As translation proceeds, nascent polypeptides pass through an exit tunnel that traverses the large ribosomal subunit. Three ribosomal proteins, termed Rpl4, Rpl17, and Rpl39 expose domains to the interior of the exit tunnel of eukaryotic ribosomes. Here we generated ribosome-bound nascent chains in a homologous yeast translation system to analyze contacts between the tunnel proteins and nascent chains. As model proteins we employed Dap2, which contains a hydrophobic signal anchor (SA) segment, and the chimera Dap2α, in which the SA was replaced with a hydrophilic segment, with the propensity to form an α-helix. Employing a newly developed FLAG exposure assay, we find that the nascent SA segment but not the hydrophilic segment adopted a stable, α-helical structure within the tunnel when the most C-terminal SA residue was separated by 14 residues from the peptidyl transferase center. Using UV cross-linking, antibodies specifically recognizing Rpl17 or Rpl39, and a His6-tagged version of Rpl4, we established that all three tunnel proteins of yeast contact the SA, whereas only Rpl4 and Rpl39 also contact the hydrophilic segment. Consistent with the localization of the tunnel exposed domains of Rpl17 and Rpl39, the SA was in contact with Rpl17 in the middle region and with Rpl39 in the exit region of the tunnel. In contrast, Rpl4 was in contact with nascent chain residues throughout the ribosomal tunnel.  相似文献

The perivertebral musculature of lizards is critical for the stabilization and the mobilization of the trunk during locomotion. Some trunk muscles are also involved in ventilation. This dual function of trunk muscles in locomotion and ventilation leads to a biomechanical conflict in many lizards and constrains their ability to breathe while running (“axial constraint”) which likely is reflected by their high anaerobic scope. Furthermore, different foraging and predator‐escape strategies were shown to correlate with the metabolic profile of locomotor muscles in lizards. Because knowledge of muscle's fiber‐type composition may help to reveal a muscle's functional properties, we investigated the distribution pattern of muscle fiber types in the perivertebral musculature in two small lizard species with a generalized body shape and subjected to the axial constraint (Dipsosaurus dorsalis, Acanthodactylus maculatus) and one species that circumvents the axial constraint by means of gular pumping (Varanus exanthematicus). Additionally, these species differ in their predator‐escape and foraging behaviors. Using refined enzyme‐histochemical protocols, muscle fiber types were differentiated in serial cross‐sections through the trunk, maintaining the anatomical relationships between the skeleton and the musculature. The fiber composition in Dipsosaurus and Acanthodactylus showed a highly glycolytic profile, consistent with their intermittent locomotor style and reliance on anaerobic metabolism during activity. Because early representatives of diapsids resemble these two species in several postcranial characters, we suggest that this glycolytic profile represents the plesiomorphic condition for diapsids. In Varanus, we found a high proportion of oxidative fibers in all muscles, which is in accordance with its high aerobic scope and capability of sustained locomotion. J. Folding of RNA molecules into their functional three-dimensional structures is often supported by RNA chaperones, some of which can catalyse the two elementary reactions helix disruption and helix formation. Hfq is one such RNA chaperone, but its strand displacement activity is controversial. Whereas some groups found Hfq to destabilize secondary structures, others did not observe such an activity with their RNA substrates. We studied Hfq’s activities using a set of short RNAs of different thermodynamic stabilities (GC-contents from 4.8% to 61.9%), but constant length. We show that Hfq’s strand displacement as well as its annealing activity are strongly dependent on the substrate’s GC-content. However, this is due to Hfq’s preferred binding of AU-rich sequences and not to the substrate’s thermodynamic stability. Importantly, Hfq catalyses both annealing and strand displacement with comparable rates for different substrates, hinting at RNA strand diffusion and annealing nucleation being rate-limiting for both reactions. Hfq’s strand displacement activity is a result of the thermodynamic destabilization of the RNA through preferred single-strand binding whereas annealing acceleration is independent from Hfq’s thermodynamic influence. Therefore, the two apparently disparate activities annealing acceleration and duplex destabilization are not in energetic conflict with each other.  相似文献

