Myocardial infarction (MI) is divided into either ST elevation MI (STEMI) or non-ST elevation MI (NSTEMI), differing in a number of clinical characteristics. We sought to identify genetic variants conferring risk to NSTEMI or STEMI by conducting a genome-wide association study (GWAS) of MI stratified into NSTEMI and STEMI in a consecutive sample of 1,579 acute MI cases with 1,576 controls. Subsequently, we followed the results in an independent population-based sample of 562 cases and 566 controls, a partially independent prospective cohort (N = 16,627 with 163 incident NSTEMI cases), and examined the effect of disease-associated variants on gene expression in 513 healthy participants. Genetic variants on chromosome 1p13.3 near the damage-regulated autophagy modulator 2 gene DRAM2 associated with NSTEMI (rs656843; odds ratio 1.57, P = 3.11 × 10−10) in the case-control analysis with a consistent but not statistically significant effect in the prospective cohort (rs656843; hazard ratio 1.13, P = 0.43). These variants were not associated with STEMI (rs656843; odds ratio, 1.11, P = 0.20; hazard ratio 0.97, P = 0.87), appearing to have a pronounced effect on NSTEMI risk. A majority of the variants at 1p13.3 associated with NSTEMI were also associated with the expression level of DRAM2 in blood leukocytes of healthy controls (top-ranked variant rs325927, P = 1.50 × 10−12). The results suggest that genetic factors may in part influence whether coronary artery disease results in NSTEMI rather than STEMI.  相似文献

The Early and Middle Flandrian geological development and paleolimnology of Lake Spitaalijärvi was studied using pollen, diatom and cladoceran analysis and 14C dating. Spitaalijärvi was isolated from the Ancylus Lake about 9000 B.P., at which time birch and pine and plants typical of open habitat communities grew on the solitary island. The rational limits for Alnus and Picea were ca. 8300 B.P. and 3700 B.P., respectively. During the first few hundred years after isolation Spitaalijärvi was probably fairly eutrophic, with a low water level. Water level began to rise before the Alnus rise (A°) and the lake became oligotrophic. After another transgression, which started before the Picea rise (P°), Tabellaria binalis and Semiorbis hemicyclus appear in the diatom stratigraphy indicating ultra-oligotrophic conditions. The main reason for the ultra-oligotrophy of Lake Spitaalijärvi is the character of the parent material in the catchment, consisting of glacial and littoral deposits derived from the local quartz sandstone of highly siliceous nature.  相似文献

Abstract: A bioluminescent assay based on the firefly luciferase reaction has been used for determination of creatine kinase activity in CSF. Activities as low as 0.1 U/L can be measured. The coefficient of variation at an activity level of 0.3–0.4 U/L was between 5 and 6%. The assay conditions optimized for serum specimens can be used for CSF. The adenylate kinase activity is almost completely inhibited, which simplifies the procedure. The creatine kinase (CK) isoenzyme distribution was obtained using the bioluminescent assay in combination with immunoinhibition or ion exchange chromatography. All specimens contained both MM and BB activity, but no MB was found. The study indicates that the bioluminescent assay is useful in the determination of CK isoenzymes in CSF. The clinical importance of the observed CK levels will be reported in a separate communication.  相似文献

The magnitude of inbreeding depression caused by recessive mutations in a population is dependent on the mutation rate and on the intensity of selection against the mutations. We studied geographical differences in the level of early inbreeding depression of Scots pine in a common garden experiment. The mean abortion rate of experimentally self-pollinated seeds was significantly lower (75.4%) among trees that originated from northern populations (66–69°N) than among trees from more southern (60–62°N) populations (86.5%). Thus, the number of embryonic lethal equivalents was lower in the northern populations (4.5) than in the southern ones (6.9). The outcrossing rate at the mature seed stage was slightly lower in the northern populations (average 0.93) than in the southern one (0.99). The estimated selfing rate at the zygote stage varied from 0–0.28 in the populations. The reduction in the magnitude of inbreeding depression in the north may have been caused by increased levels of self-fertilization in the northern populations. The proportion of self-fertilized seedlings and adults was very small in all populations (F ≈ 0), indicating high inbreeding depression also in later life stages. The high level of inbreeding depression in the partially selfing Scots pine can be explained by mutation-selection balance only if the mutation rate is high.  相似文献

