SUMMARY: Sterilized raw sewage sludge enriched with sulphate and inoculated with pure strains of Desulphovibrio desulphuricans produced negligible sulphide. Unsterilized sludge supplemented with 7% (w/v) CaSO4.2H2O and inoculated with crude cultures of sulphate-reducing bacteria obtained from sewage yielded 1·0% S2- (wt S2- produced as H2S/vol. of raw sludge) in 6 months at 30°. By repeated subculture more active cultures developed which produced 1% S2- in 7 days and 1·2–1·9% in 28 days. Digested sludge yielded only 0·1% S2-. In semicontinuous fermentations at 30°, raw sludge without added sulphate produced 20 times its own volume of gas containing 70% CH4 and 30% CO2. When 5% CaSO4.2H2O and an active crude culture of sulphate reducers were added, gas production decreased steadily to zero. There were no differences in pH, temperature and redox potential in sludges producing methane or sulphide. The chief cause of inhibition appeared to be the action of sulphide: 0·02% soluble sulphide (S2-) totally inhibited methane formation; 0·01% S2- initially decreased gas production by one-quarter but there was a slow recovery to normal, suggesting acclimatization of the methane-producing organisms to sulphide.
Linked fermentations, in which gas from a methane fermentation swept H2S from a sulphide fermentation, gave a final gas mixture of about 60% CH4, 30% CO2 and 5–10% H2S. The yield of sulphide depended on the rate of sweeping.  相似文献

There are two chemically distinct types of melanin: the red-yellow phaeomelanins and the brown-black eumelanins. While both melanins have been detected in human epidermis and cultured melanocytes, it is unknown how the phaeomelanin/eumelanin ratio in human melanocytes maintained in vitro relates to that in the epidermis from which they were isolated. This study uses high-performance liquid chromatography to quantify the eumelanin and phaeomelanin contents of epidermis and/or cultured melanocytes from 12 Europeans with lightly pigmented skin and 9 non-Europeans with more deeply pigmented skin. Epidermis from non-Europeans contained the highest levels of both eumelanin and phaeomelanin and had the lowest phaeomelanin/eumelanin ratios. In contrast, while cultured melanocytes from non-Europeans also had higher levels of eumelanin and phaeomelanin than melanocytes from Europeans, there was no difference in the phaeomelanin/eumelanin ratios in the two groups. However, the phaeomelanin/eumelanin ratios were higher in the cultured melanocytes than in the corresponding epidermis so that while eumelanin was the predominant melanin in the epidermis, phaeomelanin was the major melanin in the cultured melanocytes. These observations may have important implications for the use of cultured human melanocytes in the study of melanogenesis in man.  相似文献

Heat pulse velocity (HPV) apparatus was used to investigatethe relation between the radial HPV profile and the positionof growth rings in two Pinus patula trees (aged 4 and 7 years),growing in Frankfort State Forest in the eastern Transvaal,South Africa. Successive measurements of HPV at 3.0 mm intervalsalong each of four radii in each tree revealed a cyclical pattern,with maxima and minima correlated with summer and winter wood,respectively. We recommend that probe emplacement in fast-growingpines with relatively wide growth rings should be to randomly-assigneddepths in the sapwood. Confidence limits associated with samplemeans of wound-corrected HPV obtained with 4, 8 or 12 probesets were generated by simulating the repeated selection ofa randomly-chosen HPV measurement from each sapwood radius.We conclude from this analysis that a minimum of 12 point measurementsper tree is required to obtain an acceptable estimate of sapflow. In older trees where a decline in HPV is expected towardsthe centre of the trunk, sampling must be modified to avoidbias caused by deeper measurements representing proportionatelyless of the total sapwood area than those taken at more shallowdepths in the sapwood. The carotenoid composition of undifferentiated cauliflower budtissue is unusual in that it contains a high proportion of violaxanthinand a relatively low level of ß-carotene and lutein.The carotenoids are rapidly oxidized in damaged or disruptedtissues but may be protected by the use of an antioxidant. The three usual stem types of Phragmites communis Trin. canbe arranged in a maturation sequence. The horizontal rhizomeis the juvenile type, responsible for renewing the stand; thevertical rhizome is intermediate and bears the aerial shoots;and the aerial shoot is the mature type, photosynthesizing,potentially flowering, and short lived, but not normally capableof renewing the population. 1. Sediment and nutrient loading in freshwater systems are leading causes of aquatic habitat degradation globally. We investigated the impacts of fine-sediment and nutrient additions on the growth and survival of western toad ( Bufo boreas ) tadpoles and emergent metamorphs in mesocosm and exclosure experiments.
2. Mesocosm tanks received weekly pulses of fine sediments to create initial concentrations of 0, 130 and 260 mg L−1 of suspended sediment and either bi-weekly additions of nutrients (N = 160 μg L−1, P = 10 μg L−1) or no additions in a factorial design. Within mesocosms, tadpole exclosures allowed for quantification of tadpole grazing pressure on periphyton biomass, chlorophyll- a and sediment deposition.
3. Tadpoles receiving sediment additions experienced slower growth rates and reduced survival to metamorphosis, although no effects of treatment were detected on size at metamorphosis or time to metamorphosis. Nutrient additions also lowered survival, but had no impact on other measured parameters of tadpole fitness. Dissections and gut content analysis revealed that tadpoles ingested sediment in large quantities altering the proportion of the organic content of ingested food.
4. Together these results suggest that although sediment was readily consumed by tadpoles, its presence in the larval environment had an overall negative effect on tadpole growth and survival, although not as severe as predicted.  相似文献

