This study is designed to investigate whether substrate preference in the myocardium during the neonatal period and hypoxia-induced stress is controlled intracellularly or by extracellular substrate availability. To determine this, the effect of exogenous L-carnitine on the regulation of carbohydrate and fatty acid metabolism was determined during cardiac stress (hypoxia) and during the postnatal period. The effect of L-carnitine on long chain (palmitate) and medium chain (octonoate) fatty acid oxidation was studied in cardiac myocytes isolated from less than 24 h old (new born; NB), 2 week old (2 week) and hypoxic 4 week old (HY) piglets. Palmitate oxidation was severely decreased in NB cells compared to those from 2 week animals (0.456 ± 0.04 vs. 1.207 ± 0.52 nmol/mg protein/30 min); surprisingly, cells from even older hypoxic animals appeared shifted toward the new born state (0.695 ± 0.038 nmol/mg protein/30 min). Addition of L-carnitine to the incubation medium, which stimulates carnitine palmitoyl-transferase I (CPTI) accelerated palmitate oxidation 3 fold in NB and approximately 2 fold in HY and 2 week cells. In contrast, octanoate oxidation which was greater in new born myocytes than in 2 week cells, was decreased by L-carnitine suggesting a compensatory response. Furthermore, oxidation of carbohydrates (glucose, pyruvate, and lactate) was greatly increased in new born myocytes compared to 2 week and HY cells and was accompanied by a parallel increase in pyruvate dehydrogenase (PDH) activity. The concentration of malonyl-CoA, a potent inhibitor of CPTI was significantly higher in new born heart than at 2 weeks. These metabolic data taken together suggest that intracellular metabolic signals interact to shift from carbohydrate to fatty acid utilization during development of the myocardium. The decreased oxidation of palmitate in NB hearts probably reflects decreased intracellular L-carnitine and increased malonyl-CoA concentrations. Interestingly, these data further suggest that the cells remain compliant so that under stressful conditions, such as hypoxia, they can revert toward the neonatal state of increased glucose utilization.  相似文献

Copper (Cu) chaperones constitute a family of small Cu+-binding proteins required for Cu homeostasis in eukaryotes. The ATX1 family of Cu chaperones specifically delivers Cu to heavy metal P-type ATPases. The plant Arabidopsis thaliana expresses the ATX1-like Cu chaperone CCH, which exhibits a plant-specific carboxy-terminal domain (CTD) with unique structural properties. We show that CCH homologues from other higher plants contain CTDs with structural properties similar to Arabidopsis CCH. Furthermore, we identify a new ATX1-like Cu chaperone in Arabidopsis, AtATX1, which functionally complements yeast atx1Delta and sod1Delta associated phenotypes, and localizes to the cytosol of Arabidopsis cells. Interestingly, AtATX1, but not full-length CCH, interacts in vivo with the Arabidopsis RAN1 Cu-transporting P-type ATPase by yeast two-hybrid. We propose that higher plants express two types of ATX1-like Cu chaperones: the ATX1-type with a predominant function in Cu delivery to P-type ATPases, and the CCH-type with additional CTD-mediated plant-specific functions.  相似文献

The upper Ordovician succession of Jordan was located 60°S, less than 100 km from the Hirnantian ice sheet margin. New graptolite dates indicate glaciation ended in Jordan in the late Hirnantian (persculptus Biozone). The succession records two glacial advances within the Ammar Formation and the subsequent deglaciations. Organic-rich black shales (Batra Formation) form part of the final deglacial transgressive succession that in-filled an existing low stand glacial continental shelf topography. The base of the black shale is coincident with the maximum flooding surface. During transgression, interfluves and sub-basin margins were breached and black shale deposition expanded rapidly across the region. The top of the black shales coincides with peak highstand. The “expanding puddle model” (sensu Wignall) for black shale deposition, adapted for the peri-glacial setting, provides the best explanation for this sequence of events.

