选择沙漠常绿灌木丛枝霸王(Zygophyllum dumosum)群落及群落间的开阔地带采样,研究土壤线虫和原生动物的种群大小、物种组成及营养结构.本研究共获线虫29属,裸变形虫33属90种;在所获各类原生动物中,鞭毛虫和纤毛虫均为食细菌类群,而鞭毛虫个体数量最多;在沙漠生态系统中,土壤湿度被认为是影响生物活性的最重要因素之一.然而,本研究显示:土壤湿度对线虫和原生动物是否重要取决于这些动物对土壤湿度的生理生态适应能力;土壤线虫和原生动物的体型大小、生活史策略和活动能力与土壤湿度的有效性相关不显著,但它们对微生物和微小土壤动物区系间的相互作用有重要影响,在干旱生态系统中,这种影响能够显著改变土壤生态系统短期的养分循环.本研究还表明,土壤线虫和原生动物种群间的关系主要取决于两者营养类群组成的变化.  相似文献

Protein misfolding and deposition underlie an increasing number of debilitating human disorders and constitute a problem of major concern in biotechnology. In the last years, in vitro studies have provided valuable insights into the physicochemical principles underlying protein aggregation. Nevertheless, information about the determinants of protein deposition within the cell is scarce and only a few systematic studies comparing in vitro and in vivo data have been reported. Here, we have used the SH3 domain of α-spectrin as a model globular protein in an attempt to understand the relationship between protein aggregation in the test-tube and in the more complex cellular environment. The investigation of the aggregation in Escherichia coli of this domain and a large set of mutants, together with the analysis of their sequential and conformational properties allowed us to evaluate the contribution of different polypeptidic factors to the cellular deposition of globular proteins. The data presented here suggest that the rules that govern in vitro protein aggregation are also valid in in vivo contexts. They also provide relevant insights into intracellular protein deposition in both conformational diseases and recombinant protein production.  相似文献

The RIC-3 protein acts as a regulator of acetylcholine nicotinic receptor (nAChR) expression. In Xenopus laevis oocytes the human RIC-3 (hRIC-3) protein enhances expression of α7 receptors and abolishes expression of α4β2 receptors. In vitro translation of hRIC-3 evidenced its membrane insertion but not the role as signal peptide of its first transmembrane domain (TMD). When the TMDs of hRIC-3 were substituted, its effects on nAChR expression were attenuated. A certain linker length between the TMDs was also needed for α7 expression enhancement but not for α4β2 inhibition. A combination of increased α7 receptor steady state levels, facilitated transport and reduced receptor internalization appears to be responsible for the increase in α7 membrane expression induced by hRIC-3. Antibodies against hRIC-3 showed its expression in SH-SY5Y and PC12 cells and its induction upon differentiation. Immunohistochemistry demonstrated the presence of RIC-3 in rat brain localized, in general, in places where α7 nAChRs were found.  相似文献

Binding of agonists to nicotinic acetylcholine receptors (nAChR) is coupled to channel opening through local rearrangements of different domains of the protein. Recent structural data suggest that two of these regions could be the loop 5 (L5) and the β-strand β6', both forming the inner part of the N-terminal domain. Amino acids in these domains were mutated in α7 nAChRs, and expression levels and functional responses of mutant receptors were measured. Mutations located at the putative apex of L5, Asp97 and Glu98, and also at Phe100, gave receptors with smaller currents, showing qualitative differences with respect to muscle nAChRs. In contrast, mutations in the β-strand β6' (at Phe124 and Lys125) showed increased functional responses. Mutations affected equally the responses to acetylcholine and dimethylphenylpiperazinium, except in Phe100 where the latter was sevenfold less effective than in wild-type. Currents in mutants decayed with almost the same kinetics, ruling out large effects on desensitization. Analysis of double mutants demonstrated a functional coupling among the three electrically charged amino acids Asp97, Glu98, and Lys125, and also between Phe100 and Phe124. The results are compatible with the involvement of functional interactions between L5 and β-strand β6' during nAChR activation.  相似文献

