The effect of the thyroid activity on the formation of lipid peroxidation and on liver and heart antioxidant enzyme activities was investigated in Wistar rats. Hypothyroidism and hyperthyroidism conditions were induced for five weeks by the administration of 0.05% benzythiouracile (BTU) and L-thyroxine sodium salt (0.0012%), in drinking water, respectively. No significant effect was observed on the rates of both lipid peroxidation and the vitamin E in hepatic and cardiac tissues of hypothyroidism rats compared to the controls, contrary to the hyperthyroidism rats, which expressed a pronounced increase. The increased glutathione peroxidase activity in rats suffering from hyperthyroidism was associated with a fall of the reduced glutathione in the homogenate and a marked increase in the glutathione reductase activity. An increase in superoxide dismutase and catalase activities was also recorded in hyperthyroidism. Our results explain the thyroid activity variation in relation to the lipid peroxidation and the tissular contents of the enzymatic and the non-enzymatic antioxidants. To conclude, our results show the occurrence of a state of oxidizing stress in relation to hyperthyroidism.  相似文献

The WW-containing oxidoreductase (WWOX) tumor suppressor participates in a diverse array of cellular activities by virtue of its ability to recognize WW-binding protein 1 (WBP1) and WW-binding protein 2 (WBP2) signaling adaptors among a wide variety of other ligands. Herein, using a multitude of biophysical techniques, we provide evidence that while the WW1 domain of WWOX binds to PPXY motifs within WBP1 and WBP2 in a physiologically relevant manner, the WW2 domain exhibits no affinity toward any of these PPXY motifs. Importantly, our data suggest that while R25/W44 residues located within the binding pocket of a triple-stranded β-fold of WW1 domain are critical for the recognition of PPXY ligands, they are replaced by the chemically distinct E66/Y85 duo at structurally equivalent positions within the WW2 domain, thereby accounting for its failure to bind PPXY ligands. Predictably, not only does the introduction of E66R/Y85W double substitution within the WW2 domain result in gain of function but the resulting engineered domain, hereinafter referred to as WW2_RW, also appears to be a much stronger binding partner of WBP1 and WBP2 than the wild-type WW1 domain. We also show that while the WW1 domain is structurally disordered and folds upon ligand binding, the WW2 domain not only adopts a fully structured conformation but also aids stabilization and ligand binding to WW1 domain. This salient observation implies that the WW2 domain likely serves as a chaperone to augment the physiological function of WW1 domain within WWOX. Collectively, our study lays the groundwork for understanding the molecular basis of a key protein-protein interaction pertinent to human health and disease.  相似文献

The dinuclear terephthalato-bridged nickel(II) complexes [Ni2(cyclen)2(μ-tp)](ClO4)2 (1) [Ni2(trpn)2(μ-tp)(H2O)2](ClO4)2 (2) and [Ni2(3,3,3-tet)2(μ-tp)(H2O)2](ClO4)2 · 2H2O (3), where tp = terephthalate dianion, cyclen = 1,4,7,10-tetraazacyclododecane, trpn = tris(3-aminopropyl)amine and 3,3,3-tet = 1,5,9,13-tetraazatridecane, were synthesized and structurally characterized by X-ray crystallography. Their magnetic susceptibilities were also determined at variable temperatures over the range 2-300 K. The structures of these complexes consist of μ-tp bridging two Ni(II) centers in a bis(bidentate) bonding fashion in 1 and in bis(monodentate) bonding fashion in 2 and 3. The coordination geometry around the Ni(II) ions in these compounds has a distorted octahedral geometry with four nitrogen atoms from the amine ligand (cyclen, trpn or 3,3,3-tet) and two coordinated oxygen atoms supplied by the chelated carboxylate group of the bridged terephthalate ligand in 1, and by one tp-carboxylate-oxygen in 2 and 3. The sixth coordination site in the last two complexes 2 and 3 is achieved via an oxygen atom from a coordinated water molecule. The intradimer Ni…Ni distances in these complexes are 10.740, 11.428 and 11.537 Å for 1, 2 and 3, respectively. The electronic spectra of the complexes in aqueous solutions are in complete agreement with the assigned X-ray geometry around the Ni(II) centers. Also, the analysis of the infrared spectral data for the ν(COO) stretching frequencies of the tp-carboxalato groups reveals the existence of the bis(bidentate) and bis(monodentate) coordination modes for the bridged terephthalate ligand in 1, 2 and 3, respectively. Despite the different coordination modes of the tp bridging ligand in these complexes, they all exhibit very weak antiferromagnetic coupling. The coupling constants J were found to be −2.2, −0.6 and −1.5 cm3 K mol−1 for the complexes 1, 2 and 3, respectively. The structural and magnetic results of 1-3 are discussed in relation to the other related published μ-terephthalato dinuclear Ni(II) compounds.  相似文献

In vitro studies on the pathogenesis in swine have been hampered by the lack of relevant porcine cell lines. Since many bacterial infections are swine-specific, studies on pathogenic mechanisms require appropriate cell lines of porcine origin. We have characterized the permanent porcine intestinal epithelial cell line, IPEC-J2, using a variety of methods in order to assess the usefulness of this cell line as an in vitro infection model. Electron microscopic analyses and histochemical staining revealed the cells to be enterocyte-like with microvilli, tight junctions and glycocalyx-bound mucin. The functional integrity of monolayers was determined by transepithelial electrical resistance (TEER) measurements. Both commensal bacteria and important bacterial pathogens were chosen for study based on their principally different infection mechanisms: obligate extracellular Escherichia coli, facultative intracellular Salmonella and obligate intracellular Chlamydia. We determined the colonization and proliferation of the bacteria on and within the host cells and monitored the host cell response. We verified the expression of mRNAs encoding the cytokines IL-1α, −6, −7, −8, −18, TNF-α and GM-CSF, but not TGF-β or MCP-1. IL-8 protein expression was enhanced by Salmonella invasion. We conclude that the IPEC-J2 cell line provides a relevant in vitro model system for porcine intestinal pathogen–host cell interactions.  相似文献
