The fluorescence depolarization of 1,6-diphenyl-hexatriene was used to study the dynamic properties of the hydrophobic regions of the lipid envelopes of ortho- and paramyxoviruses as well as of the Rous sarcoma virus and of the membrane lipids of susceptible and nonsusceptible cells.The systems investigated where active and inactive influenza viruses, and NDV virus acting on chick embryo fibroblasts and Rous sarcoma virus acting on susceptible (C/E) and nonsusceptible (C/B) chicken-cell.Polarization degrees and mean rotational correlation times of DPH embedded in viral lipids were significantly higher than those of DPH in the cell membranes, due to a higher rigidity of the virus envelopes. When suspensions of labelled viruses and unlabelled cells or unlabelled viruses and labelled cells were mixed, a characteristic change of the fluorescence polarization degrees with time was observed. This behaviour was ascribed to a label transfer from virus to cell membranes or vice versa.While the rate constants of label transfer from virus to cells and cells to virus were about the same for the penetrating viruses the rate constants of label release from inactive virus to cells were much larger than for the migration in the opposite direction.  相似文献

The main potential of intrinsically fluorescent proteins (IFPs), as noninvasive and site-specific markers, lies in biological applications such as intracellular visualization and molecular genetics. However, photophysical studies of IFPs have been carried out mainly in aqueous solution. Here, we provide a comprehensive analysis of the intracellular environmental effects on the steady-state spectroscopy and excited-state dynamics of green (EGFP) and red (DsRed) fluorescent proteins, using both one- and two-photon excitation. EGFP and DsRed are expressed either in the cytoplasm of rat basophilic leukemia (RBL-2H3) mucosal mast cells or anchored (via LynB protein) to the inner leaflet of the plasma membrane. The fluorescence lifetimes (within approximately 10%) and spectra in live cells are basically the same as in aqueous solution, which indicate the absence of both IFP aggregation and cellular environmental effects on the protein folding under our experimental conditions. However, comparative time-resolved anisotropy measurements of EGFP reveal a cytoplasmic viscosity 2.5 +/- 0.3 times larger than that of aqueous solution at room temperature, and also provide some insights into the LynB-EGFP structure and the heterogeneity of the cytoplasmic viscosity. Further, the oligomer configuration and internal depolarization of DsRed, previously observed in solution, persists upon expression in these cells. DsRed also undergoes an instantaneous three-photon induced color change under 740-nm excitation, with efficiently nonradiative green species. These results confirm the implicit assumption that in vitro fluorescence properties of IFPs are essentially valid for in vivo applications, presumably due to the beta-barrel protection of the embodied chromophore. We also discuss the relevance of LynB-EGFP anisotropy for specialized domains studies in plasma membranes.  相似文献

We investigated the effect of weight loss, independent of change in diet composition, on HDL and apoAI metabolism in men with metabolic syndrome (MetS). Subjects (19 men with MetS [NCEP-ATPIII]) were fed an isoenergetic Mediterranean-style diet for 5 weeks (all foods provided). Participants then underwent a 20-week free-living period during which they were counseled to restrict energy intake, after which they were again fed an isoenergetic Mediterranean-style diet for 5 weeks. At the end of the two controlled diets, participants received a single bolus of [5,5,5-2H3] L-leucine, and fasting blood samples were collected over a 96 h period. ApoAI kinetic was assessed using multicompartmental modeling of the tracer enrichment data. Participants achieved a 9.1 ± 2.8% reduction in body weight (P < 0.001). Weight loss resulted in an increase in plasma HDL-cholesterol (HDL-C) concentrations of 6.0% (P = 0.059) and HDL3-C of 7.9% (P = 0.045), attributable to a reduction in apoAI fractional catabolic rate (−7.8%; P = 0.046) with no change in apoAI production rate (2.2%; P = 0.58). These data indicate that weight loss, independent of variation in diet composition, increases plasma HDL primarily by delaying the catabolism of apoAI.  相似文献

Although current influenza vaccines are effective in general, there is an urgent need for the development of new technologies to improve vaccine production timelines, capacities and immunogenicity. Herein, we describe the development of an influenza vaccine technology which enables recombinant production of highly efficient influenza vaccines in bacterial expression systems. The globular head domain of influenza hemagglutinin, comprising most of the protein''s neutralizing epitopes, was expressed in E. coli and covalently conjugated to bacteriophage-derived virus-like particles produced independently in E.coli. Conjugate influenza vaccines produced this way were used to immunize mice and found to elicit immune sera with high antibody titers specific for the native influenza hemagglutinin protein and high hemagglutination-inhibition titers. Moreover vaccination with these vaccines induced full protection against lethal challenges with homologous and highly drifted influenza strains.  相似文献