An in situ hybridization method using paraffin-embedded sections was used to characterize the chicken oviduct cells synthesizing ovalbumin mRNA due to the action of estrogen and progesterne. The cytodifferentiation of the oviduct cells was induced by 17β-estradiol administration to newly hatched female chicks. To avoid possible effect of estrogen on the action of progesterone the chicks were withdrawn from the estrogen by six days withdrawal period without hormone treatment. Ovalbumin mRNA was not synthesized after a period of estrogen withdrawal. Administration of estrogen induced ovalbumin mRNA in the tubular gland cells. Administration of progesterone induced the expression of ovalbumin mRNA in the surface epithelial cells. It was also found that progesterone induced mucus producing goblet cells in the surface epithelium. Estrogen did not have an effect on the mucus production, which suggests that progesterone could induce the terminal differentiation of the goblet cells. We conclude that the expression of ovalbumin in the surface epithelial cells and in the tubular gland cells is specific for progesterone and estrogen, respectively.  相似文献

Polymorphism in petal colour is common in deceptively pollinated plant species. Most of the deceptively pollinated orchids are food frauds, and in most of them, the deception is not mimetic. These plants have conspicuously coloured flowers which they use as the main attractant of naive pollinators. In a field experiment, we studied the response of bumblebees and other types of flower visitors to colour differences between experimentally paired plants of Dactylorhiza maculata , a nectarless food-deceptive species. In addition, pollen removal, an estimate of male fitness, and fruit production, an estimate of female fitness, were measured in the two colour variants. We found a trend of bumblebee preference for the dark-coloured flowers, but other flower visitors (as a group) showed no preference for any colour variant. No difference was found in the reproductive success between the two colour variants of D. maculata. The lack of a difference in reproductive success between plants with pale and dark inflorescences, despite the observed trend of bumblebee preference for dark inflorescences, suggests that there is some balancing factor in the pollination of the pale inflorescences. An excess of visits by some nocturnal species (or a group of species) which favours the pale colour of D. maculata inflorescences or an excess of visits during day time by some flower visitors other than bumblebees preferring the pale inflorescences over dark ones may form such a balancing factor.  相似文献

Environmental heterogeneity has been intensively studied, but little is known about relationships between habitat patchiness and soil processes. The aim of this study was to investigate (1) the impact of patchiness of the litter layer on the decomposer community and litter decomposition rate, and (2) whether the impact of soil fauna on the rates of processes differs in relation to patchiness. An experiment was carried out in microcosms with coniferous forest humus and four kinds of litter with different C:N ratios or stages of decomposition, either separately (i.e. in patches) or mixed with each other. Microarthropod species diversity was better maintained in the patchy systems. In the absence of soil fauna, community respiration was higher in the patchy microcosms, but in the presence of fauna the opposite pattern was observed. The contribution of soil fauna to the rate of decomposition was clearly greater in the mixed litter systems. Based on the results, a hypothesis is presented that in the patchy litter layer the soil fungi can create connections between different materials located some centimeters apart, thus enhancing decomposition, while in the mixed litter the scale of millimeters is more appropriate for the soil fauna, known to accelerate the process rates. Host organisms are believed to evolve defense mechanisms (i.e., resistance and/or tolerance) under selective pressures exerted by natural enemies. A prerequisite for the evolution of resistance and tolerance is the existence of genetic variation in these traits for natural selection to act. However, selection for resistance and/or tolerance may be constrained by negative genetic correlations with other traits that affect host fitness. We studied genetic variation in resistance and tolerance against parasitic infection and the potential fitness costs associated with these traits using a novel study system, namely the interaction between a flowering plant and a parasitic plant. In this system, parasitic infection has significant negative effects on host growth and reproduction and may thus act as a selective agent. We conducted a greenhouse experiment in which we grew host plants, Urtica dioica, that originated from a single natural population and represented 20 maternal families either uninfected or infected with the holoparasitic dodder, Cuscuta europaea. that originated from the same site. We calculated correlations among resistance, tolerance, and host performance to test for costs of resistance and tolerance. We measured resistance as parasite performance (quantitative resistance) and tolerance as the slopes of regressions relating the vegetative and reproductive biomass of host plants to damage level (measured as parasite biomass). We observed significant differences among host families in parasite resistance and in parasite tolerance in terms of reproductive biomass, a result that suggests genetic variation in these traits. Furthermore, we found differences in resistance and tolerance between female and male host plants. In addition, the correlations indicate costs of resistance in terms of host growth and reproduction and costs of tolerance in terms of host reproduction. Our results thus indicate that host tolerance and resistance can evolve as a response to infection by a parasitic plant and that costs of resistance and tolerance may be one factor maintaining genetic variation in these traits.  相似文献
