Field trials were done with four cultivars over 3 years to assess the extent to which the amount of late blight on the foliage of a potato plant could be influenced by that on a neighbouring plant of the same or a different cultivar. Drills containing the test plants were interspersed with those of spreader plants (cv. King Edward) which were artificially inoculated with Phytophthora infestans. The intensity of blight on the test plants was recorded on several occasions.
Resistant cultivars tended to be scored as less resistant in mixtures with other cultivars than in pure stands, and susceptible cultivars tended likewise to be scored as more resistant in mixed stands. However, standard analysis of variance indicated no systematic evidence of a significant effect due to neighbour cultivars, nor of interaction between cultivars and neighbour cultivars. In contrast, Kempton's (1982) neighbour model indicated a significant and positive interference coefficient (β) in each trial, which generally decreased over time. Predicted pure stand scores for each cultivar indicated that the adjustment was greatest for the most resistant and most susceptible cultivars. There was no advantage in using two-plant rather than one-plant plots in withstanding neighbour effects.  相似文献

From a survey ot the structure of the skull in rhipidistianfishes and early labylinthodont Amphibia and of the mechanismof hearing in these two groups, an account of the evolutionof the tetrapod middle ear is presented. Background

Angiogenesis is widely investigated in conjunction with cancer development, in particular because of the possibility of early stage detection and of new therapeutic strategies. However, such studies are negatively affected by the limitations of imaging techniques in the detection of microscopic blood vessels (diameter 3-5 ??m) grown under angiogenic stress. We report that synchrotron-based X-ray imaging techniques with very high spatial resolution can overcome this obstacle, provided that suitable contrast agents are used.

Results

We tested different contrast agents based on gold nanoparticles (AuNPs) for the detection of cancer-related angiogenesis by synchrotron microradiology, microtomography and high resolution X-ray microscopy. Among them only bare-AuNPs in conjunction with heparin injection provided sufficient contrast to allow in vivo detection of small capillary species (the smallest measured lumen diameters were 3-5 ??m). The detected vessel density was 3-7 times higher than with other nanoparticles. We also found that bare-AuNPs with heparin allows detecting symptoms of local extravascular nanoparticle diffusion in tumor areas where capillary leakage appeared.

Conclusions

Although high-Z AuNPs are natural candidates as radiology contrast agents, their success is not guaranteed, in particular when targeting very small blood vessels in tumor-related angiography. We found that AuNPs injected with heparin produced the contrast level needed to reveal--for the first time by X-ray imaging--tumor microvessels with 3-5 ??m diameter as well as extravascular diffusion due to basal membrane defenestration. These results open the interesting possibility of functional imaging of the tumor microvasculature, of its development and organization, as well as of the effects of anti-angiogenic drugs.  相似文献

The freshwater phreatoicidean isopod Mesamphisopus capensis has been regarded as the most widespread of the four Mesamphisopus species occurring in the Western Cape, South Africa. To determine whether this species was monotypic across its distribution over two mountainous regions, separated by a low-lying coastal plain remnant, genetic differentiation among populations from 11 localities was studied through allozyme electrophoresis of 12 loci and sequencing of a 338-bp 12S rRNA mtDNA fragment from representative individuals. Populations of the two regions were separated by a mean identity value of 0.477. Fixed allele differences at two loci distinguished these regions. Estimates of θ indicated substantial differentiation among populations across the entire sample, as well as within each of the regions. Topologies derived through parsimony and neighbour joining supported the monophyly of the two regions. On the basis of these topologies, allele frequencies and an allozyme dendrogram, five groups were identified. Discriminant function analyses, performed on body and pereopod variables independently, revealed these groups to be well differentiated with a high rate of correct a posteriori reclassification. Using genetic distance criteria these five distinct forms may be considered to be putative species. Background

In order to understand the role of herbivores in trophic webs, it is essential to know what they feed on. Diet analysis is, however, a challenge in many small herbivores with a secretive life style. In this paper, we compare novel (high-throughput pyrosequencing) DNA barcoding technology for plant mixture with traditional microhistological method. We analysed stomach contents of two ecologically important subarctic vole species, Microtus oeconomus and Myodes rufocanus, with the two methods. DNA barcoding was conducted using the P6-loop of the chloroplast trnL (UAA) intron.

Results

Although the identified plant taxa in the diets matched relatively well between the two methods, DNA barcoding gave by far taxonomically more detailed results. Quantitative comparison of results was difficult, mainly due to low taxonomic resolution of the microhistological method, which also in part explained discrepancies between the methods. Other discrepancies were likely due to biases mostly in the microhistological analysis.

Conclusion

We conclude that DNA barcoding opens up for new possibilities in the study of plant-herbivore interactions, giving a detailed and relatively unbiased picture of food utilization of herbivores.  相似文献

Modifications of a domestic freezer are described, which convert it to a versatile controlled environment chamber capable of maintaining temperatures between ambient and - 18 °C and therefore useful in investigations both of plant growth at low positive temperatures and of survival of sub-zero temperatures. The temperature control mechanism can provide a wide range of diurnal temperature regimes and of cooling and warming rates. Measurements in two such chambers showed air temperatures were always uniform to within ° 1 °C of set temperature except in a 6 cm band round the inside walls, where they were lower by 0°6 °C, at most; CO2 concentration was always close to 300 v.p.m.; relative humidity was usually in the range 80–90% and photon flux densities during the light period were 180 ° 10 μmol m-2 s-1. The uniformity of environmental conditions within each chamber was confirmed by measurements of leaf growth and plant survival of tillers of a single genotype of perennial ryegrass growing in pots of compost. These tests also confirmed the lower temperature of the 6 cm band adjacent to the chamber walls. Although temperature settings were identical, air and compost temperatures were 0°3 °C higher in one of the two chambers tested. Although this difference did not result in significant differences in plant survival, significant differences in leaf lamina extension were found between cabinets. Problems encountered in the design and testing of the chambers are discussed.  相似文献
