From strains carrying two different F-prime factors, we recovered F' derivatives that acquired the trp chromosomal region. These F'trp plasmids can be isolated at a frequency of 10-5 to 10-6. They were characterized genetically by looking at the size of the trp segment they acquired and at the location of that segment in the parental F' plasmid. Results are discussed in relationship to possible transposition mechanisms.  相似文献

HindIII restriction endonuclease fragments of DNA from temperate Bacillus subtilis bacteriophage SP02 were cloned in B. subtilis by using the plasmid pC194. Three hybrid plasmids which permit growth of the mutant SP02 susL244 in suppressor-negative bacteria were isolated. SP02 gene L is thought to code for a DNA polymerase essential for autonomous replication of SP02 DNA. Extracts of bacteria carrying one of these hybrid plasmids, pC194-96, had 10- to 30-fold increased DNA polymerase activity. The plasmid-induced DNA polymerase activity differed from that of the known B. subtilis DNA polymerases in several respects. The results of the experiments support the idea that phage SP02 codes for a new DNA polymerase.  相似文献

Pathogenic strains of Sporothrix schenkii may show triangular spores, whose angular shape is maintained by a tiebeam effect in the inner cell wall structure. This difference in wall structure lies adjacent to a folded and possibly more active part of the spore cytoplasm. The supposed generation of asci in old cultures was simulated by the death of hyphae which are reinvaded by intrahyphal growth with intrahyphal spore production, while true asci were not seen.  相似文献

Summary In order to evaluate the feasibility to localize correctly 67Ga citrate and 99mTc pertechnetate in tissues, the resolution of these radioactive compounds were measured in a model system using four different autoradiographic techniques, wet as well as dry.Wet autoradiographic techniques gave an almost complete loss of 67Ga. In deparaffinized sections of fixed and paraffin-embedded tissue the remaining 67Ga, which was probably bound to proteins, gave a resolution of 2.6 . 99mTc was either completely lost in wet techniques or the procedure could not be performed at all because of the very short half life of 99mTc.The resolution of 67Ga in a dry autoradiographic technique (according to Stumpf) was 6.9 and the resolution of 99mTc 22.6 .The technique in which frozen sections are thawed on dry film and consequently dryed, gave slightly better resolutions than the dry technique (according to Stumpf) with 67Ga as well as 99mTc.It is concluded that for the histological localization of 67Ga and 99mTc a dry technique is required. However, the use of a wet technique can be considered, provided a loss of the radioisotope is acceptable and the procedure is controlled by a dry technique.  相似文献

The conditions and requirements for an in vitro protein synthesizing system from the moss Tortula ruralis are outlined. Using this system the effects of desiccation, achieved quickly or slowly, were studied. Slowly dried moss retained fewer polyribosomes on desiccation but more active ribosomes than rapidly dried moss. Even in the completely desiccated moss the polyribosomes and/or free ribosomes present have retained their synthetic capacities. On rehydration, the slowly dried moss resumed protein synthesis more quickly than moss previously desiccated rapidly. Moss ribosomes are cycloheximide sensitive and chloramphenicol insensitive and thus the major protein synthesis occurs within the cytoplasm on rehydration. Extracted polyribosomes per se can withstand desiccation to a significant extent, suggesting that protection by the cytoplasm might not be necessary. The aquatic moss Hygrohypnum luridum can retain polyribosomal and ribosomal activity during desiccation, but this decreases greatly on rehydration.  相似文献

How environmental changes are affecting bird population dynamics is one of the most challenging conservation issues. Dietary studies of top avian predators could offer scope to monitor anthropogenic drivers of ecosystem changes. We investigated the diet of breeding Eleonora''s falcon in an area of Northeastern Algeria in the years 2010–2012. Feathers and insect remains originating from prey plucking behavior were analyzed, providing insights into the seasonally changing diet of this raptor, as well as the trans‐Mediterranean avian migration. A total of 77 species of birds (16 Sylviidae, 11 Turdidae, and 4 Emberizidae), 3 species of insects, and 1 lizard were identified among prey remains, reflecting a diverse diet. Diet composition and prey abundance varied seasonally, faithfully correlating with the passage of migrant birds as recorded from bird ring recoveries. Our findings suggest that dietary studies of predators might be deployed to investigate changes in bird migration. We discuss our results in the context of trans‐Mediterranean migration, with early‐season prey mainly comprising trans‐Saharan migrants (Apus apus and Merops apiaster) and late‐season prey being dominated by Mediterranean winter migrants (Erithacus rubecula, Turdus philomelos, Sylvia atricapilla, and Sturnus vulgaris). Notably, we observed a significant reduction in species richness of passerine remains in 2012, potentially highlighting a decline in the diversity of avian migrants.  相似文献

Upon antigen binding, the B cell receptor (BCR) undergoes clustering to form a signalosome that propagates downstream signaling required for normal B cell development and physiology. BCR clustering is dependent on remodeling of the cortical actin network, but the mechanisms that regulate actin remodeling in this context remain poorly defined. In this study, we identify the inositol 5-phosphatase INPP5B as a key regulator of actin remodeling, BCR clustering, and downstream signaling in antigen-stimulated B cells. INPP5B acts via dephosphorylation of the inositol lipid PI(4,5)P2 that in turn is necessary for actin disassembly, BCR mobilization, and cell spreading on immobilized surface antigen. These effects can be explained by increased actin severing by cofilin and loss of actin linking to the plasma membrane by ezrin, both of which are sensitive to INPP5B-dependent PI(4,5)P2 hydrolysis. INPP5B is therefore a new player in BCR signaling and may represent an attractive target for treatment of B cell malignancies caused by aberrant BCR signaling.  相似文献
