Studies on semen and sperm cells are critical to develop assisted reproductive technologies for the conservation of the collared peccary. The objective of the study was to compare the effect of different antibiotics on the bacterial load and sperm quality during short-term storage of peccary semen. Fresh semen samples from 10 males were extended in Tris-egg yolk or Tris-Aloe vera supplemented with streptomycin-penicillin (SP; 1 mg/mL - 1000 IU/mL or 2 mg/mL - 2000 IU/mL) or gentamicin (30 µg/mL or 70 µg/mL) before storage at 5°C. Bacterial load and sperm motility, membrane integrity and function, mitochondrial activity, and morphology, were evaluated at different time points for 36 h. The SP and gentamicin treatments concentration inhibited (p < 0.05) bacterial growth for 36 h regardless of the extender. Compared to the other treatments, Tris-egg yolk plus 70 µg/mL gentamicin maintained the sperm parameters for longer, including total motility (41.9 ± 6.1%) at 24 h, and membrane integrity (58.3 ± 2.1%) at 36 h. In contrast, the highest SP concentration in both extenders impaired sperm membrane integrity at 36 h (p < 0.05). For the liquid storage of collared peccary semen, it therefore is recommended to use Tris extender supplemented with egg yolk and gentamicin (70 µg/mL). 相似文献

Plant growth depressions in arbuscular mycorrhizal symbioses: not just caused by carbon drain? 总被引:1,自引:0,他引:1
* This study investigated effects of plant density and arbuscular mycorrhizal (AM) colonization on growth and phosphorus (P) nutrition of a cultivar of wheat (Triticum aestivum) that often shows early AM-induced growth depressions. * Two experiments were conducted. Expt 1 had three plant densities and one soil P concentration. Expt 2 had two plant densities and two P concentrations. Plants were grown in calcareous P-fixing soil, inoculated with Glomus intraradices or Gigaspora margarita, or noninoculated (nonmycorrhizal (NM)). Glomus intraradices colonized well and caused a growth depression only in Expt 1. Gigaspora margarita caused large growth depressions in both experiments even though it colonized poorly. * The results showed that growth depressions were mitigated by changes in relative competition for soil P by NM and AM plants, and probably by decreasing carbon costs of the fungi. * The different effects of the two fungi appear to be attributable to differences in the balance between P uptake by the fungal pathway and direct uptake via the roots. These differences may be important in other AM symbioses that result in growth depressions. From the dichloromethane extract of the flowers of Helichrysum gymnocomum (Asteraceae) two known flavonoids, 4 and 5, and a known acylphloroglucinol, 3B, were isolated. In addition to 1 and 2, the 4′,6′-dibenzyloxy-2′-hydroxy derivative of 2′,4′,6′-trihydroxychalcone and 5,7-dibenzyloxy derivative of pinocembrin, respectively, are reported in Nature for the first time. A compound 3A, related to 3B has the structure 2-methyl-1-[2,4,6-trihydroxy-3-(2-hydroxy-3-methyl-3-butenyl)phenyl]-1-propanone. Compounds 1, 2, 3A, 3B, 4 and 5 have MIC values below 64 μg/ml against a selection of pathogens, with 3B having the highest sensitivity (6.3–45 μg/ml) for eight of the ten pathogens tested, including Staphylococcus aureus (6.3 μg/ml) and methicillin and gentamycin resistant strain of S. aureus (7.8 μg/ml). The current distribution of the introduced seagrass Zostera japonica is restricted to the mid- to upper intertidal zone in the coastal Pacific Northwest region of North America. The climate in this region is cool and wet, becoming hotter and dryer with increasing distance southward. Since temperature is likely to be an important factor affecting distribution of this species, growth of two populations located near the northern and southern limits of its established range along the Pacific Coast of North America were measured in an experimental setting across a range of temperatures typical of those in the field during the growing season (10, 20, and 30 °C). The effects of temperature and population were both significant. Leaf elongation, growth, and areal productivity rates of the northern population were consistently lower than those of the southern population. Across the range of temperatures, mean leaf elongation rates ranged from 0.47 to 1.40 cm2 shoot−1 d−1; mean growth rates ranged from 0.19 to 0.52 mg dry wt shoot−1 d−1. Mean areal productivity ranged from 0.54 to 1.92 g dry wt m−2 d−1. Maximum rates of leaf elongation, growth, and areal productivity for both populations were observed at 20 °C. However, leaf elongation, growth, and areal productivity of the northern population declined markedly at 30 °C, whereas no comparable declines were observed for the southern population. This suggests that Z. japonica populations near the southern limits of its established range may be better adapted to warmer temperatures than populations near the northern range limits and further range extensions southward along the California coast may be likely. These differences could be important in predicting the outcome of competitive interactions between native and introduced seagrass species, and in determining future patterns of distribution and zonation of Pacific Coast seagrasses. 相似文献

Hutchinson-Gilford progeria syndrome is caused by the synthesis of a mutant form of prelamin A, which is generally called progerin. Progerin is targeted to the nuclear rim, where it interferes with the integrity of the nuclear lamina, causes misshapen cell nuclei, and leads to multiple aging-like disease phenotypes. We created a gene-targeted allele yielding exclusively progerin (Lmna HG) and found that heterozygous mice (Lmna HG/+) exhibit many phenotypes of progeria. In this study, we tested the hypothesis that the phenotypes elicited by the Lmna HG allele might be modulated by compositional changes in the nuclear lamina. To explore this hypothesis, we bred mice harboring one Lmna HG allele and one Lmna LCO allele (a mutant allele that produces lamin C but no lamin A). We then compared the phenotypes of Lmna HG/LCO mice (which produce progerin and lamin C) with littermate Lmna HG/+ mice (which produce lamin A, lamin C, and progerin). Lmna HG/LCO mice exhibited improved HG/LCO fibroblasts had fewer misshapen nuclei than Lmna HG/+ fibroblasts (p < 0.0001). A likely explanation for these differences was uncovered; the amount of progerin in Lmna HG/LCO fibroblasts and tissues was lower than in Lmna HG/+ fibroblasts and tissues. These studies suggest that compositional changes in the nuclear lamina can influence both the steady-state levels of progerin and the severity of progeria-like disease phenotypes. 相似文献

Creating networks of nature reserves to protect areas rich in biodiversity from the adverse impacts of anthropogenic change is a critical and urgent task. We illustrate the skewed geographical and size distributions of protected areas in the Western Hemisphere. For instance, 811 of 1413 reserves in the Western Hemisphere are smaller than 10 km2, and 35% of the total area of these reserves is in Alaska. We compile ranges for all bats in the continental Western Hemisphere and find that 82% of threatened and small‐range species are not protected adequately. Many of the most vulnerable species occur in the areas of highest human density. We provide maps delineating areas where conservation investments may have the greatest impact in preventing biodiversity losses. 相似文献
