Abstract The present study was designed to establish the susceptibility of macrophage-mediated effector functions to tetanus toxin (TT). Using the murine macrophage cell line, GG2EE, generated in vitro by v- raf /v- myc oncogenes, we have previously provided evidence that TT selectively inhibits interferon gamma (IFN-γ), but not basal, lysozyme activity. Here we show that while neither phagocytic nor candidacidal activities are affected by TT treatment, antitumoral activity is significantly impaired after exposure to TT. This phenomenon, which is dose-dependent, is fully ascribed to the holotoxin, as heat inactivated TT, C or A-B fragments result ineffective. Furthermore, C but not A-B fragment competes with TT in abrogating its inhibitory effects. Overall, these data indicate that TT is not a broad-spectrum, down-regulating signal on macrophage-mediated functions, thus implying that its toxic action is exerted on specific molecular targets. 相似文献
Low-temperature and high humidity are typical environmental factors in the plastic tunnel and solar greenhouse during the cold season that restricts plant growth and development. Herein, we investigated the impact of different combinations of low-temperature and high humidity (day/night: T1 15/10 °C?+?95%, T2 12/8 °C?+?95%, and T3 9/5 °C?+?95%) along with a control (CK 25/18 °C?+?80%) on cucumber cultivars viz: Zhongnong37 (ZN37: resistant) and Shuyanbailv (SYB: sensitive). The low-temperature and high humidity stresses increased electrolyte leakage (EL), malondialdehyde (MDA), hydrogen peroxide (H2O2) and intercellular concentration of carbon dioxide (Ci), and reduced morphological indices, relative water content (RWC), net photosynthesis rate (Pn), stomatal conductance (Gs), transpiration rate (E) and leaf pigments in both cultivars as compared to control (CK). Superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), dehydroascorbate reductase (DHAR), and glutathione reductase (GR) were decreased in cv. SYB under stress conditions as compared to cv. ZN37. Low-temperature and high humidity treatments showed an increase in proline and soluble protein content in cv. ZN37 as compared to cv. SYB. Abscisic acid (ABA) and jasmonic acid (JA) were augmented while indole-3-acetic acid (IAA), zeatin (ZT), zeatin riboside (ZR), and gibberellic acid (GA) were decreased in both cultivars. Under T3 (9/5 °C?+?95%), Pn, protoporphyrin, and ZT were extremely decreased by 71.3%, 74.3%, and 82.4%, respectively, in cv. SYB compared to control. Moreover, principal component analysis (PCA) based on physiochemical traits confirmed that cv. ZN37 had the strongest correlation with antioxidant enzymes, proline, and soluble protein content than cv. SYB under low-temperature and high humidity treatments. Our results suggest that a stress-tolerant cultivar mitigates stress damage in cucumber transplants by regulating photosynthetic efficiency, antioxidant capacity and hormonal profile when compared to a stress-sensitive cultivar.
