Background

Hepatitis C virus (HCV) is one of the leading causes of viral hepatitis worldwide and its genotype 3a is predominant in vast areas of Pakistan.

Findings

The present study reports the first full sequence of HCV 3a isolate PK-1 from Pakistan. This nucleotide sequence was compared with six other HCV genotype 3a full length sequences from different regions of the world by using statistical methods of phylogenetic analysis.

Conclusion

The nucleotide difference of these seven sequences shows that HCV genotype 3a of phylogenetically distinct origin is circulating in Pakistan.  相似文献

Chronic wasting disease (CWD) is a prion disease affecting cervids. Polymorphisms in the prion protein gene can result in extended survival of CWD-infected animals. However, the impact of polymorphisms on cellular prion protein (PrPC) and prion properties is less understood. Previously, we characterized the effects of a polymorphism at codon 116 (A>G) of the white-tailed deer (WTD) prion protein and determined that it destabilizes PrPC structure. Comparing CWD isolates from WTD expressing homozygous wild-type (116AA) or heterozygous (116AG) PrP, we found that 116AG-prions were conformationally less stable, more sensitive to proteases, with lower seeding activity in cell-free conversion and reduced infectivity. Here, we aimed to understand CWD strain emergence and adaptation. We show that the WTD-116AG isolate contains two different prion strains, distinguished by their host range, biochemical properties, and pathogenesis from WTD-116AA prions (Wisc-1). Serial passages of WTD-116AG prions in tg(CerPrP)1536+/+ mice overexpressing wild-type deer-PrPC revealed two populations of mice with short and long incubation periods, respectively, and remarkably prolonged clinical phase upon inoculation with WTD-116AG prions. Inoculation of serially diluted brain homogenates confirmed the presence of two strains in the 116AG isolate with distinct pathology in the brain. Interestingly, deglycosylation revealed proteinase K-resistant fragments with different electrophoretic mobility in both tg(CerPrP)1536+/+ mice and Syrian golden hamsters infected with WTD-116AG. Infection of tg60 mice expressing deer S96-PrP with 116AG, but not Wisc-1 prions induced clinical disease. On the contrary, bank voles resisted 116AG prions, but not Wisc-1 infection. Our data indicate that two strains co-existed in the WTD-116AG isolate, expanding the variety of CWD prion strains. We argue that the 116AG isolate does not contain Wisc-1 prions, indicating that the presence of 116G-PrPC diverted 116A-PrPC from adopting a Wisc-1 structure. This can have important implications for their possible distinct capacities to cross species barriers into both cervids and non-cervids.  相似文献

Immunosenescence, oxidative stress, and low vaccine efficacy are important symptoms of aging. The goal of our study was to test if quercetin had antiaging and stimulating effects on peripheral blood mononuclear cell (PBMC) immune cells in vitro in the presence of concanavalin a, PBMCs were isolated from healthy elderly and young people and cultured in a complete Roswell Park Memorial Institute 1640 medium supplemented with quercetin. Cell proliferation was assessed using the 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium bromide colorimetric assay after a 48-h incubation period. Spectrophotometric assays were used to assess oxidative biomarkers (proteins carbonyl [PC], malondialdehydes [MDA], and reduced glutathione [GSH]). The enzyme-linked immunosorbent assay method was used to determine the amount of interleukin (IL)-2 released. A Griess reagent was used to investigate inducible nitrite oxide synthase (iNOS) activity. When compared to young control cells, aged PBMCs had lower proliferation potency, lower IL-2 and NO release, and higher MDA and PC levels. Importantly, quercetin-treated aged PBMCs have a high proliferative response comparable to young cells, restored iNOS activity, and increased levels of GSH antioxidant defences. In comparison to untreated aged PBMCs, treated PBMCs have lower lipo-oxidative damage but higher PC levels. Quercetin may be used as a promising dietary vaccinal adjuvant in the elderly, it has significant effects in reducing immunosenescence hallmarks, as well as mitigating the lipo-oxidative stress in PBMCs cells.  相似文献

The evolution of gastric carcinogenesis remains largely unknown. We established two gastric carcinogenesis models in New-World nonhuman primates. In the first model, ACP03 gastric cancer cell line was inoculated in 18 animals. In the second model, we treated 6 animals with N-methyl-nitrosourea (MNU). Animals with gastric cancer were also treated with Canova immunomodulator. Clinical, hematologic, and biochemical, including C-reactive protein, folic acid, and homocysteine, analyses were performed in this study. MYC expression and copy number was also evaluated. We observed that all animals inoculated with ACP03 developed gastric cancer on the 9(th) day though on the 14(th) day presented total tumor remission. In the second model, all animals developed pre-neoplastic lesions and five died of drug intoxication before the development of cancer. The last surviving MNU-treated animal developed intestinal-type gastric adenocarcinoma observed by endoscopy on the 940(th) day. The level of C-reactive protein level and homocysteine concentration increased while the level of folic acid decreased with the presence of tumors in ACP03-inoculated animals and MNU treatment. ACP03 inoculation also led to anemia and leukocytosis. The hematologic and biochemical results corroborate those observed in patients with gastric cancer, supporting that our in vivo models are potentially useful to study this neoplasia. In cell line inoculated animals, we detected MYC immunoreactivity, mRNA overexpression, and amplification, as previously observed in vitro. In MNU-treated animals, mRNA expression and MYC copy number increased during the sequential steps of intestinal-type gastric carcinogenesis and immunoreactivity was only observed in intestinal metaplasia and gastric cancer. Thus, MYC deregulation supports the gastric carcinogenesis process. Canova immunomodulator restored several hematologic measurements and therefore, can be applied during/after chemotherapy to increase the tolerability and duration of anticancer treatments.  相似文献

