Mouse ficolin A is a plasma protein with lectin activity, and plays a role in host defense by binding carbohydrates, especially GlcNAc, on microorganisms. The ficolin A subunit consists of an N-terminal signal peptide, a collagen-like domain, and a C-terminal fibrinogen-like domain. In this study, we show that ficolin A can be synthesized and oligomerized in a cell and secreted into culture medium. We also identify a functionally relevant signal peptide of ficolin A by using MS/MS analysis to determine the N-terminal sequence of secreted ficolin A. When the signal peptide of mouse ficolin A was fused with enhanced green fluorescent protein (EGFP), EGFP was released into HEK 293 cell medium, suggesting that the signal peptide can efficiently direct ficolin A secretion. Moreover, our results suggest that the signal peptide of ficolin A has potential application for the production of useful secretory proteins. 相似文献

There have been numerous developments in ethanol fermentation technology since the beginning of the new millennium as ethanol
has become an immediate viable alternative to fast-depleting crude reserves as well as increasing concerns over environmental
pollution. Nowadays, although most research efforts are focused on the conversion of cheap cellulosic substrates to ethanol,
methods that are cost-competitive with gasoline production are still lacking. At the same time, the ethanol industry has engaged
in implementing potential energy-saving, productivity and efficiency-maximizing technologies in existing production methods
to become more viable. Very high gravity (VHG) fermentation is an emerging, versatile one among such technologies offering
great savings in process water and energy requirements through fermentation of higher concentrations of sugar substrate and,
therefore, increased final ethanol concentration in the medium. The technology also allows increased fermentation efficiency,
without major alterations to existing facilities, by efficient utilization of fermentor space and elimination of known losses.
This comprehensive research update on VHG technology is presented in two main sections, namely VHG brewing, wherein the effects
of nutrients supplementation, yeast pitching rate, flavour compound synthesis and foam stability under increased wort gravities
are discussed; and VHG bioethanol fermentation studies. In the latter section, aspects related to the role of osmoprotectants
and nutrients in yeast stress reduction, substrates utilized/tested so far, including saccharide (glucose, sucrose, molasses,
etc.) and starchy materials (wheat, corn, barley, oats, etc.), and mash viscosity issues in VHG bioethanol production are
detailed. Thereafter, topics common to both areas such as process optimization studies, mutants and gene level studies, immobilized
yeast applications, temperature effect, reserve carbohydrates profile in yeast, and economic aspects are discussed and future

Background
Metabolic syndrome is a risk factor for type 2 diabetes and cardiovascular disease. We identified common genetic variants that alter the risk for metabolic syndrome in the Korean population. To isolate these variants, we conducted a multiple-genotype and multiple-phenotype genome-wide association analysis using the family-based quasi-likelihood score (MFQLS) test. For this analysis, we used 7211 and 2838 genotyped study subjects for discovery and replication, respectively. We also performed a multiple-genotype and multiple-phenotype analysis of a gene-based single-nucleotide polymorphism (SNP) set.Results
We found an association between metabolic syndrome and an intronic SNP pair, rs7107152 and rs1242229, in SIDT2 gene at 11q23.3. Both SNPs correlate with the expression of SIDT2 and TAGLN, whose products promote insulin secretion and lipid metabolism, respectively. This SNP pair showed statistical significance at the replication stage.Conclusions
Our findings provide insight into an underlying mechanism that contributes to metabolic syndrome.14.

Nanotechnology has become one of the most promising new approaches for pest control in recent years. In this research, biocompatible silver nanoparticles (Btk-AgNPs) were synthesised by using the entomopathogenic bacterium, Bacillus thuringiensis kurstaki (Btk) as a low-cost and eco-friendly production system. The AgNP samples exhibited a brownish-yellow colour that is characteristic for silver nanoparticles synthesis. Btk-synthesised AgNPs were produced using both the supernatant and pellet of Bt culture at various concentrations and AgNP particles were characterised by UV-Vis spectrophotometer and Dynamic Light Scattering (DLS). The variation of hydrodynamic diameter (Dh) and UV-Vis spectra of silver particles produced by various concentration of culture showed that production of AgNPs was maximised when using 20% for either supernatant or pellet treatments of Bt of culture and the size of particles was around 85?nm for both. The insecticidal efficacy of Btk-synthesised AgNPs against larvae of the cabbage looper, Trichoplusia ni (Hübner) and black cutworm, Agrotis ipsilon (Hufnagel) was tested. Results demonstrated that the treatments of either Btk-synthesised AgNP(s) made with Bt supernatant or Btk-synthesised AgNP(p) using Bt pellet were found to be significantly more virulent toward larvae of T. ni than to A. ipsilon. 相似文献

