The assessment of data analysis methods in 1H NMR based metabolic profiling is hampered owing to a lack of knowledge of the exact sample composition. In this study, an artificial complex mixture design comprising two artificially defined groups designated normal and disease, each containing 30 samples, was implemented using 21 metabolites at concentrations typically found in human urine and having a realistic distribution of inter-metabolite correlations. These artificial mixtures were profiled by 1H NMR spectroscopy and used to assess data analytical methods in the task of differentiating the two conditions. When metabolites were individually quantified, volcano plots provided an excellent method to track the effect size and significance of the change between conditions. Interestingly, the Welch t test detected a similar set of metabolites changing between classes in both quantified and spectral data, suggesting that differential analysis of 1H NMR spectra using a false discovery rate correction, taking into account fold changes, is a reliable approach to detect differential metabolites in complex mixture studies. Various multivariate regression methods based on partial least squares (PLS) were applied in discriminant analysis mode. The most reliable methods in quantified and spectral 1H NMR data were PLS and RPLS linear and logistic regression respectively. A jackknife based strategy for variable selection was assessed on both quantified and spectral data and results indicate that it may be possible to improve on the conventional Orthogonal-PLS methodology in terms of accuracy and sensitivity. A key improvement of our approach consists of objective criteria to select significant signals associated with a condition that provides a confidence level on the discoveries made, which can be implemented in metabolic profiling studies. 相似文献

Thiocyanates (SCN-) are ubiquitous in nature. There are indispensable part of host defense system that act as a substrate for lactoperoxidase (LPO). In our study we present initial data on SCN- concentration in saliva of CF patients in comparison to healthy non-smokers and healthy smokers. 5 ml of saliva was collected from each subject to a sterile tube and thiocyanate concentration was measured in each sample. The results of the measurements are presented on Fig. 1. Mean concentration of SCN- in saliva of CF patients was 0.031 +/- 0.0052 g/l, in healthy non-smokers 0.039 +/- 0.0048 g/l and in healthy smokers 0.048 +/- 0.0161 g/l. The differences between each group were statistically significant. Studies on larger group of patients and probably on different material (BALF or induced sputum) should present interesting data complementing the in vitro studies. 相似文献

Ventilation-perfusion (V/Q) mismatch is a prominent feature of preterm infants and adults with lung disease. V/Q mismatch is known to cause arterial hypoxemia under steady-state conditions, and has been proposed as the cause of rapid arterial oxygen desaturation during apnea. However, there is little evidence to support a role for V/Q mismatch in the dynamic changes in arterial oxygenation that occur during apnea. Using a mathematical model, we quantified the effect of V/Q mismatch on the rate of desaturation during apnea to ascertain whether it could lead to rates of up to 10% s-1 as observed in preterm infants. We used a lung-body model for the preterm infant that incorporated 50 parallel alveolar-capillary units that were ventilated and perfused with the severity of V/Q mismatch (σ) defined conventionally according to σ=S.D. of the distribution of V/Q ratios. Average desaturation rate 10 s from apnea onset was strongly elevated with worsening V/Q mismatch as a result of an earlier desaturation of low V/Q units compared with high V/Q units. However, V/Q mismatch had little impact after apnea onset, with peak desaturation rate only substantially increased if mismatching caused a lowered resting arterial O2 saturation. In conclusion, V/Q mismatch causes a more immediate onset of desaturation during apnea, and therefore places preterm infants and adults with lung disease at risk of hypoxemic dips. However, V/Q mismatch does not accelerate desaturation rate beyond apnea onset and cannot, therefore, explain the rapid desaturation observed during recurrent apnea in preterm infants. 相似文献

The purpose of this study was to validate the accuracy and reliability of the Weightlifting Video Overlay System (WVOS) used by coaches and sport biomechanists at the United States Olympic Training Center. Static trials with the bar set at specific positions and dynamic trials of a power snatch were performed. Static and dynamic values obtained by the WVOS were compared with values obtained by tape measure and standard video kinematic analysis. Coordinate positions (horizontal [X] and vertical [Y]) were compared on both ends (left and right) of the bar. Absolute technical error of measurement between WVOS and kinematic values were calculated (0.97 cm [left X], 0.98 cm [right X], 0.88 cm [left Y], and 0.53 cm [right Y]) for the static data. Pearson correlations for all dynamic trials exceeded r = 0.88. The greatest discrepancies between the 2 measuring systems were found to occur when there was twisting of the bar during the performance. This error was probably due to the location on the bar where the coordinates were measured. The WVOS appears to provide accurate position information when compared with standard kinematics; however, care must be taken in evaluating position measurements if there is a significant amount of twisting in the movement. The WVOS appears to be reliable and valid within reasonable error limits for the determination of weightlifting movement technique. 相似文献