Predators exert strong selection pressures on their prey. Prey would therefore benefit by adjusting their behaviour to the risk of predation, while predators conversely would benefit from adjusting their behaviour to that of their prey. Extravagant ornamentation has evolved to attract mates and/or successfully compete with conspecifics of the same sex to secure high mating success, even if that occurs at a cost of increased risk of predation. Thus, sexually dichromatic species may be more susceptible to predation than sexually monochromatic species, and the presence of compensation is indicative of such species being more vulnerable. If extravagant ornamentation is costly in terms of predation risk, then we should expect sexually dichromatic species to have longer flight initiation distances (FID) than sexually monochromatic species. If ornamentation is acquired as a handicap with only individuals in prime condition being able to display with the smallest viability cost, we should expect sexually dichromatic individuals to have shorter FID than sexually monochromatic individuals. Such differences among individuals should, on an evolutionary time scale, translate into differences in FID being related to differences in sexual dichromatism among species. We investigated the relationship between FID and sexual dichromatism in phylogenetic analyses, while accounting for effects of continent (Australia, North America, and Europe), body mass, the interaction between sexual dichromatism and body mass and the interaction between sexual dichromatism and continent. In an analysis of 447 species we found shorter FID in sexually dichromatic than in sexually monochromatic species (consistent with the handicap hypothesis because sexually dichromatic species took greater risks), especially so at large body size. FID differed among continents and the relative difference in FID between sexually monochromatic and sexually dichromatic species was larger in Europe than in Australia and North America. These differences among continents may be attributed to latitudinal effects of predation. These findings are important for current ideas about the evolution of secondary sexual characters because they imply covarying continental differences in predation, especially for large bodied sexually dichromatic species.  相似文献

Competition between species is ubiquitous in nature and therefore widely studied in ecology through experiment and theory. One of the central questions is under which conditions a (rare) invader can establish itself in a landscape dominated by a resident species at carrying capacity. Applying the same question with the roles of the invader and resident reversed leads to the principle that “mutual invasibility implies coexistence.” A related but different question is how fast a locally introduced invader spreads into a landscape (with or without competing resident), provided it can invade. We explore some aspects of these questions in a deterministic, spatially explicit model for two competing species with discrete non-overlapping generations in a patchy periodic environment. We obtain threshold values for fragmentation levels and dispersal distances that allow for mutual invasion and coexistence even if the non-spatial competition model predicts competitive exclusion. We obtain exact results when dispersal is governed by a Laplace kernel. Using the average dispersal success, we develop a mathematical framework to obtain approximate results that are independent of the exact dispersal patterns, and we show numerically that these approximations are very accurate.  相似文献

Phosphogypsum (CaSO4), a primary by-product of phosphoric acid production, is accumulated in large stockpiles and occupies vast areas of land. It poses a severe threat to the quality of water and land in countries producing phosphoric acid. In this study, the potential of sulfate-reducing bacteria for biodegradation of this sulfur-rich industrial solid waste was assessed. The effect of phosphogypsum concentration, carbon and nitrogen sources, temperature, pH and stirring on the growth of sulfate-reducing bacteria was investigated. Growth of sulfate-reducing bacteria was monitored by measuring sulfide production. Phosphogypsum was shown to be a good source of sulfate, albeit that the addition of organic carbon was necessary for bacterial growth. Biogenic sulfide production occurred with phosphogypsum up to a concentration of 40 g L−1, above which no growth of sulfate-reducing bacteria was observed. Optimal growth was obtained at 10 g L−1 phosphogypsum. Both the gas mixture H2/CO2 and lactate supported high amounts of H2S formation (19 and 11 mM, respectively). The best source of nitrogen for sulfate-reducing bacteria was yeast extract, followed by ammonium chloride. The presence of nitrate had an inhibitory effect on the process of sulfate reduction. Stirring the culture at 150 rpm slightly stimulated H2S formation, probably by improving sulfate solubility.  相似文献

Two series of new 6-alkoxy-4-substituted-aminoquinazolines (2-4f) and their bioisoteric quinoline congeners (5-7c) were designed and synthesized. Virtual screening was carried out through docking the designed compounds into the ATP binding site of epidermal growth factor receptor (EGFR) to predict if these compounds have analogous binding mode to the EGFR inhibitors. The newly synthesized compounds were tested in vitro on human breast carcinoma cell line (MCF-7) in which EGFR is highly expressed. Most of the tested compounds exploited potent antitumor activity with IC(50) values in the nanomolar range in particular compound 3b which displayed the highest activity among the tested compounds with IC(50) equal to 0.13 nmol.  相似文献
