BACKGROUNDS: Until today, human stomach is the only recognized habitat of Helicobacter pylori. However, recruitment of DNA-based methods has made possible the detection of H. pylori in water and oral cavity, thus suggesting fecal-oral and oral-oral routes for transmission of H. pylori, respectively. In this study, yeast has been proposed as a common vector for transmission of H. pylori. Thus designed primers were recruited to target 16S rDNA and cagA genes in the oral yeasts by PCR. MATERIALS AND METHODS: Eighteen yeasts were examined microscopically for the presence of bacterial-like bodies. DNAs were extracted from oral yeasts using phenol-chloroform method. Amplification conditions were optimized as 33 cycles and annealing temperatures of 63 degrees C for 16S rDNA and 51 degrees C and 52 degrees C for cagA gene which was targeted in two steps. DNAs of H. pylori and Saccharomyces cerevisiae were used as controls. Polymerase chain reaction (PCR) products of two genes from one yeast and from H. pylori were cloned in pCAP and subsequently subcloned in pSK+ and were sequenced. RESULTS: Bacterial-like bodies were observed in all oral yeasts. The amplified products of 16S rDNA from all oral yeasts were homologous in size with those of H. pylori. Fifteen out of eighteen (83%) yeasts contained cagA gene, homologous to H. pylori. CagA was not amplified from three yeasts and S. cerevisiae. Analysis of the sequenced products of 16S rDNA and cagA from one oral yeast showed 98% homology with those of H. pylori. CONCLUSIONS: The presence of H. pylori inside the yeast was indicated by light microscopy and PCR. It appears that yeasts, which are abundant in nature and thrive the mucosal surfaces of human, might serve as reservoirs and vehicles of H. pylori.  相似文献

The six non-interacting modes for stress, strain and energy in an orthotropic elastic model of human femoral cortical bone tissue are discussed and illustrated. The stress and strain modes are illustrated using the representation of the stress and strain fields around a circular hole in a flat plate of cortical bone subjected to a uniaxial field of tension as the example. The six modes play a role in the stress analysis of orthotropic elastic materials similar to the roles played by the hydrostatic and deviatoric non-interacting stress, strain and energy modes in isotropic elasticity. The biomechanical significance of the six non-interacting modes for stress, strain and energy in hard tissue is both practical and suggestive. The modes suggest a practical scheme for the representation of stress and strain fields in hard tissue. The existence of the modes suggests physical insights, for example, possible failure mechanisms or adaptation strategies possessed by the hard tissues.  相似文献

Purpose

Variability in consumer behaviour can significantly influence the environmental performance of products and their associated impacts and this is typically not quantified in life cycle assessments. The goal of this paper is to demonstrate how consumer behaviour data can be used to understand and quantify the variability in the greenhouse gas emissions from domestic laundry washing across Europe.

Methods

Data from a pan-European consumer survey of product usage and washing habits was combined with internal company data on product format greenhouse gas (GHG) footprints and in-home measurement of energy consumption of laundry washing as well as literature data to determine the GHG footprint of laundry washing. The variability associated with four laundry detergent product formats and four wash temperature settings in washing machines were quantified on a per wash cycle basis across 23 European countries. The variability in GHG emissions associated with country electricity grid mixes was also taken into account. Monte Carlo methods were used to convert the variability in the input parameters into variability of the life cycle GHG emissions. Rank correlation analysis was used to quantify the importance of the different sources of variability.

Results and discussion

Both inter-country differences in background electricity mix as well as intra-country variation in consumer behaviour are important for determining the variability in life cycle GHG emissions of laundry detergents. The average GHG emissions related to the laundry washing process in the 23 European countries in 2014 was estimated to be 5?×?102 g CO2?eq/wash cycle, but varied by a factor of 6.5 between countries. Intra-country variability is between a factor of 3.5 and 5.0 (90% interval). For countries with a mainly fossil-based electricity system, the dominant source of variability in GHG emissions results from consumer choices in the use of washing machines. For countries with a relatively low-carbon electricity mix, variability in life cycle GHG emissions is mainly determined by laundry product-related parameters.

Conclusions

The combination of rich data sources enabled the quantification of the variability in the life cycle GHG emissions of laundry washing which is driven by a variety of consumer choices, manufacturer choices and infrastructural differences of countries. The improved understanding of the variability needs to be balanced against the cost and challenges of assessing of consumer habits.

Fibrin-platelet glue (FPG) is a blood derivative, in which platelets and fibrinogen are concentrated in a small plasma volume, by differential centrifugation and precipitation. It can form a three-dimensional and biocompatible fibrin scaffold with a myriad of growth factors and proteins that are released progressively to the local environment and contribute to the accelerated postoperative bone healing. Gelatin (Gel) is a derivative of collagen and can promote cell adhesion and proliferation due to its unique sequence of amino acids, so it is suitable for bone tissue applications. This study examined the effects of Gel, FPG and their combinations as bone scaffold on the healing of surgically created critical-size defects in rat radius. Fifty critical size defects of 5 mm long were bilaterally created in the radial diaphysis of 25 rats. The animals were randomly divided into five equal groups as empty defect, autograft, Gel, FPG and Gel–FPG groups (n = 10 in each group). Radiographs of each forelimb were taken postoperatively on the 1st day and then at the 28th and 56th days post injury to evaluate bone formation, union and remodeling of the defect. After 56 days, the rats were euthanized and their harvested healing bone samples were evaluated by histopathology, scanning electron microscopy (SEM) and biomechanical testing. The results of present study showed that the Gel alone did not significantly affect bone healing and regeneration; however, the Gel treated defects promoted healing more than those that were left untreated (negative control). Furthermore, the FPG-enhanced grafts provided a good scaffold containing numerous growth factors for proliferation of osteoinduction and was effective in improving the structural and functional properties of the newly formed bone more than that of the untreated and also the Gel treated groups. Incorporation of Gel into the FPG scaffold improved healing potential of the FPG scaffold; however, it was still inferior to the autograft (positive control). Although the Gel–FPG scaffolds had best effectiveness during bone regeneration, it still needs to be further enhanced by incorporation of the ceramic and osteoinductive biomaterials.  相似文献

