A new, continuous spectrophotometric method is described for determining lipase activity using a reverse micelle system, in which lipase (EC 3.1.1.3) and lipoxygenase (EC 1.13.11.12) are dissolved. The reverse micelle system consists of 2-ethyl hexyl sodium sulfosuccinate (AOT)-isooctane and water. Trilinolein is used as the lipase substrate; linoleate hydroperoxide is the end product of the oxidation catalyzed by lipoxygenase, which acts as an auxiliary coupled-enzyme of lipase. The method appears useful both for detailed kinetic studies of lipase and for serial analyses using sunflower oil, a cheaper substrate. This assay offers the typical advantages of the continuous direct photometric methods in that it is rapid, reproducible and sufficiently sensitive for measuring lipase activity even in some crude commercial preparations. 相似文献

With the availability of a large amount of genomic data it is expected that the influence of single nucleotide variations (SNVs) in many biological phenomena will be elucidated. Here, we approached the problem of how SNVs affect alternative splicing. First, we observed that SNVs and exonic splicing regulators (ESRs) independently show a biased distribution in alternative exons. More importantly, SNVs map more frequently in ESRs located in alternative exons than in ESRs located in constitutive exons. By looking at SNVs associated with alternative exon/intron borders (by their common presence in the same cDNA molecule), we observed that a specific type of ESR, the exonic splicing silencers (ESSs), are more frequently modified by SNVs. Chromoblastomycosis is a chronic skin infection caused by the fungus Fonsecaea pedrosoi. Exploring the reasons underlying the chronic nature of F. pedrosoi infection in a murine model of chromoblastomycosis, we find that chronicity develops due to a lack of pattern recognition receptor (PRR) costimulation. F. pedrosoi was recognized primarily by C-type lectin receptors (CLRs), but not by Toll-like receptors (TLRs), which resulted in the defective induction of proinflammatory cytokines. Inflammatory responses to F. pedrosoi could be reinstated by TLR costimulation, but also required the CLR Mincle and signaling via the Syk/CARD9 pathway. Importantly, exogenously administering TLR ligands helped clear F. pedrosoi infection in vivo. These results demonstrate how a failure in innate recognition can result in chronic infection, highlight the importance of coordinated PRR signaling, and provide proof of the principle that exogenously applied PRR agonists can be used therapeutically. 相似文献

Active lymphocytes (LY) and macrophages (MΦ) are involved in the pathophysiology of rheumatoid arthritis (RA). Due to its anti‐inflammatory effect, physical exercise may be beneficial in RA by acting on the immune system (IS). Thus, female Wistar rats with type II collagen‐induced arthritis (CIA) were submitted to swimming training (6 weeks, 5 days/week, 60 min/day) and some biochemical and immune parameters, such as the metabolism of glucose and glutamine and function of LY and MΦ, were evaluated. In addition, plasma levels of some hormones and of interleukin‐2 (IL‐2) were also determined. Results demonstrate that CIA increased lymphocyte proliferation (1.9‐ and 1.7‐fold, respectively, in response to concanavalin A (ConA) and lipopolysaccharide (LPS)), as well as macrophage H2O2 production (1.6‐fold), in comparison to control. Exercise training prevented the activation of immune cells, induced by CIA, and established a pattern of substrate utilization similar to that described as normal for these cells. Exercise also promoted an elevation of plasma levels of corticosterone (22.2%), progesterone (1.7‐fold) and IL‐2 (2.6‐fold). Our data suggest that chronic exercise is able to counterbalance the effects of CIA on cells of the IS, reinforcing the proposal that the benefits of exercise may not be restricted to aerobic capacity and/or strength improvement. The complete definition of the chemical structure of GD1b-ganglioside (GD1b) lactone isolated from human brain has been given by means of spectrometric and spectroscopic analyses. GD1h lactone contains a single ester linkage involving the external sialic acid carboxyl group and the C-9 hydroxyl group of the internal sialic acid unit. The aggressive behaviour of female and male Apennine chamois has been compared quantitatively. As opposed to what males did, females significantly attacked each other less often; preferred to gore body regions with a low risk of lethal injury, made a greater use of direct forms of aggression; seldom interacted with the opponent before attacking and gored it more often. Front clashing and fighting were very rare in both sexes. Females live in resident kin-groups, while young males disperse and adults are solitary. To a large extent habitat separation occurs between the sexes. Sexual differences in patterns of aggressive behaviour may be related to the different gregariousness of females and males, probably influenced by resource availability in the habitats they use. Chamois sexes are nearly monomorphic, females bearing slightly less hooked horns than males. This species may have evolved strongly hooked weapons as a first step to advanced wrestling or butting type horns from the ancestral stiletto shape, as goats and sheep, as well as deer, have done. 相似文献

We studied the kinetics of O2 release by oxyhemoglobin caused by sodium dithionite, in the presence and in the absence of organic cosolvents (monohydric alcohols and formamide) at 10°C. This study was performed by using standard stopped-flow techniques coupled with microprocessor-based data acquisition. We have fitted the experimental data to a mathematical expression obtained on the basis of a two-state model that takes into account the kinetic heterogeneity between α- and β-chains and the presence of αβ-dimers in oxyhemoglobin solutions. Results indicate that the cosolvents mainly affect the allosteric parameter L, i.e., the T ? R conformational equilibrium of hemoglobin, leaving the intrinsic deoxygenation rates of both R and T states almost unaltered. The L values obtained in the present work are in excellent agreement with analogous values previously estimated from oxygen equilibrium measurements. 相似文献

An efficiency assessment of a ganglioside assay procedure was carried out on human serum gangliosides from healthy subjects of different sex and age. The analysis of the gangliosides, extracted with chloroform/methanol and purified by lipid partitioning, ion exchange column chromatographic separation and desalting procedures as described by Sennet al. (1989)Eur J Biochem
181: 657–62, was performed by HPTLC followed by densitometric quantification. The yield of the procedure, expressed as radioactivity recovery, was determined by adding GM3 ganglioside, tritium labelled at the sialic acid acetyl group and at the C3 position of sphingosine, to the lyophilized serum or by associating it with the serum lipoproteins. In spite of the fact that the extraction and purification procedures were performed exactly as described we found the radioactivity recovery to be variable (25–50%) and much lower than that proposed. Much of the radioactivity was found in the organic phase after lipid partitioning, whilst all the ganglioside purification steps led to some further loss. After the introduction of some modifications to the procedure the recovery improved, reaching 67–79%.The analyses on 33 samples of 5 ml showed a human serum ganglioside content of about 10 nmol ml–1 (as corrected for the recovery), and confirmed that GM3 ganglioside is the main component of the total serum ganglioside mixture.

The developmental profiles of the gangliosides and those of the fatty acids and long-chain bases of the total ganglioside mixture of the brain of chicken were followed from the 10th day of incubation to the 63rd posthatching day. One O-acetylated polysialoganglioside that seems specific of the earlier embryonic stage and up to 21 alkali-stable components could be recognized by high-resolution two-dimensional TLC procedures and quantified by computer-assisted two-dimensional TLC densitometry. Besides a number of gangliosides identified by co-chromatography with reference standards, 10 were of unidentified structure, and within these 4 did not belong to the gangliotetrahexosyl series. Throughout embryonic life, the ceramide portion of gangliosides was found to contain the long-chain base species with 18 carbons. Those with 20 carbons, approximately 10% of the total, were found to be present only after hatching. 相似文献
