The role of bacterial 1-aminocyclopropane-1-carboxylate (ACC) deaminase activity in the interaction between tomato (Lycopersicon esculentum=Solanum lycopersicum) and Pseudomonas brassicacearum was studied in different strains. The phytopathogenic strain 520-1 possesses ACC deaminase activity, an important trait of plant growth-promoting rhizobacteria (PGPR) that stimulates root growth. The ACC-utilizing PGPR strain Am3 increased in vitro root elongation and root biomass of soil-grown tomato cv. Ailsa Craig at low bacterial concentrations (10(6) cells ml-1 in vitro and 10(6) cells g-1 soil) but had negative effects on in vitro root elongation at higher bacterial concentrations. A mutant strain of Am3 (designated T8-1) that was engineered to be ACC deaminase deficient failed to promote tomato root growth in vitro and in soil. Although strains T8-1 and 520-1 inhibited root growth in vitro at higher bacterial concentrations (>10(6) cells ml-1), they did not cause disease symptoms in vitro after seed inoculation, or in soil supplemented with bacteria. All the P. brassicacearum strains studied caused pith necrosis when stems or fruits were inoculated with a bacterial suspension, as did the causal organism of this disease (P. corrugata 176), but the non-pathogenic strain Pseudomonas sp. Dp2 did not. Strains Am3 and T8-1 were marked with antibiotic resistance and fluorescence to show that bacteria introduced to the nutrient solution or on seeds in vitro, or in soil were capable of colonizing the root surface, but were not detected inside root tissues. Both strains showed similar colonization ability either on root surfaces or in wounded stems. The results suggest that bacterial ACC deaminase of P. brassicacearum Am3 can promote growth in tomato by masking the phytopathogenic properties of this bacterium.  相似文献

The encyrtid genus Comperiella Howard has so far not been reported in the Philippines, where there is currently an outbreak of the coconut scale insect Aspidiotus rigidus Reyne particularly in the southern parts of the island of Luzon and in some areas in Mindanao. Among Comperiella species, only C. unifasciata Ishii has been reported as a parasitoid of A. rigidus. We report not only new sightings of this parasitoid genus in the Philippines from surveys conducted in parts of the provinces of Laguna and Batangas, but also the discovery of a possibly new species that, like C. unifasciata, has been found to parasitize A. rigidus at a high rate. These findings have presented a potential of biological control against the coconut scale insect problem that has threatened the coconut industry in the country.  相似文献

It is known that the Rhizobium galegae genomes contain megaplasmids. The suicide vector pSUP2111 with nifH gene of R. meliloti was introduced into the strains CIAM 0703 and CIAM 0711 of R. galegae inducing effective nodules on Galega orientalis plants. The formation of self-transmissible megaplasmids was observed. The megaplasmid transfer into non-nodulating R. meliloti mutants resulted in partial complementation of the nodulation defect in recipient strains though only one transconjugant showed the nitrogen-fixing activity in symbiosis with alfalfa and another one in symbiosis with G. orientalis plants. Among the Agrobacterium strains harbouring R. galegae megaplasmids there were four classes of transconjugants: (1) Nod+ Fix- in symbiosis with goat's rue plants (three strains); (2) Nod+ Fix- on Medicago sativa (two strains); (3) Nod+ Fix+ on M. sativa (five strains); (4) Nod- with both plant hosts (11 strains).  相似文献

The results of genetic analysis of the effects of four novel partial mouse TM-haplotypes are presented in this work. Fertility and viability of tM/tM homozygotes and tM/t6 compounds, transmission ratio distortion (trd) in males heterozygous for tM, suppression of recombination and taillessness effects were studied. Three novel t-haplotypes tM1,2,4 are viable and heterozygous for these haplotypes males T/tM show low trd (20-30%). Comparison of these data and the t6-haplotype structure suggests that the tM1,2,4-haplotypes were derived as a result of recombination events in the non-inverted T-complex fragment located between two inversions. The tM3-haplotype is semilethal and heterozygous T/tM3 males show the trd equal to that of t6-heterozygotes. Homozygous tM3/tM3 and tM3/t6 male compounds are fertile or subfertile. Potential recombination ways of derivation of tM3 are discussed.  相似文献

The results of light and electron microscopic (EM) studies of meiosis in Microtus arvalis males of the karyoform "arvalis" (2n = 46, NFa = 80), in hybrids between the chromosomal forms arvalis and obscurus (2n = 46, NFa = 68), in M. rossiaemeridionalis voles (2n = 54, NFa = 54), and in a hybrid between the species M. rossiaemeridionalis and M. kermanensis (2n = 54, NFa = 54) are presented. SC (synaptonemal complex) karyotypes of the parental forms and the hybrids were constructed on the basis of measurements of the length ofautosomal SCs revealed by the EM analysis in spermatocytes at the stage of middle pachytene. The SC karyotypes of M. arvalis and the hybrids female obscurus x male arvalis consist of 22 synaptonemal complexes of autosomal bivalents and the axial elements of the synaptonemal complexes of the sex chromosomes X and Y. The SC karyotypes of M. rossiaemeridionalis and the hybrid M. rossiaemeridionalis x M. kermanensis consist of 26 synaptonemal complexes of autosomal bivalents and a sex bivalent; they differ only in the length of the Y chromosome axis (Y chromosome in the hybrid was inherited from M. kermanensis). Asynaptic configurations of the autosomal SCs were not observed in the hybrids. The SC axial elements of the X and Y chromosomes in the parental forms and in the hybrids were located close to each other throughout pachytene, but they did not form a synaptic region. The normal synapsis in sterile hybrids (M. rossiaemeridionalis x M. kermanensis) and the behavior of the sex chromosomes in meiosis in fertile and sterile hybrids are discussed in the context of specific features of meiosis and reproductive isolation.  相似文献

During the early stages of conditioned defensive reflex formation, a dramatic increase in the acidic protein concentration can be seen in the snail CNS (the mobility 0.58 in polyacrylamide gel) with reference to water-soluble proteins of the animals' ganglions under study. Such an increase occurs at the time when the majority of neurons are involved in the learning process. The rise of the protein content is also seen upon the snail arousal, 2-6 h after being awake. It is suggested that the high level of the protein correlates with the appearance of the orientation reflexes. At the cellular level this phenomenon appears to be linked with increased excitability of a great number of neurons.  相似文献