Abstract. Research into interspecific variation in functional traits is important for our understanding of trade‐offs in plant design and function, for plant functional type classifications and for understanding ecosystem responses to shifts in species composition. Interspecific rankings of functional traits are a function of, among other factors, ontogenetic or allometric development and environmental effects on phenotypes. For woody plants, which attain large size and long lives, these factors might have strong effects on interspecific trait rankings. This paper is the first to test and compare the correspondence of interspecific rankings between laboratory grown seedlings and field grown adult plants for a wide range of functional leaf and stem traits. It employs data for 90 diverse woody and semiwoody species in a temperate British and a (sub)Mediterranean Spanish flora, all collected according to a strict protocol. For 12 out of 14 leaf and stem traits we found significant correlations between the species ranking in laboratory seedlings and field adults. For leaf size and maximum stem vessel diameter > 50 % of variation in field adults was explained by that in laboratory seedlings. Two important determinants of plant and ecosystem functioning, specific leaf area and leaf N content, had only 27 to 36 and 17 to 31 % of variation, respectively, in field adults explained by laboratory seedlings, owing to subsets of species with particular ecologies deviating from the general trend. In contrast, interspecific rankings for the same traits were strongly correlated between populations of field adults on different geological substrata. Extrapolation of interspecific trait rankings from laboratory seedlings to adult plants in the field, or vice versa, should be done with great caution.  相似文献

Aristolochia is the largest genus of the family Aristolochiaceae and the only one with large chromosome number variation. A combination of fluorochrome banding and in situ hybridization of 5S and 45S rDNA probes was used to evaluate the structural karyotype variability of representatives of two subgenera: Siphisia, which seems to have a single chromosome number (2n = 32), probably derived from an old polyploidization event, and Aristolochia, including the Old World section Diplolobus and the New World Gymnolobus. Based on chromosome morphology and on the degree of diploidization of rDNA sites, A. serpentaria (Siphisia) was identified as an old hexaploid, whereas A. paucinervis (Diplolobus) seemed to be a recent hexaploid (2n = 34). The karyotypes of the five analyzed species of section Gymnolobus were structurally more stable than those from Diplolobus, which varied considerably in the type of heterochromatin, chromosome number, and morphology. These data indicate that fluorochrome banding and rDNA localization may substantially improve the cytotaxonomical analysis of this genus.  相似文献

The aim of this work was to determine whether the stimulating effect of glucagon-like peptide (GLP)-2 on astrocyte proliferation could be reinforced by proliferating substances, including growth factors such as EGF, platelet-derived growth factor, insulin-like growth factor type I (IGF-I) or a hormone such as insulin. Both DNA synthesis and astrocyte density, as well as the expression of c-Fos, Ki-67, proliferating cell nuclear antigen and glial fibrillary acidic proteins, were found to be higher in the presence of GLP-2 than in its absence. In an attempt to get a better understanding of this process, intracellular cyclic adenosine monophosphate (cAMP) production, extracellular signal-regulated kinase (ERK) 1/2 phosphorylation and the expression of GLP-2R and IGF-I receptor (IGF-IR) mRNAs were studied in response to growth factors. Our results indicate that, in the presence of different growth factors, GLP-2 does not increase cAMP production but raises ERK 1/2 phosphorylation. In addition, GLP-2R mRNA expression was increased by IGF-I, whilst mRNA expression of IGF-IR was higher in cells incubated with GLP-2 than in control cells. These results suggest for the first time that GLP-2 and several growth factors show synergistic effects on the proliferation of rat astrocytes, a process in which an enhanced expression of GLP-2R and IGF-IR may be involved, providing additional insights into the physiological role of this novel neuropeptide, specially during astroglial regeneration.  相似文献

