The effects of prostaglandin E2 (PGE2) in association with erythropoietin on the synthesis of fetal and adult hemoglobin in peripheral blood-derived erythroid burst colonies from normal adults and from patients with sickle cell anemia were investigated. The synthesized hemoglobin at the end of 8, 14 or 18 days in culture was separated by DEAE-cellulose chromatography of 35S-methione labelled hemoglobin. Quantitative estimation of the synthesized hemoglobin phenotypes, for the three indicated culture periods, showed preferential synthesis of Hb F in addition to an overall increase in hemoglobin synthesis in PGE2 treated colonies. Furthermore, the reactivation of fetal hemoglobin production by PGE2 was more pronounced when the adherent cells were included in the culture dishes. These results indicate that the addition of PGE2 to culture dishes presumably constitutes an environmental change to promote the functional seen in the blood erythroid bursts in terms of Hb synthesis and switching. 相似文献

Genetic and physical characterization of proBA genes of the marine bacterium Vibrio parahaemolyticus
Intracellular proline pools have been implicated in the halotolerance of many organisms. To examine this relationship in a moderately halotolerant marine bacterium, Vibrio parahaemolyticus, proline biosynthesis genes were cloned in various plasmids. Some genetic and structural properties of those genes were examined. Subcloning showed that about 3.1 kilobases of V. parahaemolyticus DNA could complement proA and proB but not proC mutations of Escherichia coli. The same fragment would also complement some Pro- mutants of V. parahaemolyticus. Gamma-delta insertion mutagenesis of this subcloned fragment indicated that proB and proA genes of V. parahaemolyticus might be transcribed from different promoters. Two other genes, phoE and gpt, which map closely to the proBA genes in E. coli, were also found to be in close proximity to the proBA genes of V. parahaemolyticus. 相似文献

Vicia faba lectin contained 40-57% beta-conformation, 4-23% alpha-conformation along with random coil at pH 7.2 depending upon the analytical methods used. The percentage of beta-conformation increased with the addition of N-acetyl-D-glucosamine or methyl alpha-D-mannopyranoside. The structural transitions of V. faba lectin were affected by alkali at pH 9.6 and 10.6. Binding constants and free energy changes for the interaction between V. faba lectin and N-acetyl-D-glucosamine and methyl alpha-D-mannopyranoside were estimated at pH 7.2 using the c.d. and fluorescence methods. 相似文献

Purification and characterization of microsomal glutathione S-transferase produced by Aspergillus ochraceus TS are reported. The isozymes are located in microsomes and were active against 1-chloro-2,4-dinitrobenzene, ethacrynic acid, 1,2-dichloro-4-nitrobenzene, trans-4- phenyl-3-buten-2-one,p-nitrobenzyl chloride and bromosulphophthalein. They were inhibited by N-ethylmaleimide and bromosulphophthalein. The GST isozymes produced by Aspergillus ochraceus TS are indistinguishable in respect of their molecular mass both in native and denatured state. The subunit of the purified protein had an apparent Mr of 11 kDa while molecular mass of the native protein is around 56 kDa. The substrate specificity and pl values of the isozymes were different. The GSTs produced by Aspergillus ochraceus TS fairly share functional properties with mammalian cytosolic isozymes. 相似文献

Summary Phosphate sorption isotherms were developed for five Philippine wetland rice soils using the conventional technique and a modified one. In the conventional method, P requirements of soils varied between 280 and 810 g P/g soil. In the modified method, they varied from 160 to 540 g P/g soil at 0.2 ppm P in solution. Soils with high P-sorption capacities had vermiculite and halloysite as the dominant clay minerals. Soil reduction by flooding decreased P-sorption by 28–70 percent at 0.2 ppm P in solution. The decrease in P-sorption due to soil reduction was greatest in a crystalline soil with vermiculite and halloysite as the dominant clay minerals and least in a soil with dominant X-ray amorphous silicates in the clay fraction.Desorption of freshly adsorbed P under reduction was greater in HCO

solution than in CaCl2 and it increased with level of applied P. Desorption patterns of freshly adsorbed P were similar to adsorption patterns but values of P in solution were lower at desorption. Soils varied with respect to desorption of freshly sorbed P. Desorption studies indicate that soils vary in intensity factor with respect to P and thus influence P availability to plants. Use of P-sorption and P-desorption data obtained under reduced soil condition was proposed for detecting P needs of submerged rice soils.Results of a pot study with IR36 at different levels of solution P (reduced) in one soil indicated a high degree of correlation between adjusted P levels and the measured growth parameters.
