Delivery of DNA and siRNA into mammalian cells is a powerful technique in treating various diseases caused by single gene defects. Herein, we report a highly efficient delivery system using 1,4-butanediol diglycidyl ether (bisepoxide) crosslinked polyethylenimine (PEI) nanoparticles (PN). The nanoparticle/DNA complexes (nanoplexes) exibited approximately 2.5- to 5.0-fold gene transfer efficacy and decreased cytotoxicity in cultured cell lines, compared to the native PEI (25 kDa) (gold standard) and commercially available transfection agents such as Lipofectamine 2000 and Fugene. The bisepoxide crosslinking results in change in amine ratio in PEI; however, it retains the net charge on PN unaltered. A series of nanoparticles obtained by varying the degree of crosslinking was found to be in the size range of 69-77 nm and the zeta potential varying from +35 to 40 mV. The proposed system was also found to deliver siRNA efficiently into HEK cells, resulting in approximately 70% suppression of the targetted gene (GFP).  相似文献

A facile synthesis of 1-fluoro-1-deoxy-Delta(8)-THC analogs with side chains seven carbons in length, in the alkane/ene/yne- series (6, 5, and 4), was achieved from 1-fluoro-3,5-dimethoxybenzene (1). In vitro studies show that substitution by a fluorine has a significant detrimental effect on CB1 binding which is supported by in vivo testing. The implications of these results on the SAR of classical cannabinoids are discussed.  相似文献

Electrochemistry of microperoxidase-11 (MPx-11) anchored on the mixed self-assembled monolayer (SAM) of 2-(2-mercaptoethylpyrazine) (PET) and 4,4'-dithiodibutyric acid (DTB) on gold (Au) electrode and the biosensing of uric acid (UA) is described. MPx-11 has been covalently anchored on the mixed SAM of PET and DTB on Au electrode. MPx-11 on the mixed self-assembly exhibits reversible redox response characteristic of a surface confined species. The heterocyclic ring of PET promotes the electron transfer between the electrode and the redox protein. The apparent standard rate constant kapps obtained for the redox reaction of MPx-11 on the mixed monolayer is approximately 2.15 times higher than that on the single monolayer of DTB modified electrode. MPx-11 efficiently mediates the electrocatalytic reduction of H2O2. MPx-11 electrode is highly sensitive to H2O2 and it shows linear response for a wide concentration range. The electrocatalytic activity of the MPx-11 electrode is combined with the enzymatic activity of uricase (UOx) to fabricate uric acid biosensor. The bienzyme assembly is highly sensitive towards UA and it could detect UA as low as 2 microM at the potential of -0.1 V. The biosensor shows linear response with a sensitivity of 3.4+/-0.08 nA cm(-2) microM(-1). Ascorbate (AA) and paracetamol (PA) do not significantly interfere in the amperometric sensing of UA.  相似文献

Integrated use of inorganic fertilizer N and well decomposed cattle manure (CM) or 30-35 days old Parthenium (Parthenium hysterophorus L.), a weed grown off site as green manure (GM) under repeated applications of fertilizer P and urea N for eight years in a rice (Oriza sativa L.)-wheat (Triticum estivum L.) sequence was studied on transformation of fertilizer P applied to soil at the National Research Center for Weed Science, Jabalpur, India. Based on the results, it appeared that, repeated applications of 52 kg super-phosphate P resulted in a marked increase in Olsen P linearly with time. Conjunctive use of urea fertilizer N with organic manure resulted in a larger increase in Olsen P in the Vertisol. Studies further revealed that the greater accumulation of fertilizer P applied in excess to crop removal occurred in inorganic P in the plots receiving only fertilizer N. However, plots receiving fertilizer N along with organic manures led to P accumulation predominantly in organic forms. The study suggests that these two pools of P acted as a sink when fertilizer P was applied in excess to crop removal and are bio-chemically active. The Olsen P status after 8 cycles of rice-wheat crops revealed that the average amount of fertilizer P required after adjusting for crop uptake to increase Olsen P by 1 mg kg(-1) soil was 7.2 kg Pha(-1) in the plots receiving only fertilizer N. Whereas, application of 5t FYM or 6t GM reduced it to 4.6 kg Pha(-1). The plots receiving manure always maintained a greater concentration of Olsen P. The application of CM or GM with fertilizer N enriched short-term inorganic P as well as long-term organic P fertility. After eight years, larger concentrations of organic P in the subsurface layer (16-30 cm), compared to initial values, indicates downward movement of P in organic forms.  相似文献