Many plants have been known to be contaminated and accumulate plasticizers from the environment, including water sources, soil, and atmosphere. Plasticizers are used to confer elasticity and flexibility to various fiber and plastic products. Consumption of plasticizers can lead to many adverse effects on human health, including reproductive and developmental toxicity, endocrine disruption, and cancer. Herein, we report for the first time that two plasticizers, bis(2-ethylhexyl) terephthalate (DEHT) and bis(2-ethylhexyl) phthalate (DEHP), have been isolated from the leaves of Capparis spinosa L. (the caper bush), a plant that is widely used in food seasonings and traditional medicine. 297 mg/kg of DEHT and 48 mg/kg of DEHP were isolated from dried and grounded C. spinosa L. leaves using column chromatography and semi-preparative high-performance liquid chromatography. Our study adds to the increase in the detection of plasticizers in our food and medicinal plants and to the alarming concern about their potential adverse effects on human health. 相似文献
The Ventilago Gaertn. (Rhamnaceae) is widely distributed in pantropical areas of Africa, Asia, and Australia. However, fossil records of this taxon are sparse, which limits understanding of the evolution and biogeographic history of the genus. In the present study, we report and describe two new fossil species of Ventilago, V. siwalika sp. nov. from the Miocene sediments of Himachal Pradesh, western Himalaya, and V. pliocenica sp. nov. from the Pliocene sediments of Jharkhand, eastern India based on single-winged samaras. Ventilago pliocenica is characterized by a prominent midvein, obtuse to sub-round apex with mucronate tip, longitudinal secondary veins extending the full length of the fruit, and reticulate nature of higher-order veins, the presence of equatorial rim, the hypanthium, and short pedicel. On the other hand, V. siwalika is characterized by a prominent midvein, obtuse to sub-round apex with mucronate tip, longitudinal secondary veins extending the full length of the fruit, and reticulate nature of higher-order veins. Our discovery represents the first unambiguous fossil record of single-winged samara of Ventilago from India and provides valuable insights into the evolution of this genus. In this paper, we also review its biogeographic history and add new information to understand its hypothetical migration route. Present and earlier records of Ventilago also suggest that this genus was a common forest element during Neogene (Miocene time) in Asia. 相似文献
In the bivalve mollusc Pecten maximus, the size of the mitochondrial DNA molecules ranges from 20 to 25.8 kbp. This variability is mainly correlated with the occurrence of a variable domain composed with two to five 1.6-kbp repeated units tandemly arrayed in the genome. DNA fragments spanning the 1,586-base-pair-long repeated element and the nearest flanking gene have been cloned and sequenced. This sequence was analyzed regarding its base composition and potential secondary structures. The repeated unit domain was positioned and oriented with regard to the known flanking gene. It ends 2 base pairs upstream relative to the beginning of the tRNAgly gene. The peculiar properties of the repeated unit were compared with those of the 1,442-bp repeated element found in the mitochondrial genome of the deep sea scallop Placopecten magellanicus. This comparison provided evidence for the absence of nucleotide conservation, except for a small sequence engaged in a secondary structure, but argued for a strong pressure maintaining domains with specific nucleotide content. Ifrecovery from exercise could be enhanced in individuals with chroniclung disease, subsequent supramaximal exercise performance could also be improved. Recovery from supramaximal exercise and subsequent supramaximal exercise performance were assessed after 10 min of breathing 100% O2 or room air(RA) in 17 cystic fibrosis (CF) patients [25 ± 10 (SD) yrold, 53% men, forced expired volume in 1 s = 62 ± 21%predicted] and 17 normal subjects (25 ± 8 yr old, 59% men,forced expired volume in 1 s = 112 ± 15% predicted). Supramaximalperformance was assessed as the work of sustained bicycling at a loadof 130% of the maximum load achieved during a graded maximal exercise.Peak minute ventilation(E) andheart rate (HR) were lower in CF patients at the end of eachsupramaximal bout than in controls. In CF patients, single-exponentialtime decay constants indicated faster recovery of HR(HR = 86 ± 8 and 73 ± 6 s in RA and O2,respectively, P < 0.01). Similarly, fast and slow time constants of two-exponential equations providing thebest fit for ventilatory recovery were improved in CF patients duringO2 breathing ( = 132.1 ± 10.5 vs. 82.5 ± 10.4 s; = 880.3 ± 300.1 vs. 368.6 ± 107.1 s,P < 0.01). However, no such improvements occurred in controls. Supramaximal performance after O2 improved in CF patients (109 ± 6% of the 1st bout after O2 vs. 94 ± 6% in RA, P < 0.01).O2 supplementation had no effect on subsequent performance in controls (97 ± 3% inO2 vs. 93 ± 3% in RA). Weconclude that supplemental O2after a short bout of supramaximal exercise accelerates recovery andpreserves subsequent supramaximal performance in patients with CF. 相似文献
