In the cyanobacterium Spirulina platensis, the desaturation process is carried out by three desaturases: the 9, 12 and 6 desaturases, encoded by desC, desA and desD, respectively. The 6 desaturase is responsible for the catalysis of linoleic acid, yielding -linolenic acid (18:39,12,6), the end-product of the process. In this study, the desD gene was expressed in Escherichia coli using a pTrcHisA expression system. In order to identify the amino acid residues involved in the enzymatic activity, a sequence comparison was performed using various organisms. The alignment revealed three conserved histidine clusters, a number of conserved residues among all listed organisms and a few conserved residues among cyanobacterial species possibly involved in the desaturation activity. A series of site-directed mutations were generated in the desD gene to evaluate the role of these residues vis-à-vis the enzyme function. This approach revealed that: (1) H313 is involved in the regioselectivity of the enzyme, (2) the three histidine clusters together with H313, H315, D138 and E140 are required for enzymatic activity, most likely as providers of the catalytic Fe center and (3) W294 is also essential for the activity of 6 desaturase, possibly by forming part of the substrate-binding pocket.  相似文献

Alstonia scholaris (Dr C. Alston, 1685–1760) (Family Apocynaceae) (Chattim tree), commonly known as devil tree, is an evergreen tropical tree. The tree is native to India and also found in Sri Lanka, Southern China, throughout Malaysia to northern Australia. This plant is seriously damaged by formation of tumor like galls across the Kolkata city,West Bengal which affects its ornamental and medicinal value. Gall is formed by ovipositing adults of Pseudophacopteron alstonium Yang et Li 1983 (Hemiptera: Psyllidae: Phacopteronidae) and results in destruction of host plant. The nymphal stage undergoes moulting through first instar to third instar to reach the adult within galls. It is observed that highly infested leaves can bear 60–80 galls. The gallmaker Pseudophacopteron sp. stresses the host organ, and the host counters it with physiological activities supplemented by newly differentiated tissues. In infested leaves, chlorophyll and carbohydrate contents decreased sequentially with the age of the gall. There were no significant changes in protein and total amino acid content in gall tissue. But total lipid content was highest in mature galled leaves. Increased phenolic content after psylloid herbivory, which exerted oxidative stress on the host plants, was observed in gall infested leaves as compared to fresh ungalled leaves of Alstonia scholaris. Moisture content was highest in ungalled healthy leaves than the young galled, mature galled and perforated galled leaves.  相似文献

The effectiveness of various trace element concentrations in medicinal plants in the cure of various diseases can be determined by their quantitative estimation. Elemental concentrations of aqueous extract of F. religiosa leaves were measured by Laser-induced breakdown spectroscopy (LIBS). LIBS is a very powerful and efficient analytical tool for determining elemental constitution. The present study deals with the LIBS-based validation of elements responsible for the glycemic potential of aqueous extract of F. religiosa leaves in streptozotocin-induced diabetic models. The significant decrease in blood glucose level and marked improvement in glucose tolerance test of diabetic models is correlated to the concentration of elements present in the extract as revealed by LIBS spectra. Elements such as Mg and Ca have been observed in the LIBS spectra of F. religiosa.  相似文献

Deinococcus species exhibit an extraordinary ability to withstand ionizing radiation (IR). Most of the studies on radiation resistance have been carried out with exponential phase cells. The studies on radiation resistance of Deinococcus radiodurans R1 with respect to different phases of growth showed that late stationary phase cells of D. radiodurans R1 were fourfold more sensitive to IR and heat as compared with exponential or early stationary phase cells. The increased sensitivity of D. radiodurans R1 to IR in the late stationary phase was not due to a decrease in the intracellular Mn/Fe ratio or an increase in the level of oxidative protein damage. The resistance to IR was restored when late stationary phase cells were incubated for 15 min in fresh medium before irradiation, indicating that replenishment of exhausted nutrients restored the metabolic capability of the cells to repair DNA damage. These observations suggest that stress tolerance mechanisms in D. radiodurans R1 differ from established paradigms.  相似文献
