The region(s) of bovine galactosyltransferase that interacts with the lactose synthase regulatory protein alpha-lactalbumin was investigated using trace 3H acetylation to probe the effects of alpha-lactalbumin on the reactivities of the individual amino groups of galactosyltransferase. In the presence of Mn2+, alpha-lactalbumin was found to reduce the reactivities of lysines 93 and 181 and to increase the reactivities of one or more of lysines 230, 237, and 241. The addition of N-acetylglucosamine (20 mM), which enhances complex formation between the two proteins, did not significantly alter the pattern of perturbation. These results indicate that the NH2-terminal region of the catalytic domain of galactosyltransferase, and possibly part of the proline-rich "stem" region, is affected by the association with alpha-lactalbumin and is therefore implicated in the binding of acceptor substrates. In a separate study only cysteines 176, 266, and 342 of galactosyltransferase were found to react with [3H]iodoacetic acid under denaturing conditions. From their lack of reactivity it is deduced that the remaining two cysteines, residues 134 and 247, are joined in a disulfide linkage. From these results and those of a previous study of UDP-galactose binding (Yadav, S., and Brew, K. (1990) J. Biol. Chem. 265, 14163-14169) it appears that the soluble form of galactosyltransferase is composed of two domains, the NH2-terminal 150 residues containing the Cys134-Cys247 disulfide bond, which functions in alpha-lactalbumin and acceptor binding, and the COOH-terminal region, which is involved in UDP-galactose binding. 相似文献
Five commercial preparations of natural pesticides were tested for in vitro compatibility with muscardine fungi, Beauveria brongniartii and Metarhizium anisopliae. Neemark (azadirachtin) was found compatible with both the fungi. Phytoallexin, the natural fungicide, significantly inhibited the growth of both the fungi, while other natural pesticides showed moderate to severe inhibition. 相似文献
Rodents and shrews were screened for serologic evidence of Coxiella burnetii. Attempts were made to isolate the organism from the spleen and liver. Seroreactors: total positive/total tested (% positive), in rats (Rattus rattus, R. norvegicus), ground shrews (Suncus murinus), bandicoots (Bandicota indica, B. bengalensis) and the house mouse (Mus musculus), respectively, were 13/105 (12.38), 6/42 (14.3), 2/15 (13.3) and 1/7 (14.3). Of the eight rickettsial isolants recovered including four from field and household rats, three from ground shrew and one from bandicoots, only three comprising one each from rat, shrew and bandicoot, could be typed as C. burnetii. This appears to be the first record of rodents and an insectivore as reservoirs of C. burnetii in India. 相似文献
Conditions for the laboratory-scale production of acetoin plus diacetyl by Enterobacter Cloacae ATCC 27613 were studied. Thirty-five g acetoin plus diacetyl/50 g sucrose were obtained when fermentation was carried out in 2. 5 liter medium containing 12.5 g peptone and 12. 5 g yeast extract, at pH 7.0, in a 5 liter conical flask on a shaker (240rpm) at 28–30°C for 48 hr. The blood of two fresh water cobitids — Botia lohachata, an exclusive water breathing form and Lepidocephalus guntea a dual breather, — showed a comparatively higher range of Hb (16.0–19.0 g%), Hct (50.0–61.1%) and number of RBC (2.71–6.7 millions/mm3) than many other water and air breathing fishes. Significant sexual difference exists in these characteristics (P > 0.05).The impact of life in oxygen depleted water, also inhabited by L. guntea as a result of its air breathing habit, is well reflected in its greater RBC size (11.86 × 8.66 µm) and their larger surface area (83.96 µm2) compared to that of Botia (53.16 µm2). The smaller size (9.92 × 6.45 µm) and consequently greater number of erythrocytes (4.67 millions/mm3) in Botia, are related to its active mode of life in the swiftly flowing water of hilly rivers. Though the higher nucleo-cytoplasmic ratio of 1 : 5.2 in Botia and 1 : 6.9 in Lepidocephalus suggest a slower red cell metabolism, the greater number of erythrocytes seems to have compensated for their active mode of life. 相似文献
The need for an easy to fabricate perfect and narrowband light absorber in the visible range of electromagnetic (EM) spectrum has always been in demand for many scientific and device applications. Here, we propose a metal-dielectric-metal (MDM) 1-D grating plasmonic structure as a perfect narrow band light absorber in the visible and its application in glucose detection. The proposed structure consists of a 1- D grating of gold on the top of a dielectric layer on a gold film. Optimization for dielectric grating index (n), grating thickness (t), grating width (W), and grating period (P) has been done to improve the performance of plasmonic structure by calculating its quality factor and figure-of-merit (FOM). The optimized plasmonic structure behaves as a perfect narrowband light absorber. The flexibility to work at a specific wavelength is also offered by the proposed structure through an appropriate selection of the geometrical parameters and refractive index of the dielectric grating. The equivalent RC model is used to understand different components of the proposed structure on the optical response. The absorption response of the structure is invariant to the incident angle. Moreover, the calculated absorbance of the proposed plasmonic structure is ~ 100% with a narrow full-width half maxima (FWHM) of ~ 2.8 nm. We have numerically demonstrated a potential application of the proposed MDM absorber as a plasmonic glucose sensor in the visible range with detection sensitivity in the range of 140 to 195 nm/RIU.