Although the administration of melanocyte-stimulating hormone (MSH) peptides results in skin darkening in man, cultured human melanocytes have been reported to be unresponsive to these peptides. This may be a consequence of the conditions under which the cells were maintained in vitro, particularly the use of phorbol esters and cholera toxin as melanocyte mitogens. By culturing the cells in the absence of these additives, we demonstrate that α-MSH and its synthetic analogue Nle4DPhe7α-MSH (NDP-MSH) induce dose-related increases in melanin content and tyrosinase activity and affect cell morphology in the majority of human melanocyte cultures. In addition, NDP-MSH induces increases in tyrosinase mRNA and tyrosinase-related protein-1 (TRP-1) mRNA. The dose-response curves for the MSH peptides are sigmoidal and the two peptides are equipotent in their effects on human melanocytes. Adrenocorticotropic hormone (ACTH) also affects morphology and stimulates melanogenesis and tyrosinase activity in human melanocytes. However, the dose-response curves for ACTH are biphasic, and the melanocytes respond to lower concentrations of ACTH than MSH peptides, similar to those normally present in human plasma. These findings may be important in understanding the role of these pro-opiomelanocortin peptides in human skin pigmentation.  相似文献

Continuous measurements of CO2-release from intact roots of Lolium multiflorum growing in nutrient solution were carried out during 3–7 weeks. Periods of days with high level of irradiance and periods with low level alternated. Root respiration rate was found to depend on photosynthesis. The change in root respiration, induced by change in photosynthesis, was delayed. The root respiration rate showed diurnal fluctuations with two characteristic peaks occurring 4–6 and 14–16 hours after onset of the photoperiod. The amplitudes increased with increasing photosynthesis. The frequencies were independent of the length of photoperiod, when this varied between 8 and 16 hours. The fluctuations are discussed in relation to diurnal fluctuations in protein synthesis.  相似文献

Kinetic studies of a dithiothreitol treated membrane ATPase fraction from sugar beet roots led to the following conclusions: 1) In the presence of MgATP, Na+ and K+ stimulate the ATPase activity in different ways following simple Michaelis-Menten kinetics. Thus separate sites for Na+ and K+ are suggested. 2) In the absence of K+, Na+ acts as an uncompetitive modifier raising the apparent Km and Vmax for MgATP. 3) In the absence of Na+, K+ activates non-competitively with respect to MgATP. Thus K+ increases Vmax but does not affect the apparent affinity constant. 4) K+ and Na+ double the rate constants. 5) In the presence of Na+ or K+, Mg2+ in excess acts as a weak inhibitor to Na+ and/or K+ activity. 6) The temperature-activity dependence in the 5–40°C interval shows biphasic Arrhenius plots with the transition point between 15–18°C. The activation energy is lowered at temperatures > 18°C.  相似文献
