Calcium is an essential nutrient required for critical biological functions. Calcium supplementation is to be evaluated using immature female rats. The present study focused on some blood parameters, gonadal development and bone structure. Forty immature female Sprague–Dawley rats were randomly divided into four equal-sized groups (80 g average body weight) to receive calcium chloride dihydrate (group I: control; groups II, III and IV: received 20 mg, 40 mg and 60 mg per kg body weight, respectively) for 5 weeks. Rats were decapitated, and their trunk blood was sampled for biochemical assays. Cholesterol, triglycerides, glucose and calcium were measured. Gonadal and bone structure were histologically evaluated. Results revealed that treatment of developing female rats with three calcium doses used have no marked effect on the serum calcium and cholesterol levels. However, serum triglyceride level and body weight gain are significantly decreased in rats treated with all of the three calcium doses. Serum glucose level showed a marked increase in animals treated with the higher calcium doses. Moreover, observable histological alterations are recognized in the ovaries. Bones of the experimental animals also showed morphological alterations. These results suggest that increasing calcium supplementation decreases triglycerides and percentage body weight gain and positively affects the bone and gonadal development.  相似文献

During surveys carried out in 2008 in the nurseries of some ornamental and medical plants, about 90 plant samples belonging to six plant species were collected. Cucumber mosaic virus (CMV) was detected by routinely double antibody sandwich-enzyme linked immunosorbent assay (DAS-ELISA) in most tested plants. In Vinca rosea, Ocimum basilicum and Pelargonium sp., which reacted positively for CMV, 100% of the samples were infected. High absorbance values were obtained when extracts were prepared from these species and then examined by DAS-ELISA. The results indicated that CMV concentration on O. basilicum is greater than 50 μg ml?1 when compared with purified CMV standards. High absorbance values occur even under conditions fully unsuitable for the detection of antigens. This result suggested that the strong ELISA absorbance values were nonspecific reactions with materials in plant extracts. So, other detection methods including dot blot, Western blot and bioassay was used for comparing with ELISA test. The nonspecific reactions were substantiated when CMV was not detected by dot blot and Western blot. Also, infectious CMV was not detected in bioassay involving inoculation of extracts onto Chenopodium amaranticolor and Nicotiana tabacum var. White Burley plants as indicator hosts. Addition of bovine serum albumin (BSA) or polyvinylpyrolidene (PVP) to extraction buffer or to wells after IgG coating for DAS-ELISA reduced nonspecific reactions. Finally, CMV was isolated from V. rosea symptomatic plants, which give a positive reaction by DAS-ELISA, dot blot, Western blot and bioassay. CMV vinca-isolate was identified based on transmission, host range, serological tests, purification and electron microscope.  相似文献

The ability of the flavonoid lentinan (LAN) to enhance the repair of paclitaxel (PAC)‐induced DNA damage and apoptosis in mouse bone marrow cells was investigated. Moreover, the possible mechanism underlying this modulation was determined. LAN was neither genotoxic nor apoptogenic at doses equivalent to 1 or 2 mg/kg/day. Pretreatment of mice with LAN significantly enhances the repair of PAC‐induced DNA damage and bone marrow suppression in a dose dependent manner. Moreover, LAN affords significant protection against PAC‐induced apoptosis. A significant increase of reactive oxygen species and a decrease in reduced glutathione levels were observed after PAC treatment and prior administration of LAN before PAC challenge ameliorated these oxidative stress markers. Conclusively, our study provides, for the first time, that LAN enhances the repair of PAC‐induced DNA damage and apoptosis that resides, at least in part, on its ability to modulate the cellular antioxidant levels and consequently protect bone marrow cells from PAC genotoxicity. © 2013 Wiley Periodicals, Inc. The growth of an axon toward its target results from the reorganization of the cytoskeleton in response to environmental guidance cues. Recently developed imaging technology makes it possible to address the effect of such cues on the neural cytoskeleton directly. Although high resolution studies can be carried out on neurons in vitro, these circumstances do not recreate the complexity of the natural environment. We report here on the arrangement and dynamics of microtubules in live neurons pathfinding in response to natural guidance cues in situ using the embryonic grasshopper limb fillet preparation. A rich microtubule network was present within the body of the growth cone and normally extended into the distal growth cone margin. Complex microtubule loops often formed transiently within the growth cone. Branches both with and without microtubules were regularly observed. Microtubules did not extend into filopodia. During growth cone steering events in response to identified guidance cues, microtubule behaviour could be monitored. In turns towards guidepost cells, microtubules selectively invaded branches derived from filopodia that had contacted the guidepost cell. At limb segment boundaries, microtubules displayed a variety of behaviors, including selective branch invasion, and also invasion of multiple branches followed by selective retention in branches oriented in the correct direction. Microtubule invasion of multiple branches also was seen in growth cones migrating on intrasegmental epithelium. Both selective invasion and selective retention generate asymmetrical microtubule arrangements within the growth cone, and may play a key role in growth cone steering events.  相似文献