A simple, yet effective approach of stabilizing the nanostructure of porous metal‐based electrodes and thus, extending the life of microsolid oxide fuel cells is demonstrated. In an effort to avoid thermal agglomeration of metal electrodes, an ultrathin yttria‐stabilized zirconia (YSZ) is coated on the porous metal (Pt) cathode by the atomic layer deposition, a scalable, and potentially high‐throughput deposition technique. A very thin YSZ coating is found to maintain the morphology of its underlying nanoporous Pt during high temperature operations (500 °C). More interestingly, the YSZ coating is also found to improve oxygen reduction reaction activity by ≈2.5 times. This improvement is attributed to an enhanced triple phase area, especially in the vicinity of the Pt–electrolyte interface; cross‐sectional electron microscopy images indicate that the initially uniform ultrathin YSZ layer becomes a partially agglomerated coating, a favorable structure for a maximized reaction area and fluent oxygen access to the Pt–electrolyte interface. 相似文献

Tissue ischemia remains a common problem in plastic surgery and one for which proangiogenic approaches have been investigated. Given the recent discovery of circulating endothelial stem or progenitor cells that are able to form new blood vessels, the authors sought to determine whether these cells might selectively traffic to regions of tissue ischemia and induce neovascularization. Endothelial progenitor cells were isolated from the peripheral blood of healthy human volunteers and expanded ex vivo for 7 days. Elevation of a cranially based random-pattern skin flap was performed in nude mice, after which they were injected with fluorescent-labeled endothelial progenitor cells (5 x 10(5); n = 15), fluorescent-labeled human microvascular endothelial cells (5 x 10(5); n = 15), or media alone (n = 15). Histologic examination demonstrated that endothelial progenitor cells were recruited to ischemic tissue and first appeared by postoperative day 3. Subsequently, endothelial progenitor cell numbers increased exponentially over time for the remainder of the study [0 cells/mm2 at day 0 (n = 3), 9.6 +/- 0.9 cells/mm2 at day 3 (n = 3), 24.6 +/- 1.5 cells/mm2 at day 7 (n = 3), and 196.3 +/- 9.6 cells/mm2 at day 14 (n = 9)]. At all time points, endothelial progenitor cells localized preferentially to ischemic tissue and healing wound edges, and were not observed in normal, uninjured tissues. Endothelial progenitor cell transplantation led to a statistically significant increase in vascular density in ischemic tissues by postoperative day 14 [28.7 +/- 1.2 in the endothelial progenitor cell group (n = 9) versus 18 +/- 1.1 in the control media group (n = 9) and 17.7 +/- 1.0 in the human microvascular endothelial cell group (n = 9; p < 0.01)]. Endothelial progenitor cell transplantation also showed trends toward increased flap survival [171.2 +/- 18 mm2 in the endothelial progenitor cell group (n = 12) versus 134.2 +/- 10 mm2 in the media group (n = 12) and 145.0 +/- 13 mm2 in the human microvascular endothelial cell group (n = 12)], but this did not reach statistical significance. These findings indicate that local tissue ischemia is a potent stimulus for the recruitment of circulating endothelial progenitor cells. Systemic delivery of endothelial progenitor cells increased neovascularization and suggests that autologous endothelial progenitor cell transplantation may have a role in the salvage of ischemic tissue. 相似文献

Height is a classic polygenic trait with high heritability (h2 = 0.8). Recent genome-wide association studies have revealed many independent loci associated with human height. In addition, although many studies have reported an association between copy number variation (CNV) and complex diseases, few have explored the relationship between CNV and height. Recent studies reported that single nucleotide polymorphisms (SNPs) are highly correlated with common CNVs, suggesting that it is warranted to survey CNVs to identify additional genetic factors affecting heritable traits such as height.This study tested the hypothesis that there would be CNV regions (CNVRs) associated with height nearby genes from the GWASs known to affect height. We identified regions containing > 1% copy number deletion frequency from 3667 population-based cohort samples using the Illumina HumanOmni1-Quad BeadChip. Among the identified CNVRs, we selected 15 candidate regions that were located within 1 Mb of 283 previously reported genes. To assess the effect of these CNVRs on height, statistical analyses were conducted with samples from a case group of 370 taller (upper 10%) individuals and a control group of 1828 individuals (lower 50%).We found that a newly identified 17.7 kb deletion at chromosomal position 12q24.33, approximately 171.6 kb downstream of GPR133, significantly correlated with height; this finding was validated using quantitative PCR. These results suggest that CNVs are potentially important in determining height and may contribute to height variation in human populations. 相似文献
