To study and define the early time-dependent response (6 h) ofblocker-sensitive epithelial Na+channels (ENaCs) to stimulation ofNa+ transport by aldosterone, weused a new modified method of blocker-induced noise analysis todetermine the changes of single-channel current (iNa) channel open probability(Po), andchannel density(NT) undertransient conditions of transport as measured by macroscopic short-circuit currents(Isc). In threegroups of experiments in which spontaneous baseline rates of transportaveraged 1.06, 5.40, and 15.14 µA/cm2, stimulation of transportoccurred due to increase of blocker-sensitive channels.NT variedlinearly over a 70-fold range of transport (0.5-35µA/cm2). Relatively small andslow time-dependent but aldosterone-independent decreases ofPo occurredduring control (10-20% over 2 h) and aldosterone experimentalperiods (10-30% over 6 h). When thePo of control andaldosterone-treated tissues was examined over the 70-fold extendedrange of Na+ transport,Po was observedto vary inversely withIsc, falling from~0.5 to ~0.15 at the highest rates ofNa+ transport or ~25% per3-fold increase of transport. Because decreases ofPo from anysource cannot explain stimulation of transport by aldosterone, it isconcluded that the early time-dependent stimulation ofNa+ transport in A6 epithelia isdue exclusively to increase of apical membraneNT.

Aim This paper documents reconstructions of the vegetation patterns in Australia, Southeast Asia and the Pacific (SEAPAC region) in the mid‐Holocene and at the last glacial maximum (LGM). Methods Vegetation patterns were reconstructed from pollen data using an objective biomization scheme based on plant functional types. The biomization scheme was first tested using 535 modern pollen samples from 377 sites, and then applied unchanged to fossil pollen samples dating to 6000 ± 500 or 18,000 ± 1000 14C yr bp . Results 1. Tests using surface pollen sample sites showed that the biomization scheme is capable of reproducing the modern broad‐scale patterns of vegetation distribution. The north–south gradient in temperature, reflected in transitions from cool evergreen needleleaf forest in the extreme south through temperate rain forest or wet sclerophyll forest (WSFW) and into tropical forests, is well reconstructed. The transitions from xerophytic through sclerophyll woodlands and open forests to closed‐canopy forests, which reflect the gradient in plant available moisture from the continental interior towards the coast, are reconstructed with less geographical precision but nevertheless the broad‐scale pattern emerges. 2. Differences between the modern and mid‐Holocene vegetation patterns in mainland Australia are comparatively small and reflect changes in moisture availability rather than temperature. In south‐eastern Australia some sites show a shift towards more moisture‐stressed vegetation in the mid‐Holocene with xerophytic woods/scrub and temperate sclerophyll woodland and shrubland at sites characterized today by WSFW or warm‐temperate rain forest (WTRF). However, sites in the Snowy Mountains, on the Southern Tablelands and east of the Great Dividing Range have more moisture‐demanding vegetation in the mid‐Holocene than today. South‐western Australia was slightly drier than today. The single site in north‐western Australia also shows conditions drier than today in the mid‐Holocene. Changes in the tropics are also comparatively small, but the presence of WTRF and tropical deciduous broadleaf forest and woodland in the mid‐Holocene, in sites occupied today by cool‐temperate rain forest, indicate warmer conditions. 3. Expansion of xerophytic vegetation in the south and tropical deciduous broadleaf forest and woodland in the north indicate drier conditions across mainland Australia at the LGM. None of these changes are informative about the degree of cooling. However the evidence from the tropics, showing lowering of the treeline and forest belts, indicates that conditions were between 1 and 9 °C (depending on elevation) colder. The encroachment of tropical deciduous broadleaf forest and woodland into lowland evergreen broadleaf forest implies greater aridity. Main conclusions This study provides the first continental‐scale reconstruction of mid‐Holocene and LGM vegetation patterns from Australia, Southeast Asia and the Pacific (SEAPAC region) using an objective biomization scheme. These data will provide a benchmark for evaluation of palaeoclimate simulations within the framework of the Palaeoclimate Modelling Intercomparison Project.  相似文献

