Class I Major Histocompatibility Complex (MHC) molecules evolved to sample degraded protein fragments from the interior of the cell, and to display them at the surface for immune surveillance by CD8+ T cells. The ability of these lymphocytes to identify immunogenic peptide-MHC (pMHC) products on, for example, infected hepatocytes, and to subsequently eliminate those cells, is crucial for the control of hepatitis B virus (HBV). Various protein scaffolds have been designed to recapitulate the specific recognition of presented antigens with the aim to be exploited both diagnostically (e.g. to visualize cells exposed to infectious agents or cellular transformation) and therapeutically (e.g. for the delivery of drugs to compromised cells). In line with this, we report the construction of a soluble tetrameric form of an αβ T cell receptor (TCR) specific for the HBV epitope Env183–191 restricted by HLA-A*02:01, and compare its avidity and fine-specificity with a TCR-like monoclonal antibody generated against the same HLA target. A flow cytometry-based assay with streptavidin-coated beads loaded with Env183–191/HLA-A*02:01 complexes at high surface density, enabled us to probe the specific interaction of these molecules with their cognate pMHC. We demonstrate that the TCR tetramer has similar avidity for the pMHC as the antibody, but they differ in their fine-specificity, with only the TCR tetramer being capable of binding both natural variants of the Env183–191 epitope found in HBV genotypes A/C/D (187Arg) and genotype B (187Lys). Collectively, the results highlight the promiscuity of our soluble TCR, which could be an advantageous feature when targeting cells infected with a mutation-prone virus, but that binding of the soluble oligomeric TCR relies considerably on the surface density of the presented antigen. 相似文献

Regions of homozygosity (ROHs) are more abundant in the human genome than previously thought. These regions are without heterozygosity,
i.e. all the genetic variations within the regions have two identical alleles. At present there are no standardized criteria
for defining the ROHs resulting in the different studies using their own criteria in the analysis of homozygosity. Compared
to the era of genotyping microsatellite markers, the advent of high-density single nucleotide polymorphism genotyping arrays
has provided an unparalleled opportunity to comprehensively detect these regions in the whole genome in different populations.
Several studies have identified ROHs which were associated with complex phenotypes such as schizophrenia, late-onset of Alzheimer’s
disease and height. Collectively, these studies have conclusively shown the abundance of ROHs larger than 1 Mb in outbred
populations. The homozygosity association approach holds great promise in identifying genetic susceptibility loci harboring

The purpose of this study was to explore the use of molecular bio-imaging systems and biomechanical dynamics to elucidate the fate of a nanocomposite hydrogel system prepared by merging FITC-labeled nanolipobubbles within a cross-linked hydrogel network. The nanocomposite hydrogel system was characterized by size distribution analysis and zeta potential as well as shears thinning behavior, elastic modulus (G’), viscous loss moduli (G”), TEM, and FTIR. In addition, molecular bio-imaging via Vevo ultrasound and Cell-viZio techniques evaluated the stability and distribution of the nanolipobubbles within the cross-linked hydrogel. FITC-labeled and functionalized nanolipobubbles had particle sizes between 135 and 158 nm (PdI?=?0.129 and 0.190) and a zeta potential of ?34 mV. TEM and ultrasound imaging revealed the uniformity and dimensional stability of the functionalized nanolipobubbles pre- and post-embedment into the cross-linked hydrogel. Biomechanical characterization of the hydrogel by shear thinning behavior was governed by the polymer concentration and the cross-linker, glutaraldehyde. Ultrasound analysis and Cell-viZio bio-imaging were highly suitable to visualize the fluorescent image-guided nanolipobubbles and their morphology post-embedment into the hydrogel to form the NanoComposite system. Since the nanocomposite is intended for targeted treatment of neurodegenerative disorders, the distribution of the functionalized nanolipobubbles into PC12 neuronal cells was also ascertained via confocal microscopy. Results demonstrated effective release and localization of the nanolipobubbles within PC12 neuronal cells. The molecular structure of the synthetic surface peptide remained intact for an extended period to ensure potency for targeted delivery from the hydrogel ex vivo. Aim Twelve per cent of the Earth’s terrestrial surface is covered by protected areas, but neither these areas nor the biodiversity they contain are evenly distributed spatially. To guide future establishment of protected areas, it is important to understand the factors that have shaped the spatial arrangement of the current protected area system. We used an information‐theoretic approach to assess the ability of vertebrate biodiversity measures, resource consumption and agricultural potential to explain the global coverage pattern of protected areas. Location Global. Methods For each of 762 World Wildlife Fund terrestrial ecoregions of the world, we measured protected area coverage, resource consumption, terrestrial vertebrate species richness, number of endemic species, number of threatened species, net primary production, elevation and topographic heterogeneity. We combined these variables into 39 a priori models to describe protected area coverage at the global scale, and for six biogeographical realms. Using the Akaike information criterion and Akaike weights, we identified the relative importance and influence of each variable in describing protected area coverage. Results Globally, the number of endemic species was the best variable describing protected area coverage, followed by the number of threatened species. Species richness and resource consumption were of moderate importance and agricultural potential had weak support for describing protected area coverage at a global scale. Yet, the relative importance of these factors varied among biogeographical realms. Measures of vertebrate biodiversity (species richness, endemism and threatened species) were among the most important variables in all realms, except the Indo‐Malayan, but had a wide range of relative importance and influence. Resource consumption was inversely related to protected area coverage across all but one realm (the Palearctic), most strongly in the Nearctic realm. Agricultural potential, despite having little support in describing protected area coverage globally, was strongly and positively related to protection in the Palearctic and Neotropical realms, as well as in the Indo‐Malayan realm. The Afrotropical, Indo‐Malayan and Australasian realms showed no clear, strong relationships between protected area coverage and the independent variables. Main conclusions Globally, the existing protected area network is more strongly related to biodiversity measures than to patterns of resource consumption or agricultural potential. However, the relative importance of these factors varies widely among the world’s biogeographical realms. The following study evaluates the overt toxic potential of carprofen (CRP), flunixin (FXN) and phenylbutazone (PBZ) in Old world vultures in relation to historic toxicity data for diclofenac and ketoprofen, with the Cape vulture (Gyps coprotheres) being the indicator species. The toxic potential of a single oral dose of CRP (11.5 mg/kg), FXN (1 mg/kg),PBZ (1.7 mg/kg) or water was evaluated by means of a four-way parallel study (n = 2), as means of ascertaining if these drugs were as toxic as diclofenac in the vulture. No unscheduled deaths or pathological lesions were noted following exposure. Clinical signs of lethargy and depression were, however, noted in one CRP, two FXN and one PBZ treated birds. Mild reversible inhibition of UA excretion was evident in all three groups, although UA remained within the population reference interval in contrast to the effects previously described for diclofenac and ketoprofen. All treatment groups had a drug concentration responsive increase in alanine transferase activity. CRP, FXN and PBZ were characterised by a maximum plasma concentration (Cmax) of 1051.8 ± 620.7 ng/ml, 335.9 ± 36.3 ng/ml and 11150 ± 2474.9 ng/ml at 4 ± 4.3, 0.45 ± 0.02 and 5.3 ± 5.2 hours (Tmax) respectively and a half-life of elimination of 13.3 ±5, 1.8±1 and 18.7 ±11.4 hours respectively. While we could not demonstrate a lethal effect of the tested substances, the presence of toxic clinical signs, clinical pathological changes and/or long half-lives of elimination suggests that all three drugs have a potential for toxicity in a larger population or on repeat administration. In conclusion while the studied substances were not as overtly toxic as diclofenac, they are of safety concern. 相似文献
