Identification of new TRAP markers linked to chlorophyll content, leaf senescence, and cell membrane stability in water-stressed wheat 总被引:1,自引:0,他引:1

In order to identify target region amplification polymorphism (TRAP) markers linked to three physiological traits in wheat (Triticum aestivum L.), the segregating F4 population from the cross between drought-sensitive (Yecora Rojo) and drought-tolerant (Pavon 76) genotypes was made. The parents and 150 F4 families were evaluated phenotypically for drought tolerance using two irrigation treatments [2.5 and 7.5 m3(H2O) m?2(soil)]. Using 40 different TRAP primer combinations tested for polymorphism in parental and F4 family genotypes, the results revealed that quantitative trait locus (QTL) for chlorophyll content was associated with TRAP 5, TRAP 14, and TRAP 20 and explained 18, 16, and 23 % phenotypic variation, respectively. The genetic distance between chlorophyll content QTL and TRAP 5, TRAP 14, and TRAP 20 were 12.3, 19.8, and 13.6 cM, respectively. QTL for flag leaf senescence was associated with TRAP 2, TRAP 3, TRAP 15, and TRAP 16 and explained 33, 27, 28, and 23 % phenotypic variations, respectively. The genetic distance between flag leaf senescence QTL and TRAP 2, TRAP 3, TRAP 15, and TRAP 16 were 9.4, 14.7, 18.1, and 17.3 cM, respectively. QTL for cell membrane stability was associated with TRAP 8, TRAP 9, and TRAP 37 and explained 27, 30, and 24 % phenotypic variation, respectively. The markers TRAP 8, TRAP 9, and TRAP 37 had genetic distances of 17.0, 10.0, and 9.0 cM, respectively. Therefore, these TRAP markers can be used in breeding for drought tolerance in wheat. 相似文献

In Egypt, disposing of partially treated or untreated domestic and industrial wastewater into agricultural drains deteriorates their water quality. A growing interest in effective low-cost treatment of polluted water and wastewater has resulted in many studies on constructed wetlands.This study evaluates free water surface constructed wetlands (by far the largest application project is named “Lake Manzala Engineered Wetland [Egypt]”) utilized to improve the water quality in Bahr El Baqar drain, which is located at the northeastern edge of the Nile Delta. This drain discharges its water into Manzala Lake, which in turn has many fishing activities and is connected to the Mediterranean Sea. The full capacity of the constructed wetland system is 25,000 m3/day. Three various flow rate wetlands were investigated; five wetland beds of high flow rate of 0.344 m3/m2-day, five wetland beds of low flow rate of 0.048 m3/m2-day and reciprocated cells of flow of 500 m3/day.The concentrations of different contaminants along the constructed wetlands system were measured to determine the treatment efficiency. The effluent was compared with the Egyptian standards of water quality in agricultural drains (Law 48/1982). Due to the high percentage of the agricultural water drain, the concentrations of contaminants in the influent were relatively low. The percentages of removal for the different contaminants were BOD5: 52%, COD: 50%, TSS: 87%, TDS: 32%, NH4-N: 66%, PO4: 52%, Fe: 51%, Cu: 36%, Zn: 47% and Pb: 52%. The natural vegetation considerably increased the value of dissolved oxygen in the treated effluent. Eight reference genes of Brassica napus were evaluated using quantitative real-time polymerase chain reaction (qRT-PCR) data, focusing on vegetative tissues and developing embryos. Analyses of expression stability indicated that UP1, UBC9, UBC21, and TIP41 were the top four choices as stably expressed reference genes for vegetative tissues, whereas ACT7, UBC21, TIP41, and PP2A were the top four choices for maturing embryos. In addition, radiolabeling of overall messenger RNA (mRNA) of maturing embryos indicated that the expression patterns of the top four ranked reference genes reflected the overall mRNA content changes in maturing embryos. 相似文献

Aerobic metabolism requires a complex antioxidative system to balance reactive oxygen species (ROS). When in excess, ROS disrupt cellular activities and molecular structures. Owing to the variety of ROS, there are different antioxidative activities that require various tests for their detection. The so-called 'total antioxidative capacity' inhibition assay presented in this paper can be used to quantify the overall activity of low-molecular-weight antioxidants (AOs) in biological samples. The assay is based on enhanced horseradish peroxidase-catalyzed luminol chemiluminescence. It can be fine-tuned so that the biological samples meet the requirements of the light detector. A detailed protocol describing all relevant parameters is provided. The procedure is quick, inexpensive and reproducible. The assay can be used with diverse biological materials such as plant extracts and blood plasma. Hence, it is applicable to fields as diverse as crop breeding, medical diagnostics or food sciences. The time needed for the assay depends on the number of samples and their AO content. The protocol takes one working day to complete when five samples with five replicates are measured sequentially. 相似文献

