the workshop Molecular Radiation Biology. This is the first report correlating levels of polyamines and its fractions with differentiation in Dictyostelium discoideum. Temporal changes in endogenous levels of free, conjugated and bound putrescine, spermidine and spermine were analysed at critical stages of morphogenesis in this organism. No spermine was found at any given stage and putrescine was the most abundant polyamine. There was a sharp increase in the levels of both free (and total) and conjugated forms of putrescine and spermidine at the slug stage as compared to the growth phase. The levels of putrescine and spermidine were found to be higher in isolated prespore cells as compared to the prestalk cells. Remarkably, the levels of polyamine decreased at the early culminant stage. Data suggest that a moderate level of polyamines is needed for growth but it is important to have high levels of polyamines at the time of differentiation.  相似文献

Better breeding strategies for captive Asian elephants in range countries are needed to increase populations; this requires a thorough understanding of their reproductive physiology and factors affecting ovarian activity. Weekly blood samples were collected for 3.9 years from 22 semi-captive female Asian elephants in Thai elephant camps to characterize LH and progestin patterns throughout the estrous cycle. The duration of the estrous cycle was 14.6+/-0.2 weeks (mean+/-S.E.M.; n=71), with follicular and luteal phases of 6.1+/-0.2 and 8.5+/-0.2 weeks, respectively. Season had no significant effect on the overall length of the estrous cycle. However, follicular and luteal phase lengths varied among seasons and were negatively correlated (r=-0.658; P<0.01). During the follicular phase, the interval between the decrease in progestin concentrations to baseline and the anovulatory LH (anLH) surge varied in duration (average 25.9+/-2.0 days, range 7-41, n=23), and was longer in the rainy season (33.4+/-1.8 days, n=10) than in both the winter (22.2+/-4.5 days, n=5; P<0.05) and summer (18.9+/-2.6 days, n=8; P<0.05). By contrast, the interval between the anLH and ovulatory LH (ovLH) surge was more consistent (19.0+/-0.1 days, range 18-20, n=14). Thus, seasonal variation in estrous cycle characteristics were mediated by endocrine events during the early follicular phase, specifically related to timing of the anLH surge. Overall reproductive hormone patterns in Thai camp elephants were not markedly different from those in western zoos. However, this study was the first to more closely examine how timing of the LH surges impacted estrous cycle length in Asian elephants. These findings, and the ability to monitor reproductive hormones in range countries (and potentially in the field), should improve breeding management of captive and semi-wild elephants.  相似文献

Fine needle aspiration cytology in the diagnosis of tuberculous mastitis   总被引:2,自引:0,他引:2  
OBJECTIVE: To study the relationship between granulomas in the breast and tuberculous mastitis. STUDY DESIGN: Retrospective analysis of 22 breast aspirates that showed epithelioid cell granulomas. The aspirates were reviewed and the cytomorphologic findings summarized. RESULTS: Aspiration cytology revealed epithelioid cell granulomas along with giant cells, necrosis and inflammatory cell infiltrate. Overall acid-fast bacillus (AFB) positivity was 22.7%. AFB positivity was greater in the presence of necrosis when epithelioid cells were absent. CONCLUSION: In a country like India, the diagnosis of granulomatous mastitis must be made with caution, even in the absence of AFB. Only after a sufficient trial of antituberculosis treatment has been given and the patient fails to respond should an alternative diagnosis be suggested.  相似文献
