We have recently found that matrix metalloproteinases (MMPs) are targets for T-cell and B-cell reactivity in experimental arthritis. In the present article, we investigate whether modulation of MMP-specific T-cell responses could influence the course of adjuvant arthritis (AA). Lewis rats were treated nasally with MMP peptides prior to or after AA induction. Administration of the MMP-10 or the MMP-16 peptide prior to AA induction reduced the arthritic symptoms. In contrast, administration of the MMP-10 peptide after AA induction aggravated the arthritic symptoms. The present study shows the possible usefulness of MMP peptides for immunotherapy. However, a clear understanding of proper timing of peptide administration is crucial for the development of such therapies.  相似文献

The conventional random pore model assumes a homogeneous cell distribution in the gel matrix used to immobilize cells. However, the validity of this model is restricted to values of the exponent alpha, between 1.8 and 2.25, of a model power function relating the diffusivity coefficient in the matrix with the overall cell volume fraction in the system. Based on the analysis of published data for diffusion in gels with immobilized cells and on the homogeneous approach for the random pore model developed in a previous work, a new, nonhomogeneous approach is proposed for alpha values outside the range 1.8-2.25. To explain these data, two main types of nonhomogeneous cell distribution were considered: (1) nonhomogeneous cell distribution in the gel for alpha > 2.25 (type 1) and (2) nonhomogeneity related with anisotropy of cell space orientation when alpha < 1.8 (type 2). In the case of nonhomogeneity of type 1, the cell volume fraction in the layers occupied by cells must be considered in place of the concept previously used for homogeneous distribution, viz., the average cell volume fraction. This model underlines that accumulation of cells in a thin layer close to the surface improves their nutrient intake. For nonhomogeneity of type 2, the tortuosity of such a system is smaller than should be expected if spherical cells were considered, thereby changing the effective diffusion. The model proposed in this work proved to fit into several real cases reported in the literature.  相似文献

Background. Noninvasive diagnostic tests are useful as screening tools for Helicobacter pylori infection in pediatric populations. The aim of this study was to evaluate performance of the immunoblot assay, Helico Blot 2.1, for the diagnosis of H. pylori infection in symptomatic children. Materials and Methods. Immunoblot assay was used for detection of IgG antibodies to specific H. pylori proteins and to a recombinant H. pylori antigen, CIM marker. The study was performed on sera collected from 134 symptomatic, untreated children (mean age, 9.1 ± 3.2 years; range, 1–14 years). H. pylori infection status was determined by culture, histology and rapid urease test. Results. Immunoblot assay yielded a positive result in 71 of the 72 infected patients (sensitivity 98.6%) and in eight of the 62 noninfected ones (specificity 87.1%). The predictive values for a positive and a negative result were 89.9% and 98.2%, respectively. The performance of the CIM band alone, as a marker for H. pylori infection status, was also evaluated. This band was present on the blot of 71 infected patients and on four of the 62 H. pylori‐negative patients. The sensitivity, specificity, PPV and NPV of the CIM antigen were 98.6%, 93.5%, 94.7% and 98.3%, respectively. Conclusions. The immunoblot assay Helico Blot 2.1 is a suitable noninvasive test for the serodiagnosis of H. pylori infection in children. The good level of performance demonstrated by the novel recombinant antigen CIM suggests it may be a useful contribution to the qualitative and quantitative performance of the Helico Blot 2.1 in pediatric populations.  相似文献

RAPD analyses have been used to determine the genetic diversity and the population structure of the endangered Blanca Cacere?a bovine breed. Genetic variability was evaluated on the basis of 1048 loci produced by 71 primers. RAPD produced a number of polymorphic loci (30.44%), and it has been proved to be a useful method for evaluating polymorphisms in this breed. The dendrograms based on simililarity indexes and on Nei's genetic distances between 60 animals and the value of genetic differentiation among subpopulations (F(ST)) showed a clear population substructure defined by herds and a scarce genetic flow among herds. Analysis of molecular variance (AMOVA) showed that 32.4% of the total variance was due to differences among herds and confirmed the clustering found. The results of the present study allow us to plan more adequate mating in order to maintain the genetic diversity and to improve the efficiency of conservation for the Blanca Cacere?a bovine breed.  相似文献

In the sea slug Aplysia, buccal synapses of cerebral-buccal interneurons (CBIs) CBI-2 and CBI-12 exhibit short-term synaptic enhancement (STE), including frequency-dependant facilitation and augmentation/post-tetanic potentiation (AUG/PTP). The STE that results from driving CBI-2 or CBI-12 is associated with significantly decreased latency to burst onset in buccal premotor neurons and motor neurons, increased cycle frequency of ingestion buccal motor programs (iBMPs) and increased intraburst firing frequency of buccal neurons during iBMPs. Tests of paired-pulse facilitation during AUG/PTP suggest that the locus for this plasticity is presynaptic. The AUG/PTP is not elicited by heterosynaptic pathways, indicating that its origin is homosynaptic. At low CBI-2 and CBI-12 firing frequencies, STE is likely to contribute to iBMP initiation, while at higher firing frequencies, STE is correlated with increased cycle frequency of iBMPs. Thus, STE is an important component of the mechanisms whereby cerebral neurons regulate cyclic feeding programs and likely contributes to observed variations in behavioral responses, including feeding arousal. Twenty-seven oxygenated xanthones have been assessed for their capacity to inhibit in vitro the growth of three human cancer cell lines, MCF-7 (breast cancer), TK-10 (renal cancer) and UACC-62 (melanoma). The effect of these xanthones on the proliferation of human T-lymphocytes was also evaluated. Differences on their potency towards the effect on the growth of the human cancer cell lines as well as on the proliferation of human T-lymphocytes can be ascribed to the nature and positions of the substituents on the xanthonic nucleus.  相似文献

The maturation in the vasodilator response to nitric oxide (NO) in isolated intrapulmonary arteries was analyzed in newborns and 15- to 20-day-old piglets. The vasodilator responses to NO gas but not to the NO donor sodium nitroprusside increased with age. The inhibitory effects of the superoxide dismutase inhibitor diethyldithiocarbamate and xanthine oxidase plus hypoxanthine and the potentiation induced by superoxide dismutase and MnCl(2) of NO-induced vasodilatation were similar in the two age groups. Diphenyleneiodonium (NADPH oxidase inhibitor) potentiated the response to NO, and this effect was more pronounced in the older animals. The nonselective cyclooxygenase inhibitors indomethacin and meclofenamate and the preferential cyclooxygenase-1 inhibitor aspirin augmented NO-induced relaxation specifically in newborns, whereas the selective cycloxygenase-2 inhibitor NS-398 had no effect. The expressions of alpha-actin, cycloxygenase-1, and cycloxygenase-2 proteins were similar, whereas Cu,Zn-superoxide dismutase decreased with age. Therefore, the present data suggest that the maturational increase in the vasodilatation of NO in the pulmonary arteries during the first days of extrauterine life involves a cycloxygenase-dependent inhibition of neonatal NO activity.  相似文献