Summary: Changes in intraspecific social organization are of particular interest to sociobiologists, because an understanding of the genetic and ecological determinants of different social forms bears on the origins of sociality and mating isolation. The Southern Yellowjacket, Vespula squamosa (Drury), occurs as both polygyne and single queen nests. This study examined intra-caste size differences between both social forms of nests. Four polygyne and 13 single queen nests were collected from counties in Florida and Georgia. All single queen nests were one season old, whereas polygyne nests were at least 2 years old. The latter had significantly more workers per nest (p < 0.005), but mean values varied by over 50-fold. Workers and gynes from each social form were compared for six measures of body size: weight, wing length, femur length, thorax length, thorax width and head width. Correlation coefficients for pairs of all traits were highest between head width and both measures of thorax size (r > 0.72), whereas the lowest positive correlations were obtained between weight and all other measures of size (r < 0.64). Gynes of both social forms differed significantly in weight (p < 0.001) and thorax length (p < 0.04), whereas workers differed significantly for all six measures of size (p < 0.04 to p < 0.001).  相似文献

The fish fauna of Gebel Aulia reservoir (White Nile) consists of 52 species withAlestes baremoze Joannis andBrycinus nurse Rüppell dominating in numbers and biomass. The seasonality and condition ofA. baremoze, A. dentex (L.) andB. nurse are described in relation to various limnological factors in the reservoir, close to the dam. The zooplankton assemblage is small-sized, consists mainly of small Cladocera and shows striking seasonality. The hydrological regime, temperature and the grazing effect of fish are the major factors influencing its development.The three species belong to a eurytopic but basically riverine fish guild. They show a seasonal cycle of abundance with high numbers at periods of water storage (October–April) and low numbers during riverine conditions. This seasonal pattern of occurrence, the size structure of the population and the maturity indices give strong evidence that no resident population is present at the study site, but upstream migrations of the immature fish occur at low water level (May–September). Return migrations of juvenile fish into the area at the dam proper take place from October until April.

Die quantitative Bedeutung und das jahreszeitliche Auftreten vonA. baremoze Joannis,A. dentex Cuvier andB. nurse Rüppell wurde im Zusammenhang mit dem saisonalen Zyklus der hydrologischen und limnologischen Bedingungen untersucht. Die Häufigkeit der Fische im Staubereich ist in der Stauphase (Oktober–April) hoch und in den Monaten, in denen kein Rückstau erfolgt (Mai–September), gering.Das saisonale Muster des Auftretens, die größenstruktur der Fischpopulation and die Maturitätsfaktoren im eigentlichen Staubereich keine seßhafte Population vorhanden ist. Die Fische unternehmen kurz vor der Geschlechtsreife anadrome Wanderungen, die Jungfische wandern katadrom.

Three networks/projects involving 27 European countries were established to investigate the quality of second-line drug (SLD) susceptibility testing with conventional and molecular methods. 1. The “Baltic-Nordic TB-Laboratory Network” comprised 11 reference laboratories in the Baltic-Nordic States. They performed SLD testing in the first phase with a panel of 20 Mycobacterium tuberculosis strains. After several laboratories made technical changes a second panel of 10 strains with a higher proportion of resistant strains were tested. Although the concordance for Ofloxacin, Kanamycin, and Capreomycin was consistently high, the largest improvements in performance were achieved for the analysis of Ofloxacin resistant (from 88.9 to 95.0%), and Capreomycin resistant (from 71.0 to 88.9%) strains. 2. Within the FP7 TB PAN-NET project (EU Grant agreement 223681) a quality control panel to standardize the EQA (External Quality Assurance) for first-line drugs (FLD) and SLD testing for phenotypic and molecular methods was established. The strains were characterized by their robustness, unambiguous results when tested, and low proportion of secondary drug resistances. 3. The (European Reference Laboratory Network-TB) ERLN-TB network analyzed four different panels for drug resistance testing using phenotypic and molecular methods; in two rounds in 2010 the 31 participating laboratories began with 5 strains, followed by 10 strains and 6 additional crude DNA extracts in 2011 and 2012 were examined by conventional DST and molecular methods. Overall, we demonstrated the importance of developing inter-laboratory networks to establish quality assurance and improvement of SLD testing of M. tuberculosis.  相似文献

Streptozotocin induces rapid degradation of deoxyribonucleic acid (DNA) in actively dividing or resting Bacillus subtilis cells. Difference spectroscopy showed that the antibiotic interacts specifically with cytosine containing mononucleotides in vitro. This interference occurs only within the very narrow pH range of 5 to 5.5 and is reversed immediately upon lowering or increasing the pH. No measurable interaction was observed between isolated DNA and streptozotocin. The possibility is discussed that interaction of streptozotocin with cytosine residues in cellular DNA, although possibly taking place at a very low frequency, may constitute the primary step inducing DNA strand breakage.  相似文献

The principal methods for determining the central venous pressure have been compared in 26 patients undergoing cardiac catheterization. The method in which the reference point is taken to be 10 cm. above the surface supporting a horizontal patient gave false high-pressure values.
