We use an Australian freshwater invertebrate species, Daphnia carinata, to assess whether variation in habitat permanence influences life-history traits in subpopulations. Using a life table experiment, we measure the life-history traits of populations from both permanent and temporary pools. We show that these habitat classes are associated with clear differences in important life-history traits and evidence of trade-offs in important traits influencing reproduction, diapause, and growth rate and suggest this is evidence for local adaptation. Here we use Daphnia from Australian populations spanning semi-arid and temperate climates generating results that are in broad agreement with similar studies in the northern hemisphere, and so extend these results to a new continent and its particular climate. Variation in habitat permanence, it appears, is a very general driver of life-history divergence.
The extinction debt, delayed species extinctions following landscape degradation, is a widely discussed concept. But a consensus about the prevalence of extinctions debts is hindered by a multiplicity of methods and a lack of comparisons among habitats. We applied three contrasting species–area relationship methods to test for plant community extinction debts in three habitats which had different degradation histories over the last century: calcareous grassland, heathland and woodland. These methods differ in their data requirements, with the first two using information on past and current habitat area alongside current species richness, whilst the last method also requires data on past species richness. The most data‐intensive, and hence arguably most reliable method, identified extinction debts across all habitats for specialist species, whilst the other methods did not. All methods detected an extinction debt in calcareous grassland, which had undergone the most severe degradation. We conclude that some methods failed to detect an extinction debt, particularly in habitats that have undergone moderate degradation. Data on past species numbers are required for the most reliable method; as such data are rare, extinction debts may be under‐reported. 相似文献
The lipid-rich cell wall of Mycobacterium tuberculosis, the agent of tuberculosis, serves as an effective barrier against many chemotherapeutic agents and toxic host cell effector molecules, and it may contribute to the mechanism of persistence. Mycobacterium tuberculosis strains mutated in a 13-gene operon called mce1, which encodes a putative ABC lipid transporter, induce aberrant granulomatous response in mouse lungs. Because of the postulated role of the mce1 operon in lipid importation, we compared the cell wall lipid composition of wild type and mce1 operon mutant M. tuberculosis H37Rv strains. High resolution mass spectrometric analyses of the mce1 mutant lipid extracts showed unbound mycolic acids to accumulate in the cell wall. Quantitative analysis revealed a 10.7 fold greater amount of free mycolates in the mutant compared to that of the wild type strain. The free mycolates were comprised of alpha, methoxy and keto mycolates in the ratio 1:0.9:0.6, respectively. Since the mce1 operon is regulated in vivo, the free mycolates that accumulate during infection may serve as a barrier for M. tuberculosis against toxic products and contribute to the pathogen’s persistence. 相似文献
Estimates of soil seed banks are important to many ecological investigations and plant conservation, yet seed banks are among the most difficult plant community attributes to accurately quantify. To compare extraction and emergence seed bank characterization methods, we collected 0- to 5-cm soil seed bank samples and measured plant community composition in six microsite types (below different perennial plant species and interspaces) at 10 field sites in the Mojave Desert, USA. Extraction detected five times more species sample?1 and orders of magnitude greater seed density than emergence, though evaluating viability of extracted seed was not straightforward. Only 13 % of 847 tested seeds from extraction emerged in follow-up assays. Considering all sites, species detection was more similar between methods: 21 taxa for emergence and 28 for extraction. Results suggest that: (i) capturing microsite variation is critical for efficiently estimating site-level desert seed banks; (ii) method comparisons hinged on the scale of analysis for species richness, as differences in species detection between methods diminished when increasing resolution from the sample to the regional scale; (iii) combining data from all seed bank methods provided the strongest correlation with vegetation; and (iv) improving knowledge of seed germinability is important for advancing both seed bank methods, including for extraction to evaluate the proportion of extracted seeds that are viable. Multifactor approaches that balance several effectiveness measures (e.g., both seed density and species detection at multiple scales) and procedural challenges are most likely to accurately represent complexity in tradeoffs for choosing methods to quantify soil seed banks. 相似文献
A drawback of targeting soluble antigens such as cytokines or toxins with long-lived antibodies is that such antibodies can prolong the half-life of the target antigen by a “buffering” effect. This has motivated the design of antibodies that bind to target with higher affinity at near neutral pH relative to acidic endosomal pH (~pH 6.0). Such antibodies are expected to release antigen within endosomes following uptake into cells, whereas antibody will be recycled and exocytosed in FcRn-expressing cells. To understand how the pH dependence of antibody-antigen interactions affects intracellular trafficking, we generated three antibodies that bind IL-6 with different pH dependencies in the range pH 6.0–7.4. The behavior of antigen in the presence of these antibodies has been characterized using a combination of fixed and live cell fluorescence microscopy. As the affinity of the antibody:IL-6 interaction at pH 6.0 decreases, an increasing amount of antigen dissociates from FcRn-bound antibody in early and late endosomes, and then enters lysosomes. Segregation of antibody and FcRn from endosomes in tubulovesicular transport carriers (TCs) into the recycling pathway can also be observed in live cells, and the extent of IL-6 association with TCs correlates with increasing affinity of the antibody:IL-6 interaction at acidic pH. These analyses result in an understanding, in spatiotemporal terms, of the effect of pH dependence of antibody-antigen interactions on subcellular trafficking and inform the design of antibodies with optimized binding properties for antigen elimination. 相似文献
