Studies of recombination in vertebrates have rather lagged behind those in yeast and bacteria in large part due to the relative genetic intractability of vertebrate model systems. Immunoglobulin diversification in the chicken cell line DT40 provides a powerful combination of a physiological recombination process coupled with facile genetic modification. The immunoglobulin variable regions of DT40 constitutively diversify by a combination of gene conversion, in which sequence changes are templated from one of a number of upstream pseudogenes or by non-templated point mutation. Both of these events are initiated by abasic sites in the variable region DNA generated following the targeted deamination of cytidine by activation induced deaminase. Recent work has shown that the two outcomes, gene conversion and somatic mutation, are likely to reflect alternate pathways for the processing of these abasic sites. In this review I will discuss the current data on avian Ig gene diversification and examine how the immunoglobulin loci of DT40 may provide a useful model system for studying the mechanisms and interactions of vertebrate recombination and pathways of DNA damage tolerance.  相似文献

The effect of temperature on post-tetanic potentiation (PTP) has been examined in the muscles of small mammals but not in human skeletal muscle. We examined PTP in the ankle dorsiflexor muscles of 10 young men by evoking twitches before and after a 7-second tetanus at 100 Hz in a control (room air approximately 21 degrees C) condition and after immersion of the lower leg in warm (45 degrees C) and cold (10 degrees C) water baths for 30 min. Exposure to cold decreased tetanus and pre-tetanus twitch peak torque, but increased rise time, half-relaxation time, and muscle action potential (M-wave) amplitude; exposure to warm water had little effect. PTP was smallest in cold exposure 5 s post-tetanus, but persisted throughout the 12 min test period, whereas PTP had subsided by 6 min post-tetanus in control and warm exposures. M-wave amplitude initially decreased after exposure to warm water, recovered, then decreased again by 11 min post-tetanus. In contrast, exposure to cold had no initial effect but did increase the M-wave amplitude during the last half of the 12 min test period, similar to that seen in the control. The greatest immediate decrease in rise time and half-relaxation time was observed in the control; however, by 12 min post-tetanus warm exposure showed the greatest increase in rise time and half-relaxation time above pre-tetanus values. The decrease in the unpotentiated twitch torque with cooling in human dorsiflexors is typical for muscles with a predominance of type I (slow) fibres. The effect of cold on PTP is similar to that seen previously in mammalian muscles with a predominance of type II (fast) fibres, although the underlying mechanism of the cooling effect appears to differ.  相似文献

There appears to be widespread acceptance that for a population to persist, some demographic parameter must be density dependent at some place or time. In this paper, we question the veracity and heuristic value of treating this statement as a general principle of ecology. We also point out that some processes that have recently been defined as density dependent are, in fact, not. Taken in its original sense, density dependence implies a change in demographic rates based on biological (generally negative) feedback. Situations exist, however, in which demographic rates change in relation to density without negative biological feedback. For example, per capita recruitment in marine populations will decrease as local population size increases even as absolute numbers of arriving larvae do not change. The failure to separate these density-related processes from true density-dependent processes affects our understanding of population regulation and of the way in which the natural world functions. Furthermore, focusing solely on density-dependent processes and their role in population regulation neglects to address numerous density-independent processes like disturbance and climatic variation that may have important impacts in determining population size. In small mammals, muscles with shorter twitch contraction times and a predominance of fast-twitch, type II fibers exhibit greater posttetanic twitch force potentiation than muscles with longer twitch contraction times and a predominance of slow-twitch, type I fibers. In humans, the correlation between potentiation and fiber-type distribution has not been found consistently. In the present study, postactivation potentiation (PAP) was induced in the knee extensors of 20 young men by a 10-s maximum voluntary isometric contraction (MVC). Maximal twitch contractions of the knee extensors were evoked before and after the MVC. A negative correlation (r = -0. 73, P < 0.001) was found between PAP and pre-MVC twitch time to peak torque (TPT). The four men with the highest (HPAP, 104 +/- 11%) and lowest (LPAP, 43 +/- 7%) PAP values (P < 0.0001) underwent needle biopsies of vastus lateralis. HPAP had a greater percentage of type II fibers (72 +/- 9 vs. 39 +/- 7%, P < 0.001) and shorter pre-MVC twitch TPT (61 +/- 12 vs. 86 +/- 7 ms, P < 0.05) than LPAP. These data indicate that, similar to the muscles of small mammals, human muscles with shorter twitch contraction times and a higher percentage of type II fibers exhibit greater PAP.  相似文献

