A biogenic route was adopted towards the synthesis of gold nanoparticles using the extract of a novel strain, Talaromyces flavus. Reduction of chloroauric acid by the fungal extract resulted in the production of gold nanoparticle, which was further confirmed by the concordant results obtained from UV–visible spectroscopy, energy dispersive spectroscopy (EDS), and dynamic light scattering (DLS) analysis. Morphology and the crystal nature of the synthesized nanoparticles were characterized using transmission electron microscopy (TEM), X-ray diffraction (XRD) and selected area electron diffraction (SAED). A direct correlation was observed between nanoparticle formation and the concentration of reducing agent present in the fungal extract. The time-dependent kinetic study revealed that the bioreduction process follows an autocatalytic reaction. Crystalline, irregular, and mostly flower-shaped gold nanoparticles with a mean hydrodynamic radius of 38.54?±?10.34 nm were obtained. pH played a significant role on production of mono-dispersed nanoparticle. FTIR analysis partially deciphered the involvement of –NH2, ?SH, and –CO groups as the probable molecules in the bio-reduction and stabilization process. Compared to the conventional methods, a time-resolved, green, and economically viable method for floral-shaped nanoparticle synthesis was developed.  相似文献

The present communication reports an endangered catfish of the family Siluridae, Pterocryptis barakensis (Vishwanath & Sharma) for the first time outside its type locality from Brahmaputra drainage. The specimens were collected from the River Tissa, Longding district, Arunachal Pradesh foothills of Himalayan region of North East India. The influence of phytohormones, salicylic acid (SA) and methyl jasmonate (MJ) on the antioxidant systems in Haematococcus pluvialis was investigated. Both SA and MJ at 500 μM concentration reduced the growth of alga with salicylic acid, having more pronounced effect. Carotenoid and chlorophyll contents were decreased by SA and increased by MJ. Salicylic acid (100 μM) increased astaxanthin content to 6.8-fold under low light (30 μmol m−2 s−1), while MJ (10 μM) showed marginal increase in astaxanthin. Salicylic acid (500 μM) increased superoxide dismutase activity to 4.5- and 3.3-fold and ascorbate peroxidase (APX) activity to 15.5- and 7.1-fold under low and high light, respectively. Methyl jasmonate increased catalase activity (1.4-fold) under high light and APX activity (5.4-fold) under low light. Different mechanism of oxidative stress induced antioxidant production may be the plausible reason for this varied response for salicylic acid and methyl jasmonate. Higher concentrations of SA and MJ inhibited astaxanthin accumulation by different mechanisms either by scavenging the free radicals or by increasing primary carotenoids production. At lower concentrations, these phytohormones could be used for elicitation of secondary carotenoid production.  相似文献

A novel zinc transporter has been purified and cloned from rat renal brush border membrane. This transporter was designated as Zip10 encoded by Slc39a10 gene and characterized as zinc importer. Present study documents the impact of thyroid hormones on the expression of Zip10 encoded by Slc39a10 gene in rat model of hypo and hyperthyroidism. Serum T(3) and T(4) levels were reduced significantly in hypothyroid rats whereas these levels were significantly elevated in hyperthyroid rats as compared to euthyroid rats thereby confirming the validity of the model. Kinetic studies revealed a significant increase in the initial and equilibrium uptake of Zn(++) in both intestinal and renal BBMV of hyperthyroid rats in comparison to hypothyroid and euthyroid rats. By RT-PCR, Slc39a10 mRNA expression was found to be significantly decreased in hypothyroid and increased in hyperthyroid as compared to euthyroid rats. These findings are in conformity with the immunofluorescence studies that revealed markedly higher fluorescence intensity at periphery of both intestinal and renal cells isolated from hyperthyroid rats as compared to hypothyroid and euthyroid rats. Higher expression of Zip10 protein in hyperthyroid group was also confirmed by western blot. These findings suggest that expression of zinc transporter protein Zip10 (Slc39a10) in intestine and kidney is positively regulated by thyroid hormones.  相似文献

Propolis is a natural substance collected by honey bees from various plants such as, poplar, palm, pine, conifer secretions, gums, resins, mucilage and leaf buds. It is collected and brought very painstakingly by honey bees to be used for sealing cracks and crevices occurring in their hives. Originally, it as an antiseptic meant for preventing bee-hive from microbial infections along with preventing decomposition of intruders. Additionally, propolis has been used in folk medicine for centuries. The biological characteristics of propolis depend upon its chemical composition, plant sources, geographical zone and seasons. More than 300 compounds have been identified in propolis such as, phenolic compounds, aromatic acids, essential oils, waxes and amino acids. Recombinant transmembrane adenylate cyclase (AC) was incorporated into membranes of giant liposomes using membrane fusion between liposomes and baculovirus-budded virus (BV). AC genes were constructed into transfer vectors in a form fused with fluorescent protein or polyhistidine at the C-terminus. The recombinant BVs were collected by ultracentrifugation and AC expression was verified using western blotting. The BVs and giant liposomes generated using gentle hydration were fused under acidic conditions; the incorporation of AC into giant liposomes was demonstrated by confocal laser scanning microscopy through the emission of fluorescence from their membranes. The AC-expressing BVs were also fused with liposomes containing the substrate (ATP) with/without a specific inhibitor (SQ 22536). An enzyme immunoassay on extracts of the sample demonstrated that cAMP was produced inside the liposomes. This procedure facilitates direct introduction of large transmembrane proteins into artificial membranes without solubilization.  相似文献

The stress-related corticotropin-releasing hormone (CRH) was first identified by isolation of its cDNA from the brain of the Japanese eel Anguilla japonica. CRH cDNA encodes a signal peptide, a cryptic peptide and CRH (41 amino acids). The sequence homology to mammalian CRH is high. Next, the distribution of CRH-immunoreactive (ir) cell bodies and fibers in the brain and pituitary were examined by immunohistochemistry. CRH-ir cell bodies were detected in several brain regions, e.g., nucleus preopticus pars magnocellularis, nucleus preopticus pars gigantocellularis and formatio reticularis superius. In the brain, CRH-ir fibers were distributed not only in the hypothalamus but also in various regions. Some CRH-ir fibers projected to adrenocorticotropic hormone (ACTH) cells in the rostral pars distalis of the pituitary and also the α-melanocyte-stimulating hormone (α-MSH) cells in the pars intermedia of the pituitary. Finally, the neuroanatomical relationship between the CRH neurons and gonadotropin-releasing hormone (GnRH) neurons was examined by dual-label immunohistochemistry. CRH-ir fibers were found to be in close contact with GnRH-ir cell bodies in the hypothalamus and in the midbrain tegmentum and GnRH-ir fibers were in close contact with CRH-ir cell bodies in the nucleus preopticus pars magnocellularis. These results suggest that CRH has some physiological functions other than the stimulation of ACTH and α-MSH secretion and that reciprocal connections may exist between the CRH neurons and GnRH neurons in the brain of the Japanese eel.  相似文献