Enduring forms of synaptic plasticity are thought to require ongoing regulation of adhesion molecules, such as N-cadherin, at synaptic junctions. Little is known about the activity-regulated trafficking of adhesion molecules. Here we demonstrate that surface N-cadherin undergoes a surprisingly high basal rate of internalization. Upon activation of NMDA receptors (NMDAR), the rate of N-cadherin endocytosis is significantly reduced, resulting in an accumulation of N-cadherin in the plasma membrane. Beta-catenin, an N-cadherin binding partner, is a primary regulator of N-cadherin endocytosis. Following NMDAR stimulation, beta-catenin accumulates in spines and exhibits increased binding to N-cadherin. Overexpression of a mutant form of beta-catenin, Y654F, prevents the NMDAR-dependent regulation of N-cadherin internalization, resulting in stabilization of surface N-cadherin molecules. Furthermore, the stabilization of surface N-cadherin blocks NMDAR-dependent synaptic plasticity. These results indicate that NMDAR activity regulates N-cadherin endocytosis, providing a mechanistic link between structural plasticity and persistent changes in synaptic efficacy.  相似文献

Ganges river dolphin abundance has undergone a predominant decline across its range since monitoring began. In Nepal, disappearance from some of the rivers it once used has already occurred. Today this species can only be found in three river systems in Nepal, the Karnali, Sapta Koshi, and Narayani, but numbers are low in these locations. To determine the abundance of dolphins remaining in the Karnali system (which includes the Karnali, Geruwa, and Mohana), and factors affecting dolphin habitat use, we conducted surveys where we recorded dolphin presence. Dolphins within this river system were sighted only in the Karnali and an abundance estimate of 5.04 ± 0.753 SE was calculated. This pattern of ranging differed from that previously reported (from previous sightings only in the Geruwa to current sightings only in the Karnali). River depth likely contributed to the presence or absence of dolphins. Shifts in available habitat between the Geruwa and Karnali have resulted from changes in the course of the main stream Karnali following construction of the Chisapani irrigation intake. Because of the low numbers of dolphins reported, there is great concern that loss of this species in Nepal is likely in the near future.  相似文献

Bacterial blight (BB) caused by Xanthomonas oryzae pv. oryzae (Xoo) is a major disease of rice in the tropics for which genetic resistance in the host plants is the only effective solution. This study aimed at identification of resistance gene combinations effective against Xoo isolates and fingerprinting of the Xoo isolates of Andaman Islands (India). Here, we report the reaction of 21 rice BB differentials possessing Xa1 to Xa21 genes individually and in different combinations to various isolates of pathogen collected from Andaman Islands. Pathological screening results of 14 isolates revealed that among individual genes tested across 2 years, Xa4, Xa7 and Xa21 conferred resistance reaction across all isolates, whereas among combinations, IRBB 50 (Xa4 + xa5), IRBB 52 (Xa4 + Xa21) and IRBB 60 (Xa4 + xa5 + xa13 + Xa21) conveyed effective resistance against tested isolates. The nature of genetic diversity among four isolates selected on the basis of geographical isolation in the islands was studied through DNA finger printing. The RAPD primers S111, S119, S1117, S1109, S1103, S109 and S105 were found to be better indicators of molecular diversity among isolates than JEL primers. The diversity analysis grouped 14 isolates into three major clusters based on disease reaction wherein isolate no. 8 was found the most divergent as well as highly virulent. The remaining isolates were classified into two distinct groups. The importance of the study in the context of transfer of resistance gene(s) in the local cultivars specifically for tropical island conditions is presented and discussed.  相似文献