We propose a hypothesis in which anoxic conditions were initiated beneath the halocline in a salinity stratified water column; a fresher surface layer resulted from ice meltwater generated during early deglaciation. During the initial stages of marine incursion, nutrients in the monimolimnion were isolated from the euphotic zone by the halocline. Increasing total organic carbon (TOC) and δ13Corg up section indicates the organic carbon content of the shales was controlled mainly by increasing bioproductivity in the mixolimnion (the Strakhov model). Mixolimnion nutrient levels were sustained by a continual and increasing supply of meltwater-derived nutrients, modulated by obliquity changes in high latitude insolation. Anoxia was sustained over tens to hundreds of thousands of years. The formation of black shales on the north Gondwana shelf was little different to those observed in modern black shale environments, suggesting that it was the nature of the Ordovician seas that pre-disposed them to anoxia.  相似文献

Since its opening 5.33 million years ago, the Gibraltar Strait has always contributed to the Mediterranean fauna and flora. Despite the increasing importance of the phenomenon, ecological determinants underlying colonization success of Atlantic fishes in the Mediterranean Sea have been poorly investigated. Here we reconstruct the recent historical colonization of the whole Mediterranean Sea by Atlantic fishes and we aim to determine where Atlantic fishes preferentially establish and whether some biological traits and ecological factors can be correlated to the colonization success (climate match, position in the water column, maximum body length, propagules, confamilial resistance, depth). A database on Atlantic fish species records from 1810 to 2006 was built and the colonization rate of each introduced species was estimated. Analysis of Variance, Chi squared test and logistic regression were used to investigate the relationships between ecological variables and colonization success. In addition, an index of asymmetry was used to analyse the relative colonization on the two sides of the Mediterranean Sea. Overall 48.33% of Atlantic species introduced in the Mediterranean Sea succeeded in colonizing eastwards. We found that habitat depth of Atlantic species is significantly related to their colonization success due to the obstacle of the shallow depth of the Gibraltar Strait (300 m). It also appears that despite the cyclonic general water circulation on the North Western basin, the northern side is more colonized than the southern one: 70.40% of the studied species colonize the northern side, while only 29.62% colonize the southern one. Two hypotheses may explain this trend: the bottom-up process that enhances the colonization success of Atlantic fishes along the Spanish coast of Alboran Sea owning to its high productivity and the intensiveness of the scientific explorations along the northern Mediterranean side. We conclude that crossing the Gibraltar Strait does not guarantee the colonization success and that species life-history and functional traits are poor predictors. Instead we suggest that environmental factors may determine favourable locations for invasive species installation. Escherichia coli ribosomal protein S1 is required for the translation initiation of messenger RNAs, in particular when their Shine–Dalgarno sequence is degenerated. Closely related forms of the protein, composed of the same number of domains (six), are found in all Gram-negative bacteria. More distant proteins, generally formed of fewer domains, have been identified, by sequence similarities, in Gram-positive bacteria and are also termed ‘S1 proteins’. However in the absence of functional information, it is generally difficult to ascertain their relationship with Gram-negative S1. In this article, we report the solution structure of the fourth and sixth domains of the E. coli protein S1 and show that it is possible to characterize their β-barrel by a consensus sequence that allows a precise identification of all domains in Gram-negative and Gram-positive S1 proteins. In addition, we show that it is possible to discriminate between five domain types corresponding to the domains 1, 2, 3, 4–5 and 6 of E. coli S1 on the basis of their sequence. This enabled us to identify the nature of the domains present in Gram-positive proteins and, subsequently, to probe the filiations between all forms of S1.  相似文献

The genetic characterization of natural populations of common pandora (Pagellus erythrinus, Sparidae) was studied using allozyme markers. Fish samples (n = 360) were collected from five locations along the Tunisian coasts and one from the Libyan coast. From eight enzymatic systems, 12 putative enzyme-coding loci were resolved, 8 of which were polymorphic at a level of 99%. F statistics indices revealed a significant departure from panmixia with heterozygote deficiencies and a slight genetic differentiation between samples. Neighbor-joining dendrogram constructed by Reynolds genetic distance showed that the populations sampled from the southeastern Mediterranean Sea were more differentiated from the others.  相似文献
