To investigate the integrated evolution of mitochondrial and nuclear genomes in the eukaryotic cell, we have focused our attention on OXPHOS (oxidative phosphorylation) gene families which encode proteins involved in the main mitochondrial function. The present study reports the phylogenetic analysis of two OXPHOS gene families: ATP synthase subunit c (or lipid binding protein, LBP) and Cytochrome c (Cytc). Both gene families possess a higher expansion trend than the typically low duplication rate of OXPHOS genes in Metazoa, but follow a completely different evolutionary history, especially in mammals. LBP is represented by three well conserved isoforms in all mammals (P1, P2, P3): only P3 possesses a clearly conserved isoform in all Vertebrates, P1 and P2 were already present before the bird-mammal divergence and there are preliminary evidence from the in silico analysis that P1, the most evolutionary divergent isoform, is poorly expressed and not regulated by NRF1. In contrast, Cytc family presents at least two duplicated genes in all the analysed Vertebrates, is subject to a high expansion trend, especially of processed pseudogenes in mammals, and some events of gain and loss of function can be supposed.  相似文献

Background and aims

Approximately 50 % of belowground organic carbon is present in the northern permafrost region and due to changes in climate there are concerns that this carbon will be rapidly released to the atmosphere. The release of carbon in arctic soils is thought to be intimately linked to the N cycle through the N cycle’s influence on microbial activity. The majority of new N input into arctic systems occurs through N2-fixation; therefore, N2-fixation may be the key driver of greenhouse gases from these ecosystems.

Methods

At Alexandra Fjord lowland, Ellesmere Island, Canada concurrent measurements of N2-fixation, N mineralization and nitrification rates, dissolved organic soil N (DON) and C, inorganic soil N and surface greenhouse gas fluxes (CO2, N2O and CH4) were taken in two ecosystem types (Wet Sedge Meadow and Dryas Heath) over the 2009 growing season (June-August). Using Structural Equation Modelling we evaluated the hypothesis that CO2, CH4 and N2O flux are linked to N2-fixation via the N cycle.

Results

The soil N cycle was linked to CO2 flux in the Dryas Heath ecosystem via DON concentrations, but there was no link between the soil N cycle and CO2 flux in the Wet Sedge Meadow. Methane flux was also not linked to the soil N cycle, nor surface soil temperature or moisture in either ecosystem. The soil N cycle was closely linked to N2O emissions but via nitrification in the Wet Sedge Meadow and inorganic N in the Dryas Heath, indicating the important role of nitrification in net N2O flux from arctic ecosystems.

Conclusions

Our results should be interpreted with caution given the high variability in both the rates of the N cycling processes and greenhouse gas flux found in both ecosystems over the growing season. However, while N2-fixation and other N cycling processes may play a more limited role in instantaneous CO2 emissions, these processes clearly play an important role in controlling N2O emissions.  相似文献

Microorganisms colonizing surfaces can exude a wide range of substances, generally called Extracellular Polymeric Substances (EPS). While EPS has often been visualized as thick mature strata embedding microbes, the initial phases of EPS production, its structure at the micro- and nanoscale and the microbial wall areas involved in its exudation are less known. In this work we use Atomic Force Microscopy to image EPS produced by the fungus Paxillus involutus on phyllosilicate surfaces. Hyphal tips initially deposit EPS which assumes the shape of a “halo” surrounding hyphae. The fusion of adjacent EPS halos is likely responsible for the creation of EPS monolayers covering mineral surfaces. It is also proposed that a specific region of hyphae initiates the formation of mineral channels produced by fungi. The results presented here permit for the first time to propose a model for the initial stages of EPS accumulation in fungi and filamentous microorganisms in general.  相似文献