Ideal coatings for orthopedic implants should be able to induce excellent osseointegration with host bone tissue, which requires good osteogenic responses and limited inflammatory reactions. Cerium oxide (CeO2) ceramics have anti-oxidative properties and can be used to decrease mediators of inflammation, making them attractive for biomedical application. In this study, two kinds of CeO2 incorporated calcium silicate coatings (CS-10Ce and CS-30Ce) were prepared via plasma spraying technique, and the effects of CeO2 addition on the responses of bone mesenchymal stem cells (BMSCs) and RAW264.7 macrophages were evaluated. The CS-10Ce and CS-30Ce coatings were characterized by X-ray diffraction, scanning electron microscopy, and X-ray photoelectron spectroscopy. An increase in CeO2 content in the coatings resulted in enhanced chemical stability and better BMSCs osteogenic behaviors in terms of cell adhesion, proliferation, ALP activity, and mineralized nodule formation. With respect to either ZrO2-added or unmodified CS coating, the CS-30Ce coating elicited higher effects on the macrophages, suppressing the gene expressions of pro-inflammatory (M1) markers (CCR7, IL-6, and TNF-α), while upregulating the expressions of anti-inflammatory (M2) markers (CD206, IL-1ra, and IL-10); moreover, it could also increase the expression of osteoinductive molecules (BMP2 and TGF-β1) by the macrophages. The results suggested that the regulation of BMSCs behaviors and macrophage-mediated responses at the coating’s surface was related to CeO2 incorporation. The incorporation of CeO2 in CS coatings can be a valuable strategy to promote osteogenic responses and mitigate inflammatory reactions.  相似文献

This is the first report on the degradation of poly(3-hydroxybutyrate) (PHB), and its copolymers poly(3-hydroxyvalerate) P(3HB-co-10-20% HV) by Nocardiopsis aegyptia, a new species isolated from marine seashore sediments. The strain excreted an extracellular PHB depolymerase and grew efficiently on PHB or its copolymers as the sole carbon sources. The degradation activity was detectable by the formation of a transparent clearing zone around the colony on an agar Petri plate after 25 days, or a clearing depth under the colony in test tubes within 3 weeks. The previous techniques proved that the bacterium was able to assimilate the monomeric components of the shorter alkyl groups of the polymers. Nocardiopsis aegyptia hydrolyzed copolymers 10-20% PHBV more rapidly than the homopolymer PHB. The bacterial degradation of the naturally occurring sheets of poly(3-hydroxybutyrate), and its copolymer poly(3-hydroxybutyrate-co-3-hydroxyvalerate) was observed by scanning electron microscopy (SEM). The samples were degraded at the surface and proceeded to the inner part of the materials. Clear morphological alterations of the polymers were noticed, indicating the degradative capability of the bacterium. Plackett-Burman statistical experimental design has been employed to optimize culture conditions for maximal enzyme activity. The main factors that had significant positive effects on PHB depolymerase activity of Nocardiopsis aegyptia were sodium gluconate, volume of medium/flask and age of inoculum. On the other hand, MgSO4.7H2O, KH2PO4, K2HPO4 and NH4NO3 exhibited negative effects. Under optimized culture conditions, the highest activity (0.664 U/mg protein) was achieved in a medium predicted to be near optimum containing (in g/L): PHB, 0.5; C6H11O7Na, 7.5; MgSO4.7H2O, 0.35; K2HPO4, 0.35; NH4NO3, 0.5; KH2PO4, 0.35; malt extract, 0.5 and prepared with 50% seawater. The medium was inoculated with 1% (v/v) spore suspension of 7 days old culture. Complete clarity of the medium was achieved after 3 days at 30 degrees C.  相似文献

The flavonoid glycosides of Medicago radiata as well as M. arabica, M. polymorpha, M. sativa, Trigonella coerulescens, T. foenum-graecum and T. spicata were studied in detail. Major glycosides identified were the 7-glucuronides of apigenin, luteolin, chrysoeriol and tricin, as well as lesser amounts of di- and triglucuronides of chrysoeriol and tricin. Also identified were the 3-robinobioside and 3,7-diglucoside of kaempferol, along with lesser amounts of quercetin-3,7-diglucoside, 4′,7-dihydroxyflavone, 3′,4′,7-trihydroxyflavone, formononetin and daidzein. Twelve other Medicago and Trigonella species were also studied for their flavonoid aglycones. The systematic position of M. radiata is discussed.  相似文献

The antioxidant status as well as protein composition of faba bean leaves infected with Bean yellow mosaic virus (BYMV) and the effect of salicylic acid application was examined in this paper. Some modifications in the antioxidant status were observed by changing some antioxidant enzymes activities and contents of antioxidant metabolites. BYMV-infected leaves revealed POD, CAT, APX and SOD induced activities while SA treatments could inhibit POD, CAT activities but induced SOD activity. The enzyme activities seemed to be SA concentration dependant. Higher H2O2 and MDA concentrations were recorded in virus-infected leaves than those of the corresponding controls while treatment with SA followed by virus inoculation caused lowering of MDA concentration and reducing the damage due to lipid peroxidation. Moreover, because of virus infection and/or SA treatments, an increase in the amounts of phenolics and flavonoids was noticed. As compared to the control, BYMV infection or SA application caused pronounced increase in the antioxidant activity of leaf extracts detected by DPPH assay, indicating an increase in the amounts of antioxidant compounds occurred. To test the protein composition, the contents of each protein fractions (soluble, insoluble and total) were analyzed and the change in protein patterns was visualized using SDS-PAGE. The BYMV-infected bean leaves had protein contents higher than the control indicating accumulation of pathogenesis-related proteins. Moreover, spraying SA with or without virus inoculation could accumulate soluble, insoluble and total proteins and the pattern of increase was in accordance with SA concentration. Alterations in protein patterns were observed in faba bean leaves (Vicia faba cv Giza 461) in response to BYMV infection and SA treatments. Because of BYMV infection and SA treatments, the protein profiles showed new bands in comparison to the control. Some polypeptides were highly accumulated in treatments of SA followed by virus inoculation. Changing antioxidant status and accumulation of some antioxidant metabolites as well as the pronounced alterations in the protein composition indicate a kind of plant response against pathogen invasion and in case of SA treatment it is considered a way by which a defence response was initiated and/or activated.  相似文献