4-Aminophenol (4-AP) and D-serine are established rodent nephrotoxins that selectively damage renal proximal tubules. In an attempt to understand the mechanism of action of these toxicants in greater detail, a high throughput proteomics approach was used to profile protein changes in the plasma of animals treated with these compounds. Male Fischer 344 and Alderley Park rats were treated with increasing doses of 4-AP or D-serine and plasma samples were collected over time. Control groups received either saline or the non-toxic enantiomer, L-serine. Using high throughput two-dimensional gel analysis, a number of plasma proteins showing dose- and time-dependent regulation were identified. One toxicity-associated plasma protein was identified as the cellular enzyme fumarylacetoacetate hydrolase (FAH), which is known to be required for tyrosine metabolism. The FAH gene is mutated in the human genetic disorder type I tyrosinaemia, which is associated with liver and kidney abnormalities and neurological disorders. FAH was elevated in the plasma of animals treated with 4-AP and D-serine at early time points and returned to baseline levels after 3 weeks. The protein was not elevated in the plasma of control animals or those treated with L-serine. The presence of FAH in plasma is intriguing as it is normally a cellular enzyme with no known function in plasma. It is possible that 4-AP and D-serine may work through a previously unknown mechanism in the kidney via regulation of tyrosine metabolism or FAH activity. Therefore, FAH may function in a fashion analogous to the aspartate aminotransferase (AST) and alanine aminotransferase (ALT) enzymes that are used to measure liver injury. The link between kidney toxicants and inherited tyrosinaemia also raises the possibility that FAH may be a marker of kidney toxicity in humans. These observations highlight the value of proteomics in identifying new biomarkers and providing new unprecedented insights into complex biological mechanisms.  相似文献

This protocol details a method to derive human embryonic stem (hES) cells from single blastomeres. Blastomeres are removed from morula (eight-cell)-stage embryos and cultured until they form multicell aggregates. These blastomere-derived cell aggregates are plated into microdrops seeded with mitotically inactivated feeder cells, and then connected with neighboring microdrops seeded with green fluorescent protein-positive hES cells. The resulting blastomere-derived outgrowths are cultured in the same manner as blastocyst-derived hES cells. The whole process takes about 3-4 months.  相似文献

Natural selection drives adaptive evolution, but contrasting environmental pressures may lead to trade-offs between phenotypes that confer different performances. Such trade-offs may weaken the strength of selection and/or generate complex fitness surfaces with multiple local optima that correspond to different selection regimes. We evaluated how differences in patterns of phenotypic selection might promote morphological differences between subpopulations of the amphidromous Hawaiian waterfall-climbing goby, Sicyopterus stimpsoni. We conducted laboratory experiments on fish from the islands of Kaua‘i and Hawai‘i (the “Big Island”) to compare patterns of linear and nonlinear selection, and the opportunity for selection, that result from two contrasting pressures, predator evasion and waterfall climbing, which vary in intensity between islands. We found directional and nonlinear selection were strongest when individuals were exposed to their primary selective pressures (predator evasion on Kaua‘i, waterfall climbing on the Big Island). However, the opportunity for selection was greater for the non-primary pressure: climbing on Kaua‘i, predator evasion on the Big Island. Canonical rotation of the nonlinear gamma matrix demonstrated that individuals from Kaua‘i and the Big Island occupy regions near their local fitness peaks for some traits. Therefore, selection for predator evasion on Kaua‘i and climbing on the Big Island may be less effective in promoting morphological changes in this species, because variation of functionally important traits in their respective environments may have been reduced by directional or stabilizing selection. These results demonstrate that despite constraints on the opportunities for selection, population differences in phenotypic traits can arise due to differences in selective regimes. For S. stimpsoni, sufficient variation exists in other locomotor traits, allowing for necessary levels of performance in the contrasting selective regime (i.e., climbing on Kaua‘i and predator evasion on the Big Island) through many-to-one-mapping, which may be essential for the survival of local populations in an evanescent island environment.  相似文献
