Abstract We have studied the response of two methylotrophic yeasts ( Hansenula polymorpha and Candida boidinii ) to toxin T-514, a toxin lethal to man, extracted from the shrub Karwinska homboldtiana . Growth experiments indicated a dose-response effect; at enhanced concentrations (50 μg/ml) the different subcellular organelles rapidly disintegrated resulting in death of the cultures. At non-lethal concentrations (<2 μg / ml ) growth ceased initially, but resumed after a lag period of 4 h. At the subcellular level a specific effect was observed on peroxisomal integrity. Distinct holes appeared in the peroxisomal membranes, resulting in leakage of matrix proteins from these organelles. In addition, import of newly synthesized proteins appeared to be blocked since cytosolic aggregates of matrix proteins were formed. The peroxisomal damage was probably irreversible since affected organelles were degraded at later stages of incubation. Upon restoration of growth on methanol, new peroxisomes developed from those which had escaped degradation.  相似文献

A central theme in ecological research is to understand how species interactions contribute to community dynamics. Species interactions are the basis of parametric (model-driven) and nonparametric (model-free) approaches in theoretical and empirical work. However, despite their different interpretations across these approaches, these measures have occasionally been used interchangeably, limiting our opportunity to use their differences to gain new insights about ecological systems. Here, we revisit two of the most used measures across these approaches: species interactions measured as constant direct effects (typically used in parametric approaches) and local aggregated effects (typically used in nonparametric approaches). We show two fundamental properties of species interactions that cannot be revealed without bridging these definitions. First, we show that the local aggregated intraspecific effect summarizes all potential pathways through which one species impacts itself, which are likely to be negative even without any constant direct self-regulation mechanism. This property has implications for the long-held debate on how communities can be stabilized when little evidence of self-regulation has been found among higher-trophic species. Second, we show that a local aggregated interspecific effect between two species is correlated with the constant direct interspecific effect if and only if the population dynamics do not have any higher-order direct effects. This other property provides a rigorous methodology to detect direct higher-order effects in the field and experimental data. Overall, our findings illustrate a practical route to gain further insights about non-equilibrium ecological dynamics and species interactions.  相似文献

Abstract: Angiotensin-converting enzyme (ACE) activity was measured by a radiochemical assay in 30 specific areas of the rat brain stem. ACE activity is unevenly distributed, with a 60-fold difference between the lowest and the highest activity. The area postrema exhibits the highest activity. The substantia nigra (pars reticulata), the locus coeruleus, the areas A1 and A2, the nuclei commissuralis, and tractus solitarii have a substantial ACE activity, whereas the lowest activity is found in the raphe nuclei and the nuclei of the reticular formation.  相似文献

A standard DNA modification methyltransferase (MTase) selection protocol was followed to clone the BstVI restriction and modification system from Bacillus stearothermophilus in Escherichia coli. Both genes were contained in a 4.4-kb EcoRI fragment from B. stearothermophilus V chromosomal DNA. The heterologous expression of these genes did not depend on their orientation in the vector, suggesting that the genes are expressed in E. coli under the control of promoters located on the cloned fragment. Subcloning experiments demonstrated that the bstVIR gene was expressed in the absence of its cognate MTase.  相似文献

The foundational concepts behind the persistence of ecological communities have been based on two ecological properties: dynamical stability and feasibility. The former is typically regarded as the capacity of a community to return to an original equilibrium state after a perturbation in species abundances and is usually linked to the strength of interspecific interactions. The latter is the capacity to sustain positive abundances on all its constituent species and is linked to both interspecific interactions and species demographic characteristics. Over the last 40 years, theoretical research in ecology has emphasized the search for conditions leading to the dynamical stability of ecological communities, while the conditions leading to feasibility have been overlooked. However, thus far, we have no evidence of whether species interactions are more conditioned by the community''s need to be stable or feasible. Here, we introduce novel quantitative methods and use empirical data to investigate the consequences of species interactions on the dynamical stability and feasibility of mutualistic communities. First, we demonstrate that the more nested the species interactions in a community are, the lower the mutualistic strength that the community can tolerate without losing dynamical stability. Second, we show that high feasibility in a community can be reached either with high mutualistic strength or with highly nested species interactions. Third, we find that during the assembly process of a seasonal pollinator community located at The Zackenberg Research Station (northeastern Greenland), a high feasibility is reached through the nested species interactions established between newcomer and resident species. Our findings imply that nested mutualistic communities promote feasibility over stability, which may suggest that the former can be key for community persistence.  相似文献