A series of 2-fluorophenyl-4,6-disubstituted [1,3,5]triazines (1) and (2) were synthesized and evaluated for their antimicrobial activity against three representative gram-positive bacteria and two fungi. The structure–activity relationship (SAR) demonstrates that the 3- or 4-fluorophenyl component attached directly to the triazine ring was essential for activity. Of these compounds, 14, 15, and 25 demonstrated significant activity against all selected organisms compared to control. Effects of reversal coenzyme specificity toward NADP+ and thermostabilization of xylitol dehydrogenase (XDH) from Pichia stipitis on fermentation of xylose to ethanol were estimated using a recombinant Saccharomyces cerevisiae expressing together with a native xylose reductase from P. stipitis. The mutated XDHs performed the similar enzyme properties in S. cerevisiae cells, compared with those in vitro. The significant enhancement(s) was found in Y-ARSdR strain, in which NADP+-dependent XDH was expressed; 86% decrease of unfavorable xylitol excretion with 41% increased ethanol production, when compared with the reference strain expressing the wild-type XDH. 相似文献

Neurons in the anterior ventral (AV) thalamic nucleus of human adults were impregnated by Golgi-Kopsch impregnation method.
Results showed that at least three morphological types of neurons could be recognized in the human AV thalamic nucleus. Type
I neurons were medium to large with rich dendritic arborization. Both tufted and radiating dendritic branching patterns were
seen in almost every neuron of this type. Only the initial axonal segments of these cells were impregnated suggesting that
these axons were heavily myelinated. Type II neurons were medium in size with poor to moderate dendritic arborization. Many
of these cells possess a few dendritic grape-like appendages. Long segments (up to 300 μm) of their axons were impregnated
suggesting that these axons were either unmyelinated or thinly myelinated. These axons change their direction and form loops
very often. No local branches were seen for these axons suggesting that they could be projection axons. Type III neurons were
small with only one or two dendrites with poor arborization. No axons for these cells were seen in this study. The three neuronal
types in the human AV thalamic nucleus were compared with neuronal types already described in other thalamic nuclei of human
and non-human species. The results of this study might provide a morphological basis for further electrophysiological and

No past studies of acute diarrhea in Tunisia have examined the phenotypic and genotypic profiles of enterotoxigenic Escherichia coli (ETEC) isolates. We determined 65 ETEC isolates derived from a total of 327 E. coli isolates collected from a previous study (acute diarrheal and healthy persons, children and adults n = 214) and 32 E. coli isolates derived from an acute diarrheal outbreak in Kabaria-Ennour city, Tunis. All E. coli isolates were screened by polymerase chain reaction (PCR) for ETEC virulence genes: sta (heat-stable toxin gene) and elt (heat-labile toxin gene). Seventy-two percent (47 of 65) of ETEC strains expressed the sta gene only, 21.5% (14 of 65) expressed the elt gene and 6.1% (4 of 65) expressed both genes. For the outbreak isolates, the elt gene was predominant (10 isolates out of 14). Ganylioside GM1 enzyme-linked immunosorbent assay (GM1-ELISA) was used to validate the PCR results and this was confirmed by dot blot assay. The same results were obtained. The most common colonization factors (CFs) were CFA/I (44.6%) and coli surface antigen 6 (CS6) (11%), and 44.6% of the isolates showed no association with either CFAs. Resistance of ETEC isolates to tetracycline (38.5%), streptomycin (26%), and beta-lactam agents (ticarcillin 26%, amoxicillin 24.6%, cephalotin 21.5%) was common. Regarding serotypes, the majority of ETEC isolates serotyped as O86:H(-) (n = 16), O128:H2 (n = 11), and O127:H21 (n = 10). Other serotypes found were O111:H(-) (n = 6) and O126: H(-) (n = 5). DNA macrorestriction fragment analysis by pulsed-field gel electrophoresis (PFGE) using the XbaI enzyme was conducted to investigate the epidemiological clonal relationship among ETEC isolates. Major patterns were identified among which some of outbreak ETEC isolates belonged. These data suggest that a proportion of acute diarrhea in Tunis represents the confluence of small epidemics by clonality-related ETEC isolates that are transiently introduced or that persist in our community. 相似文献