The transformer gene in Ceratitis capitata (Cctra ep ) is the founding member of a family of related SR genes that appear to act as the master epigenetic switch in sex determination in insects. A functional protein seems to be produced only in individuals with a female XX karyotype where it is required to maintain the productive mode of expression through a positive feedback loop and to direct female development by instructing the downstream target genes accordingly. When zygotic activation of this loop is prevented, male development follows. Recently, tra ep orthologues were isolated in more distantly related dipteran species including Musca domestica, Glossina morsitans and Lucilia cuprina and in the Hymenopterans Apis mellifera and Nasonia vitripennis. All of these tra ep orthologues seem to act as binary switches that govern all aspects of sexual development. Transient silencing leads to complete masculinization of individuals with a female karyotype. Reciprocally, in some systems it has been shown that transient expression of the functional TRA product is sufficient to transactivate the endogenous gene and implement female development in individuals with a male karyotype. Hence, a mechanism based on tra ep epigenetic autoregulation seems to represent a common and presumably ancestral single principle of sex determination in Insecta. The results of these studies will not only be important for understanding divergent evolution of basic developmental processes but also for designing new strategies to improve genetic sexing in different insect species of economical or medical importance.  相似文献

Microcosms with Pinus sylvestris seedlings in symbiosis with the fungus mycorrhizal Paxillus involutus were established, and atomic force microscopy (AFM) was used to characterise plant photosynthate-driven fungal interactions with mineral surfaces. Comparison of images of the same area of the minerals before and after mycorrhizal fungal colonization showed extensive growth of hyphae on three different mineral surfaces – hornblende, biotite and chlorite. A layer of biological exudate, or biolayer, covered the entire mineral surface and was composed of globular features of diameter 10–80 nm, and the morphology of the biolayer differed among mineral types. Similar-sized components were found on the fungal hyphae, but with a more elongated profile. Biolayer and hyphae surfaces both appeared to be hydrophobic with the hyphal surfaces yielding higher maximal adhesive interactions and a wider range of values: the mean (± SE) adhesive forces were 2.63 ± 0.03 and 3.46 ± 0.18 nN for biolayer and hypha, respectively. The highest adhesion forces are preferentially localized at the hyphal surface above the Spitzenkörper region and close to the tip, with a mean interaction force in this locality of 5.24 ± 0.49 nN. Biolayer thickness was between 10 and 40 nm. The underlying mineral was easily broken up by the tip, in contrast to the native mineral. These observations of mineral surfaces colonised by mycorrhizal fungus demonstrate how fungal hyphae are able to form a layer of organic exudates, or biolayer, and its role in hyphal attachment and potential weathering of ferromagnesian silicates, which may supply nutrients to the plant.  相似文献

Mitochondrial DNA variability of West New Guinea populations.   总被引:3,自引:0,他引:3  
This paper reports human mitochondrial DNA variability in West New Guinea (the least known, western side of the island of New Guinea), not yet described from a molecular perspective. The study was carried out on 202 subjects from 12 ethnic groups, belonging to six different Papuan language families, representative of both mountain and coastal plain areas. Mitochondrial DNA hypervariable region 1 (HVS 1) and the presence of the 9-bp deletion (intergenic region COII-tRNA(Lys)) were investigated. HVS 1 sequencing identified 73 polymorphic sites defining 89 haplotypes; the 9-bp deletion, which is considered a marker of Austronesian migration in the Pacific, was found to be absent in the whole West New Guinea study sample. Statistical analysis applied to the resulting haplotypes reveal high heterogeneity and an intersecting distribution of genetic variability in these populations, despite their cultural and geographic diversity. The results of subsequent phylogenetic approaches subdivide mtDNA diversity in West New Guinea into three main clusters (groups I-III), defined by sets of polymorphisms which are also shared by some individuals from Papua New Guinea. Comparisons with worldwide HVS 1 sequences stored in the MitBASE database show the absence of these patterns outside Oceania and a few Indonesian subjects, who also lack the 9-bp deletion. This finding, which is consistent with the effects of genetic drift and prolonged isolation of West New Guinea populations, lead us to regard these patterns as New Guinea population markers, which may harbor the genetic memory of the earliest human migrations to the island.  相似文献