The purpose of this study was to determine whether neural and/or muscular factors contributed to the inferior strength-related motor performances of obese adolescents. Subjects were 10 non-obese (14.6% fat) and 11 obese (32.3% fat) males matched for age (15-18 years), level of maturity (Tanner stages IV and V), lean body mass, and height. Peak torque (PT) was measured during maximal voluntary isometric (IS) and isokinetic (IK) knee extension (KE). Peak twitch torque (TT), time to peak torque (TPT), and half-relaxation time (HRT) of the knee extensors were elicited by percutaneous electrical stimulation. The interpolated twitch technique was used to determine the extent of motor unit activation (% MUA) during maximal voluntary IS KE. Knee extensor cross-sectional area (CSA) was determined by computed axial tomography taken at the mid-thigh. All strength and area measurements were made on the right side of the body. Obese subjects had significantly (P less than 0.05) lower maximal voluntary IS and IK KE strength normalized for body weight, and significantly lower % MUA during IS KE. There were no significant differences (P greater than 0.05) between groups for absolute or normalized (for the product of muscle CSA and height) ISPT, IKPT, and TT, knee extensor CSA, or TPT and HRT. These results suggest that reduced MUA and a lower strength per mass ratio (due to excess fat) are probably important contributing factors to the poorer motor performances of the obese, especially for complex motor tasks involving large muscle groups and the support or moving of body weight.  相似文献

Walking ability, though important for quality of life and participation in social and economic activities, can be adversely affected by neurological disorders, such as Spinal Cord Injury, Stroke, Multiple Sclerosis or Traumatic Brain Injury. The aim of this study is to evaluate if the energy cost of walking (CW), in a mixed group of chronic patients with neurological diseases almost 6 months after discharge from rehabilitation wards, can predict the walking performance and any walking restriction on community activities, as indicated by Walking Handicap Scale categories (WHS). One hundred and seven subjects were included in the study, 31 suffering from Stroke, 26 from Spinal Cord Injury and 50 from Multiple Sclerosis. The multivariable binary logistical regression analysis has produced a statistical model with good characteristics of fit and good predictability. This model generated a cut-off value of.40, which enabled us to classify correctly the cases with a percentage of 85.0%. Our research reveal that, in our subjects, CW is the only predictor of the walking performance of in the community, to be compared with the score of WHS. We have been also identifying a cut-off value of CW cost, which makes a distinction between those who can walk in the community and those who cannot do it. In particular, these values could be used to predict the ability to walk in the community when discharged from the rehabilitation units, and to adjust the rehabilitative treatment to improve the performance.  相似文献

Objective

The present study shows the results of a double-blind sham-controlled pilot trial to test whether measurable stimulus-specific functional connectivity changes exist after Automatic Mechanical Peripheral Stimulation (AMPS) in patients with idiopathic Parkinson Disease.

Methods

Eleven patients (6 women and 5 men) with idiopathic Parkinson Disease underwent brain fMRI immediately before and after sham or effective AMPS. Resting state Functional Connectivity (RSFC) was assessed using the seed-ROI based analysis. Seed ROIs were positioned on basal ganglia, on primary sensory-motor cortices, on the supplementary motor areas and on the cerebellum. Individual differences for pre- and post-effective AMPS and pre- and post-sham condition were obtained and first entered in respective one-sample t-test analyses, to evaluate the mean effect of condition.

Results

Effective AMPS, but not sham stimulation, induced increase of RSFC of the sensory motor cortex, nucleus striatum and cerebellum. Secondly, individual differences for both conditions were entered into paired group t-test analysis to rule out sub-threshold effects of sham stimulation, which showed stronger connectivity of the striatum nucleus with the right lateral occipital cortex and the cuneal cortex (max Z score 3.12) and with the right anterior temporal lobe (max Z score 3.42) and of the cerebellum with the right lateral occipital cortex and the right cerebellar cortex (max Z score 3.79).

Conclusions

Our results suggest that effective AMPS acutely increases RSFC of brain regions involved in visuo-spatial and sensory-motor integration.

Classification of Evidence

This study provides Class II evidence that automatic mechanical peripheral stimulation is effective in modulating brain functional connectivity of patients with Parkinson Disease at rest.

IntroductionMicrochimeric male fetal cells (MFCs) have been associated with systemic lupus erythematosus, and published studies have further correlated MFC with lupus nephritis (LN). In the present study, we evaluated the frequency of MFC in the renal tissue of patients with LN.MethodsTwenty-seven renal biopsies were evaluated: Fourteen were from women with clinical and laboratory findings of LN, and thirteen were from controls. Genomic DNA was extracted from kidney biopsies, and the male fetal DNA was quantified using real-time quantitative polymerase chain reactions for the detection of specific Y chromosome sequences.ResultsMFCs were detected in 9 (64%) of 14 of patients with LN, whereas no MFCs were found in the control group (P = 0.0006). No differences in pregnancy history were found between patients with LN and the control group. Significantly higher amounts of MFCs were found in patients with LN with serum creatinine ≤1.5 mg/dl. Furthermore, women with MFCs had significantly better renal function at the time of biopsy (P = 0.03). In contrast, patients with LN without MFCs presented with more severe forms of glomerulonephritis (World Health Organization class IV = 60% and class V = 40%).ConclusionsOur data indicate a high prevalence of MFCs in renal biopsy specimens from women with LN, suggesting a role for MFCs in the etiology of LN. The present report also provides some evidence that MFCs could have a beneficial effect in this disease.