The present study describes the chemical composition and the antibacterial activity of extracts from Cordia verbenacea DC (Borraginaceae), a traditional medicinal plant that grows widely along the southeastern coast of Brazil. The extracts were obtained using different extraction techniques: high-pressure operations and low-pressure methods. The high-pressure technique was applied to obtain C. verbenacea extracts using pure CO2 and CO2 with co-solvent at pressures up to 30 MPa and temperatures of 30, 40 and 50 °C. Organic solvents such as n-hexane, ethyl acetate, ethanol, acetone and dichloromethane were used to obtain extracts by low-pressure processes. The antibacterial activity of the extracts was also subjected to screening against four strains of bacteria using the agar dilution method. The extraction yields were up to 5.0% w/w and up to 8.6% w/w for supercritical fluid extraction with pure CO2 and with ethyl acetate as co-solvent, respectively, while the low-pressure extraction indicates yields up to 24.0% w/w in the soxhlet extraction using water and aqueous mixture with 50% ethanol as solvents. The inhibitory activity of the extracts in Gram-positive bacteria was significantly higher than in Gram-negative. The quantification and the identification of the extracts recovered were accomplished using GC/MS analysis. The most important components identified in the extract were artemetin, β-sitosterol, α-humulene and β-caryophyllene, among others.  相似文献

The mitochondrial respiratory chain of Ustilago maydis contains two terminal oxidases, the cytochrome c oxidase (COX) and the alternative oxidase (AOX). To understand the biochemical events that control AOX activity, we studied the regulation and function of AOX under oxidative stress. The activity of this enzyme was increased by both pyruvate (K05 = 2.6 mM) and purine nucleotides (AMP, K05 = 600 μM) in mitochondria using succinate as respiratory substrate. When U. maydis cells were grown in the presence of antimycin A, the amount of AOX in mitochondria was markedly increased and its selectivity towards AMP and pyruvate changed, suggesting that post-translational events may play a role in the regulation of AOX activity under stress conditions. Addition of antimycin A to isolated mitochondria induced the inactivation of AOX, the formation of lipid peroxides and the loss of glutathione from mitochondria. The two last processes are probably related with the time dependent inactivation of AOX, in agreement with the inhibition of the enzyme by tert-butyl hydroperoxide. Our results suggest that the in vivo operation of AOX in U. maydis depends on the mitochondrial antioxidant machinery, including the glutathione linked systems.  相似文献

Temporary soil flooding before cultivation alleviates iron chlorosis in crops grown on some calcareous Mexican Vertisols. In order to investigate the effectiveness of such practice we carried out experiments with ten calcareous Vertisols from Mexico and eight calcareous Inceptisols from Spain. In an incubation experiment, we studied the release of Fe2+ into the solution of soil suspensions in sealed vials with 5 m M CaCl2. In a pot experiment, we measured the leaf SPAD value (i.e. an estimate of leaf chlorophyll concentration) of lupin and strawberry sequentially grown on a soil-sand mixture previously flooded for 30 days (SPADf value) and on a non-flooded (control) mixture (SPADc value). The amount of Fe2+ released by the soil at day 58 and the increase in oxalate-extractable Fe (Feo) upon incubation in vials were larger on average for the Inceptisols than for the Vertisols. The SPADc values for lupin and strawberry were (i) larger for the Vertisols than for the Inceptisols (probably because the Vertisols contain little carbonate and induce less Fe chlorosis than the Inceptisols) and (ii) correlated with Feo, and with citrate/ascorbate- and DTPA-extractable Fe (Feca, FeDTPA). The SPADf-SPADc differencewas (i) much larger for the Inceptisols than for the Vertisols and (ii) correlated with the increases in Feo and Feca caused by flooding and with the amount of Fe2+ released in the incubation experiment. We hypothesize that the weak response of the Vertisols to flooding was partly a result of their history including flooding episodes in the field, so a steady state had been reached in which the pool of Fe compounds undergoing reductive dissolution and reprecipitating upon oxidation as poorly crystalline Fe oxides (the main source of phytoavailable Fe) remained relatively constant and thus changed little after pot flooding. The Inceptisols, which had never been flooded in the field, were capable of releasing Fe from sources other than poorly crystalline Fe oxides upon flooding, thus making this treatment effective against Fe chlorosis. Our results point to the need to further study those soil chemical and mineralogical properties that are related to increases in Fe phytoavailability upon temporary soil flooding.  相似文献