Common safety practices in pesticide use were assessed among apple farmers in Ardabil Province, Iran. With reference to the place of storing pesticides, most farmers (60%) stated that they store the pesticides in stalls and warehouses, while 8.5% stated that they store the pesticides in their houses. The majority of the farmers (71.5%) stated that they prepare the pesticide sprays in the orchards or next to irrigation wells. Almost one out of three farmers (32.8 %) reported that they dump the empty containers in the orchard fields and almost an equal proportion (30.2%) reported that they usually bury the empty containers. Irritating (burning) eyes and blurred vision were symptoms that the majority of respondents had experienced. Most farmers used trousers, long-sleeved blouses/shirts, and gloves when spraying, but coveralls and goggles were rarely used. Age and farming experience negatively affected PPE use and safety behavior, namely old and experienced farmers did not follow safety rules. On the contrary, education, information about pesticides, and farming as the main profession promoted safety behavior. Overall, significant levels of exposure among farmers can be concluded due to pesticide use. The extension services should aim human safety in relation to pesticide use in agriculture.  相似文献

The present study provides the length–weight and length–length relationships information (LWR & LLR) for three Blenny species; Antennablennius adenensis from the coast of Larak Island in the Persian Gulf and Istiblennius pox and Omobranchus mekranensis, which were captured in the coasts of Chabahar city in the Gulf of Oman, Iran. Total length (TL) and standard length (SL) were determined as well as weight. Specimens were collected occasionally in summer 2017 and winter 2018 by Hand net. The length–weight parameter b for these species was 2.84, 3.29 and 3.06 respectively with a high coefficient of determination (r2 ≥ 0.95). All LLRs were highly correlated (r2 > 0.99). All three species had no previous report for LWR and LLR estimates in FishBase.  相似文献

The geographical distribution and ecological preferences of Haemaphysalis in domestic animals in Iran were studied 4 times a year from April 2003 to March 2005. A total of 1,622 ixodid tick specimens were collected from 3 different zones. Among them, 108 (6.7%) Haemaphysalis ticks, consisting of 6 species, were identified; H. punctata (3.4%), H. parva (0.5%), H. sulcata (0.6%), H. choldokovskyi (1.7%), H. concinna (0.06%) and Haemaphysalis sp. (0.6%). H. punctata was the most abundant species, whereas H. concinna was the rarest species collected in humid and sub-humid zones on cattle, sheep and goats. H. choldokovskyi was principally collected from sheep and goats grazed in cold mountainous areas. The infested areas consisted of Caspian Sea (Guilan, Mazandaran, Golestan, and central provinces), mountainous (Azarbaiejan, Ardebil, Kohgilouyeh, and Kordestan) and semi-dessert (Khorasan, Semnan, Kerman, Sistan, and Baluchestan) zones. The Caspian Sea zone (23.6%) was the most highly infested region. The results show that various species of Haemaphysalis ticks infest domestic ruminants in Iran and each tick species show characteristic geographical distributions.  相似文献

Adipose derived mesenchymal stem cells (ASCs) transplantation is a novel immunomodulatory therapeutic tool to ameliorate the symptom of inflammatory bowel disease (IBD). The objective of this study was to investigate the therapeutic effects of combined sufasalazine and ASCs therapy in a rat model of IBD. After induction of colitis in rats, ASCs were cultured and intraperitoneally injected (3 × 106cells/kg) into the rats on Days 1 and 5 after inducing colitis, in conjunction with daily oral administration of low dose of sulfasalazine (30 mg/kg). The regenerative effects of combination of ASCs and sulfasalazine on ulcerative colitis were assessed by measuring body weight, colonic weight/length ratio, disease activity index, macroscopic scores, histopathological examinations, cytokine, and inflammation markers profiles. In addition, western blot analysis was used to assess the levels of nuclear factor-kappa B (NF-κB) and apoptosis related proteins in colitis tissues. Simultaneous treatment with ASCs and sulfasalazine was associated with significant amelioration of disease activity index, macroscopic and microscopic colitis scores, as well as inhibition of the proinflammatory cytokines in trinitrobenzene sulfonic acid (TNBS)-induced colitis. Moreover, combined ASCs and sulfasalazine therapy effectively inhibited the NF-κB signaling pathway, reduced the expression of Bax and prevented the loss of Bcl-2 proteins in colon tissue of the rats with TNBS-induced colitis. Furthermore, combined treatment with ASCs and sulfasalazine shifted inflammatory M1 to anti-inflammatory M2 macrophages by decreasing the levels of MCP1, CXCL9 and increasing IL-10, Arg-1 levels. In conclusion, combination of ASCs with conventional IBD therapy is potentially a much more powerful strategy to slow the progression of colitis via reducing inflammatory and apoptotic markers than either therapy alone.  相似文献
