A partially purified protein (the SR fraction) of porcine and human origin has been extensively characterized as Follicular Regulatory Protein (FRP). In the current study, 1A8D5, one of several monoclonal antibodies raised against FRP, was used to further purify the protein. The monoclonal antibody cross-reacted only with porcine plasminogen, a key fibrinolytic proenzyme. A commercial polyclonal antibody for human plasminogen confirmed the relationship between plasminogen and bands of the SR fraction of the porcine follicular fluid. Sequencing of the N-terminal amino acids (54 kd) of the SR fraction indicated that it shared 100% identity with the short form of porcine plasminogen chain A and 93% identity to human plasminogen. Moreover, we demonstrated that this purified protein from human follicular fluid inhibited aromatase activity of granulosa cells, a key biological property of FRP. Given that plasminogen possesses most of the proposed properties of the protein termed FRP, we conclude that FRP is likely plasminogen itself or a plasminogen-related protein and not a novel protein. 相似文献

Recent evidence has suggested that apoptosis plays an important role in various diseases, but concerns about the specificity of the TUNEL method for detecting apoptosis have been raised. The purpose of the present study was to investigate the specificity of the TUNEL method by using immersion and perfusion fixed tissues from both normal rats and rats with heart failure. Although a few positive cells were observed in perfusion fixed tissues, a significant number of positive cells were observed in immersion fixed tissues, especially when fixed tissues were kept for an extended time before the TUNEL assay was applied. The results of TUNEL staining should be interpreted with caution. When immersion fixation is used, fixed tissues should be assayed by the TUNEL method before the DNA degenerates. 相似文献

Pyrroles and its fused forms possess antimicrobial activities, they can easily interact with biomolecules of living systems. A series of substituted pyrroles, and its fused pyrimidines and triazines forms have been synthesised, all newly synthesised compound structures were confirmed by spectroscopic analysis. Generally, the compounds inhibited growth of some important human pathogens, the best effect was given by: 2a, 3c, 4d on Gram-positive bacteria and was higher on yeast (C. albicans), by 5c on Gram-negative bacteria and by 5a then 3c on filamentous fungi (A. fumigatus and F. oxysporum). Such results present good antibacterial and antifungal potential candidates to help overcome the global problem of antibiotic resistance and opportunistic infections outbreak. Compound 3c gave the best anti-phytopathogenic effect at a 50-fold lower concentration than Kocide 2000, introducing a safe commercial candidate for agricultural use. Little is known on the diversity and public health significance of Echinococcus species in livestock in Egypt. In this study, 37 individual hydatid cysts were collected from dromedary camels (n=28), sheep (n=7) and buffalos (n=2). DNA was extracted from protoscoleces/germinal layer of individual cysts and amplified by PCR targeting nuclear (actin II) and mitochondrial (COX1 and NAD1) genes. Direct sequencing of amplicons indicated the presence of Echinococcus canadenesis (G6 genotype) in 26 of 28 camel cysts, 3 of 7 sheep cysts and the 2 buffalo derived cysts. In contrast, Echinococcus granulosus sensu stricto (G1 genotype) was detected in one cyst from a camel and 4 of 7 cysts from sheep, whereas Echinococcus ortleppi (G5 genotype) was detected in one cyst from a camel. To improve the ultraviolet (UV) water disinfection process and to better understand the impact of the harmful effects of germicidal radiation on the DNA molecule, we have developed a reliable biological monitoring system based on PCR 16S ribosomal DNA (rDNA) and terminal restriction fragment length polymorphism analysis. The PCR analysis was performed using the bacteria-specific 27F and 905R primers to replicate a fragment of the rDNA gene. This new and versatile method can be used to evaluate the effects of direct UV radiation on DNA (UV dose/response) and to estimate the potential of bacteria to mitigate UV lethal effects via photoreactivation and dark repair. 相似文献

In order to establish an updated checklist of terrestrial gastropod from Tunisia, a revision of the species of Sphincterochilidae is presented, using bibliographic and museum records and the results of our own field work. As a result, only two species, Sphincterochila candidissima and Sphincterochila tunetana, are accepted to occur in Tunisia, and their type specimens are illustrated. The study of the morphological characters of the genital organs of both species clarified their subgeneric affiliation. Comparison of Sphincterochila tunetana with Sphincterochila cariosa from Lebanonshowed that the first has to be classified within the subgenus Albea, and the latter within Sphincterochila s. str.; the subgenus Rima Pallary, 1910 remains in the synonymy of Sphincterochila s. str. Bibliographic records of Sphincterochila baetica and Sphincterochila otthiana from Tunisia could not be confirmed, the latter probably lives close to the border with Algeria. 相似文献
