Captive Asian elephant (Elephas maximus) populations are decreasing due to low birth rates compared to wild elephants. Improving oestrous detection in female elephants is required to ensure successful mating in captive and semi-captive herds. Responsive behaviours of eight semi-captive bull elephants to the uro-genital area (genital inspection test) or urinary pheromones (urine test) of 14 female elephants throughout the oestrous cycle were evaluated. Weekly blood samples were collected for 27 consecutive months (14 months for the genital inspection test and 13 months for the urine test) from female elephants to characterize the patterns of circulating progestagen. Responsive behaviours of bulls were compared between females in the follicular versus the luteal phase of the cycle. The sensitivity and specificity of the genital inspection test were 65% and 68%, while those of the urine test were 52% and 61%, respectively. The bulls showed significantly higher “genital inspection”, “flehmen from genital area” and “trunk on back” behaviours during the genital inspection test, and “flehmen” behaviours during the urine test in oestrous than in non-oestrous females. In sum, this study showed that monitoring sexual behaviours of Asian elephant bulls towards females or their urine can be used to detect the oestrous period. Although the sensitivity and specificity of both tests were not as high as expected, still, these methods appear to be more efficient at detecting oestrous than traditional methods based on mahout estimations of female receptivity. The use of genital inspection and urine tests may lead to more successful matings and thus to creating self-sustaining populations of captive elephants in range countries.  相似文献

The insecticidial and biological activity of the cyano-substituted neonicotinoid acetamiprid was determined against the western subterranean termite, Reticulitermes hesperus Banks (Isoptera: Rhinotermitidae). Acetamiprid was very active against termites in topical applications, with an LD50 = 0.02 ng per termite. Even though acetamiprid was extremely toxic in topical applications, deposits > or =50 ppm on sand were required to consistently provide >90% kill of termites within 7 d after a 1-h exposure. Termites were quickly affected by brief exposures to sand treated with 1 ppm acetamiprid and within 1 h, their locomotion was dramatically impaired. Acetamiprid was transferred from donors to recipients only when donors were held on deposits > or =50 ppm for 1 h. Deposits even as low as 1 ppm were repellent and termites failed to tunnel into treated sand, and there was no significant mortality. Exposure to acetamiprid impaired locomotion of termites as did other slow-acting neonicotinoids, such as imidacloprid. Acetamiprid was repellent at all concentrations tested, acting like type I pyrethroid treatments in soil. A new subcategory of type III soil termiticides is proposed that incorporates the sublethal and delayed effects observed in neonicotinoid insecticides, and repellency at certain concentrations.  相似文献

Three genes coding for xylanase synthesis in Clostridium thermocellum were cloned and expressed in Escherichia coli. Genomic DNA from Clostridium thermocellum was digested to completion with HindIII, BamHI, and SalI. The fragments were ligated into the corresponding sites of pUC19 and transformed into Escherichia coli. Two of the genes encoded for xylanases which depolymerized xylans but were unable to extensively convert these substrates to reducing sugar. The third gene encoded for an enzyme that extensively hydrolyzed xylan. The insert containing the latter gene was subjected to extensive mapping and was found to encode for a xylanase with a molecular weight of approximately 25,000. The protein product of the cloned gene was obtained in a relatively pure form by heat treatment, ion exchange and gel permeation steps. Transient spectra and kinetic data of Tiron (1,2-dihydroxybenzene-3,5-disulphonic acid) are reported, obtained after pulse-radiolytic oxidation by hydroxyl radicals (°OH), superoxide anions (O2?) or a combination of both oxygen radicals. The rate constant with °OH radicals was determined at 1.0·109 M?1·s?1. Contrary to a previous report (Greenstock, C.L. and Miller, R.W. (1975) Biochim. Biophys. Acta 396, 11–16), the rate constant with O2? of 1.0·107 M?1·s?1 is lower by one order of magnitude; also the semiquinone absorbs at 300 nm rather than at 400 nm. The ratio of the rate constants with °OH and O2? of 100 again demonstrates that any oxidation reaction by the latter radical is unspecific due to the more efficient reaction of °OH radicals, leading to the same products with catechol compounds.  相似文献

The pulse-radiolytic oxidation of hydroxylamine by either hydroxyl radicals (OH), superoxide anions (O2?), or a combination of both radicals was investigated. It was found that only OH radicals efficiently attack the substrate, while O2? is necessary for the subsequent formation of nitrite. Determination of the latter reaction thus allows the detection of the combined presence of both oxygen radical species.  相似文献
