Calf skin acid-soluble collagen, containing about 34 residues of lysine plus hydroxylysine per 100,000 dalton polypeptide chain, was treated with [14C]glucose in the presence or absence of NaCNBH3. In 144 h, under the conditions employed, the presence of NaCNBH3 increased the extent of glycosylation from 8 to 15% of the total residues of lysine plus hydroxylysine. The extent of glycosylation was estimated, using acid hydrolysates of the protein, by isolation and determination of reduced adducts (1-lysinohexitols) employing a system of paper chromatography followed by chromatography on an amino acid analyzer. By those means the difficulties of using specific color reactions such as that with thiobarbituric acid were obviated. Identification of the reduced adducts as forms of 1-lysinohexitol was made by comparison of that substance prepared by treatment of polylysine with [14C]glucose in the presence of NaCNBH3. Of interest is the fact that treatment of the polymer with glucose for 144 h under conditions similar to those used for the collagen, resulted in an increase of extent of glycosylation from 3 to about 50% of the total lysine residues when NaCNBH3 was present in the incubation medium. The greater degree of glycosylation of lysine residues in polylysine as compared with collagen (15 versus about 50%) may be ascribed to the different orders of macromolecular structure in the protein that could sequester certain of the residues from reaction with glucose. 1-Lysinohexitol was also identified in hydrolysates of neutral salt-soluble guinea pig skin collagen that had been reacted with glucose and then treated with NaB3H4. The glycosylated collagens were fragmented by treatment with CNBr, and modified lysine residues were found to occur along the entire length of the collagen chains. The use of NaCNBH3 in the manner described above permits measurement of both aldimine and ketoamine forms of the adducts made with glucose. The possible physiological significance of the reversibility of the ketoamine form of adduct is discussed briefly. 相似文献

Malonyl-CoA synthetase fromPseudomonas fluorescens was inactivated by diethylpyrocarbonate (DEP) with the second-order rate constant of 775 M–1 min–1 atpH 7.0, 25°C, showing a concomitant increase in absorbance at 242 nm due to the formation of N-carbethoxyhistidyl derivatives. The inactivated enzyme at low concentration of DEP (<0.2 mM) could be completely reactivated by hydroxylamine but not completely reactivated at high concentration (>0.5 mM), indicating that there may be another functional group modified by DEP. strain DC5-E2 that lacks mtDNA () can be cotransformed with a mixture of mtDNA and the plasmid YEp13 (LEU2/2μ/pBR322) to produce Leu+ transormants which, on being mated to tester strains, generate respiratory competent diploids (such events are denoted marker rescue). In this work strain DC5-E2 was transformed with recombinant molecules consisting of a mtDNA segment including the gene inserted into YEp13. The Leu+ transformants made with the recombinant plasmids were unable to rescue testers carrying mutations in the region, in contrast to Leu+ cotransformants made with mixtures of YEp13 and mtDNA. We conclude that marker rescue events occur as a result of interactions between the mtDNA of the tester and the mtDNA sequences introduced by transformation. Such interactions cannot occur when the latter mtDNA is forced to replicate in covalent association with YEp13, probably in the nucleus. 相似文献

We have examined the uptake of creatine by cultured monolayers of human IMR-90 flbroblasts, human uterine smooth muscle cells, calf aortic smooth muscle cells, and myoblasts and myotubes of the L6E9 rat skeletal muscle cell line. Creatine uptake is dependent on temperature and sensitive to the presence of Na+ in the extracellular medium. It is saturable, apparently concentrative, and inhibited by ouabain and structural analogs of creatine. In these respects, it resembles the process of creatine uptake by isolated preparations of skeletal muscle and brain tissues. Lineweaver-Burk plots of the data for variation in rate of uptake with concentration of creatine in the medium are nonlinear, suggesting that the process of uptake may be heterogeneous. Assuming the operation of two saturable processes of uptake, we calculated two values for apparent Km and V for each cell line. Kinetic parameters of creatine uptake by the different cell types are similar. The lower values of Km (0.02–0.04 mm) are in the physiological range of creatine concentration in mammalian plasma. 相似文献

As ultraviolet wavelengths are used in normal avian colour perception, the maintenance of captive birds under artificial lighting (which is normally UV-deficient) may have welfare implications. European starling Sturnus vulgaris juveniles kept in UV-deficient light environments had significantly higher basal plasma corticosterone concentrations than those kept under full spectrum lighting, in the second of two experimental blocks. UV-deficient conditions also led to significant changes in behaviour indicative of escape (less perching and more hanging on the cage and pecking at it). However, the birds from the first block, where the interval between transfer to the experimental set-up from the wild was short (2 days), showed significantly higher basal and maximum plasma corticosterone concentrations than those in the second block and no additional effect of light environment on either corticosterone or behaviour. We hypothesise that this difference between blocks was due to the overriding initial stress of being in captivity swamping any treatment effects. Capture stress had declined in the second set of birds, which entered the experiment after 7–14 days in captivity. Stress effects of UV-deficient lighting appear small relative to the overall impact of captivity, but may nevertheless become apparent after the initial effects of capture subside. 相似文献