Snow cover is a key environmental component for tundra wildlife that will be affected by climate change. Change to the snow cover may affect the population dynamics of high‐latitude small mammals, which are active throughout the winter and reproduce under the snow. We experimentally tested the hypotheses that a deeper snow cover would enhance the densities and winter reproductive rates of small mammals, but that predation by mustelids could be higher in areas of increased small mammal density. We enhanced snow cover by setting out snow fences at three sites in the Canadian Arctic (Bylot Island, Nunavut, and Herschel Island and Komakuk Beach, Yukon) over periods ranging from one to four years. Densities of winter nests were higher where snow depth was increased but spring lemming densities did not increase on the experimental areas. Lemmings probably moved from areas of deep snow, their preferred winter habitat, to summer habitat during snow melt once the advantages associated with deep snow were gone. Our treatment had no effect on signs of reproduction in winter nests, proportion of lactating females in spring, or the proportion of juveniles caught in spring, which suggests that deep snow did not enhance reproduction. Results on predation were inconsistent across sites as predation by weasels was higher on the experimental area at one site but lower at two others and was not higher in areas of winter nest aggregations. Although this experiment provided us with several new insights about the impact of snow cover on the population dynamics of tundra small mammals, it also illustrates the challenges and difficulties associated with large‐scale experiments aimed at manipulating a critical climatic factor.  相似文献

Mesothelin is a cell surface associated antigen expressed on mesothelial cells and in some malignant neoplasms. Mesothelin-targeted therapies are in phase I/II clinical trials. The clinicopathologic and prognostic significance of mesothelin expression in triple negative breast carcinomas (TNBC) has not been fully assessed. We evaluated the expression of mesothelin and of basal markers in tissue microarrays of 226 TNBC and 88 non-TNBC and assessed the clinicopathologic features of mesothelin-expressing breast carcinomas. Furthermore, we investigated the impact of mesothelin expression on the disease-free and overall survival of patients with TNBC. We found that mesothelin expression is significantly more frequent in TNBC than in non-TNBC (36% vs 16%, respectively; p = 0.0006), and is significantly correlated with immunoreactivity for basal keratins, but not for EGFR. Mesothelin-positive and mesothelin-negative TNBC were not significantly different by patients’ race, tumor size, histologic grade, tumor subtype, lymphovascular invasion and lymph node metastases. Patients with mesothelin-positive TNBC were older than patients with mesothelin-negative TNBC, developed more distant metastases with a shorter interval, and had significantly lower overall and disease-free survival. Based on our results, patients with mesothelin-positive TNBC could benefit from mesothelin-targeted therapies.  相似文献

Receptor-like kinases (RLKs) are a family of transmembrane proteins with a variable ligand-binding extracellular domain and a cytoplasmic kinase domain. In Arabidopsis, there are ∼600 RLKs believed to have diverse functions during plant growth, development and interactions with the environment. Based on the variable extracellular domain, RLKs can be classified into different subfamilies. The CrRLK subfamily contains 17 members in Arabidopsis and characterization of some of its members suggests a role for these proteins in the regulation of growth and reproduction. This review focuses on the roles of CrRLKs in the regulation of polarized growth with emphasis on the newly identified signal transduction pathways activated downstream of CrRLKs. A picture is emerging where CrRLKs are part of a conserved signal transduction cascade important for growth maintenance in different cell types.Key words: CrRLKs, FERONIA, RAC/ROP, ROS, polar growthThe ability of plants to perceive and process environmental and internal information into coordinated responses is crucial to their adaptability and survival in constantly changing environments. Most of signal perception occurs at the plasma membrane of cells where membrane-associated receptors receive signals to activate downstream signaling cascades that regulate growth and development. In plants and animals alike, receptor-like kinases (RLKs) mediate many of the signaling events at the cell surface and in the model plant Arabidopsis they comprise a monophyletic family with more than 600 members.1 RLKs are transmembrane proteins with a variable N-terminal extracellular domain and a Ser/Thr intracellular kinase domain. The diversity of their extracellular domains suggests involvement in the transduction of a wide range of signals and allows them to be classified into different sub-families.2 The CrRLK1L subfamily (from here on referred to as CrRLK) is named after the first member characterized in Catharanthus roseus cell cultures3 and contains 17 members in Arabidopsis.4 Several members of this family have now been implicated in growth regulatory processes.THESEUS1 (THE1) was identified through a suppressor screen of a cellulose-deficient mutant (prc1-1) which has a short hypocotyl phenotype.5 Loss of THE1 function resulted in reduced growth inhibition in the prc1-1 the1 double mutant. Interestingly, the the1 mutation itself has no effect in wild type background, thus leading to the suggestion that THE1 functions as a sensor of cell wall integrity in situations where the cell wall is weakened and organ elongation would be detrimental for the plant.4,5A second CrRLK, FERONIA (FER), was first implicated in the regulation of female control of fertility. In the female gametophyte FER is involved in sensing pollen tube arrival and promoting its rupture which is necessary for double fertilization to occur.6,7 FER is in fact involved in several processes depending on the tissue where it is expressed. In hypocotyls, FER is involved in the integration of ethylene and brassinosteroid (BR) signals to regulate hypocotyl elongation in the dark.8 Moreover, FER, THE1 and the closely related HERCULES1 (HERK1), were found to regulate cell elongation by interacting with BR signaling.9 More recently, roles for FER in the regulation of root hair development and fungal invasion have been established.10,11 The pollen-specific ANXUR1 (ANX1) and ANXUR2 (ANX2) are closely related to FER and act redundantly to maintain pollen tube growth integrity during its journey through the style and ensure against precocious pollen tube rupture before reaching the ovule.12,13Apparently with different biological roles, all the CrRLK members analyzed thus far have an effect on the growth of plant cells. The present review focuses on their role during cell growth with emphasis on polar cell growth and the downstream pathways activated by CrRLKs.  相似文献