In this study, the effect of different preparations made from Bacillus thuringiensis var. thuringiensis (strains: CCEB 555 and CCEB 058) on ants, Monomorium pharaonis, under laboratory conditions is reported. The different preparations tested consisted of (1) a liquid culture of the strain B. thuringiensis CCEB 555 (containing spores and exotoxin), (2) the supernatant of the culture broth of strain CCEB 555 (containing exotoxin), and (3) the biological preparation “Bathurin” prepared from the strain B. thuringiensis CCEB 058 (containing spores and inclusions, without exotoxin). The preparations were used either pure or in alternation with borax, i.e., 1 wk borax, 3 wk the respective preparation for several months. All preparations were found to be toxic to M. pharaonis and their effect was characterized by a slow extinction of the ant colony. Administration of “Bathurin” (1.3%) yielded a 100% mortality after 20 wk. Using a liquid culture of B. thuringiensis var. thuringiensis, 100% mortality was recorded after 21 wk, a period of time which did not differ from that obtained with the supernatant of the culture containing exotoxin. The alternation with borax was found to accelerate ant mortality by 9–10 wk after administration. In all experiments, the worker ants died first, the queen ants surviving them by 1–3 wk.In experiments employing worker ants only, a 100 and 98% mortality, respectively, occurred within 3 wk after administration of a liquid culture of B. thuringiensis and “Bathurin” supplemented with borax. 相似文献

A survey of 466 Caribbean lizards found Plasmodium parasites present in Anolis species only of five islands. Parasites presently considered to be P. floridense occurred on Grand Cayman, North Bimini, Jamaica, Hispaniola (Haiti), and Puerto Rico. A second species, P. azurophilum, is described as new from Anolis cybotes of Haiti, A. krugi of Puerto Rico, and A. lineatopus and A. grahami of Jamaica. It lacks visible pigment in erythrocytic host cells but can produce it occasionally. Both asexual and sexual forms occur in a variety of white blood cells, notably in azurophil granulocytes and polymorphonuclear leucocytes. Experimental infections indicate that the leucocytic phase occurs after the acute erythrocytic infection declines, thus suggesting that the schizogonic and gametogonic cycles in white cells may represent an adaptive defense against immune mechanisms of the host. Mean numbers of nuclei in schizonts and mean gametocyte size are influenced by host species and type of host cell. 相似文献

The possibility whether alterations in the cyclic AMP-adenylate cyclase-phosphodiesterase system play a role in the action of 1,1,1-trichloro-2,2-bis-(p-chlorophenyl)ethane (DDT) on hepatic and renal carbohydrate metabolism was investigated. Administration of exogenous cyclic AMP (10mg/100g) was found to mimic the action of DDT which enhanced the activities of pyruvate carboxylase, phosphoenolpyruvate carboxylase, fructose 1,6-diphosphatase and glucose 6-phosphatase in both liver and kidney cortex, elevated the concentration of blood glucose and urea and decreased the amount of hepatic glycogen. Treatment with theophylline augmented the effects of a submaximal dose of this halogenated hydrocarbon on serum urea and glucose as well as the key gluconeogenic enzymes in liver and kidney cortex. Addition of DDT in vitro to liver and kidney homogenates resulted in a significant enhancement of adenylate cyclase activity. Hepatic and renal slices from rats already treated with DDT displayed an increased ability to convert [(3)H]adenosine into cyclic [(3)H]AMP. Whereas kidney-cortex slices excised from rats given caffeine and DDT produced an even greater amount of cyclic [(3)H]AMP, imidazole, propranolol and hydrazine prevented the insecticide-stimulated rise in cyclic nucleotide production. In contrast, prostaglandin E(1) failed to exert any significant effect on DDT-induced increases in cyclic [(3)H]AMP synthesis from radioactive adenosine. The present study and our previous findings (Kacew & Singhal, 1973e) support the concept that the DDT-induced alterations in carbohydrate metabolism of liver and kidney cortex may be related to an initial stimulation of the cyclic AMP-adenylate cyclase system in these tissues. 相似文献

“Capillary permeability” to serum albumin has been measured in patients with collagen vascular diseases by a method which compares the dilution of intravenously injected 131I-human serum albumin and 51Cr-R.B.C.s. The results indicate an increased capillary permeability comparable to that which occurs in patients with extensive inflammatory skin disease. We suggest that this increased capillary permeability may be the cause of the episodes of oedema which occur in patients with collagen vascular diseases such as disseminated lupus erythematosus, systemic sclerosis, dermatomyositis, polyarteritis nodosa, and rheumatoid arthritis. “Spontaneous periodic oedema” may be the presenting feature of collagen vascular disease and is due to increased capillary permeability. 相似文献
