Genetic data are useful for estimating the genealogical relationship or relatedness between individuals of unknown ancestry. We present a computer program, ml ‐relate that calculates maximum likelihood estimates of relatedness and relationship. ml ‐relate is designed for microsatellite data and can accommodate null alleles. It uses simulation to determine which relationships are consistent with genotype data and to compare putative relationships with alternatives. ml ‐relate runs on the Microsoft Windows operating system and is available from http://www.montana.edu/kalinowski .  相似文献

A few days of thermal acclimation (to 5 °C versus 15 °C) may strongly affect tolerance to drought stress in Collembola. To better understand this phenomenon, the effect of acclimation on water loss rate and its consequence for survival in the species Pogonognathellus flavescens Tullberg (Tomoceridae) is investigated. Acclimation does not affect the water content of hydrated animals but animals exposed to 15 °C and 76% relative humidity lose water much faster after having been acclimated to 5 °C rather than 15 °C. Tolerance to water loss is not affected; in both treatment groups, animals survive up to 40% loss of the water content recorded when fully hydrated. The percentage water content of hydrated animals decreases with size, which may explain why the proportion of initial water lost appears to be a better predictor for survival than the amount of remaining water. The proportion of initial water lost per unit time is little influenced by size in animals acclimated to 15 °C but increases with decreasing size in the group at 5 °C, indicating that acclimation affects a physiological protection against water loss.  相似文献

Carbon flux in arid and semiarid area shrublands, especially in old‐growth shrub ecosystems, has been rarely studied using eddy covariance techniques. In this study, eddy covariance measurements over a 100‐year old‐growth chamise‐dominated chaparral shrub ecosystem were conducted for 7 years from 1996 to 2003. A carbon sink, from −96 to −155 g C m−2 yr−1, was determined under normal weather conditions, while a weak sink of −18 g C m−2 yr−1 and a strong source of 207 g C m−2 yr−1 were observed as a consequence of a severe drought. The annual sink strength of carbon in the 7‐year measurement period was −52 g C m−2 yr−1. The results from our study indicate that, in contrast to previous thought, the old‐growth chaparral shrub ecosystem can be a significant sink of carbon under normal weather conditions and, therefore, be an important component of the global carbon budget.  相似文献

When woody plant abundance increases in grasslands and savannas, a phenomenon widely observed worldwide, there is considerable uncertainty as to whether aboveground net primary productivity (ANPP) and ecosystem carbon (C) and nitrogen (N) pools increase, decrease, or remain the same. We estimated ANPP and C and N pools in aboveground vegetation and surface soils on shallow clay and clay loam soils undergoing encroachment by Prosopis glandulosa in the Southern Great Plains of the United States. Aboveground Prosopis C and N mass increased linearly, and ANPP increased logarithmically, with stand age on clay loam soils; on shallow clays, Prosopis C and N mass and ANPP all increased linearly with stand age. We found no evidence of an asymptote in trajectories of C and N accumulation or ANPP on either soil type even following 68 years of stand development. Production and accumulation rates were lower on shallow clay sites relative to clay loam sites, suggesting strong edaphic control of C and N accumulation associated with woody plant encroachment. Response of herbaceous C mass to Prosopis stand development also differed between soil types. Herbaceous C declined with increasing aboveground Prosopis C on clay loams, but increased with increasing Prosopis C on shallow clays. Total ANPP (Prosopis+herbaceous) of sites with the highest Prosopis basal area were 1.2 × and 4.0 × greater than those with the lowest Prosopis basal area on clay loam and shallow clay soils, respectively. Prosopis ANPP more than offset declines in herbaceous ANPP on clay loams and added to increased herbaceous ANPP on shallow clays. Although aboveground C and N pools increased substantially with Prosopis stand development, we found no corresponding change in surface soil C and N pools (0–10 cm). Overall, our findings indicate that Prosopis stand development significantly increases ecosystem C and N storage/cycling, and the magnitude of these impacts varied with stand age, soil type and functional plant traits  相似文献

ABSTRACT. Cell division in higher eukaryotes is mainly controlled by p34 cdc2 or related kinases and by other components of these kinase complexes. We present evidence that cdc2 -like kinases also occur in Paramecium. Two polypeptides reacted with an antibody directed against the perfectly conserved PSTAIR region found in cdc2 kinases in other eukaryotes. Only the less abundant peptide bound to p13 suc1 from Schizosaccharomyces pombe. Using centrifugal elutriation to select cells on the basis of size, we isolated highly synchronous Paramecium G1 cells. With this procedure, we demonstrated that the p13suc1-associated cdc2 -like histone H1 kinase was activated before cell division at the point of commitment to division in Paramecium. Further, we show that Paramecium cdc2 -like proteins occurred principally as monomers and that these monomers were active as histone H1 kinases in vitro.  相似文献

Feather melanocytes in the Barred Plymouth Rock (BPR) and White Leghorn (WL) chickens die prematurely in vivo when compared to the wild type Jungle Fowl (JF) chicken. Since these mutant melanocytes live in vitro, an environmental factor in the feather must precipitate their death. Results show that the addition of selected antioxidants, glutathione (GSH) and superoxide dismutase (SOD), can rescue these mutant melanocytes in vitro that have been placed under stress conditions that cause their premature cell death. Measurements of in vivo levels of GSH, catalase, and SOD show no significant difference in catalase activity between the JF, BPR, and WL feathers but do show a significant reduction in GSH activity in both the BPR and WL feathers to approximately 66% of the GSH concentration found in JF feathers. SOD activity in the BPR tissue is reduced significantly to approximately 50% of the JF activity and the WL SOD activity is reduced significantly to approximately 50% of the BPR SOD activity. Preliminary results of measurements of glutathione peroxidase activity indicate there is no difference in the levels of this enzyme in JF, BPR and WL feathers. A working hypothesis, based on current results, is proposed for premature cell death in BPR and WL feather melanocytes. The BPR melanocytes are genetically sensitive due to a defect in their SOD and GSH levels caused by the barring gene (B) and their death, due to reactive species of oxygen radicals, is precipitated in the poorly vascularized feather by the accumulation of oxygen radicals due to the low turnover of tissue fluids. The WL chicken carries the dominant white gene (I) in addition to the B gene. This gene directs the further reduction of the level of SOD and, when combined with the cell death mechanism already present in the BPR chicken, causes the WL feather melanocytes to die much earlier than the BPR feather melanocytes which in turn die much earlier than the wild type JF melanocytes. This same mechanistic hypothesis could apply as a cause of premature melanocyte cell death in human vitiligo wherein the vitiliginous melanocytes may have a genetic defect in their oxygen radical protection system.  相似文献

SYNOPSIS. The structure and morphogenesis of the ventral ciliature of Paraurostyla hymenophora (Stokes) are described. The oral primordium apparently originates in association with transverse cirrus #6, from which it migrates anteriorly simultaneous with kinetosomal proliferation. The primordium eventually forms an elongate ciliary field from which the future opisthe's fronto-ventro-transverse (FVT) and undulating membrane primordial fields arise. Concomitantly, the future proter's FVT primordial field is initiated by the disaggregation of frontal cirri #4, #5, and #6. Primordia then develop simultaneously within marginal and ventral cirral rows by a disaggregation of cirri within the respective rows, and do not give rise to new cirri until the FVT fields complete segregation into discrete cirri. Near the completion of cirral production from the FVT primordia, each ventral cirral primordium (VCP) forms the 2 rightmost transverse cirri. Segregation of new cirri within the marginal cirral primordia and VCP then occurs, eventually replacing all old cirri within their respective marginal and ventral cirral rows. At the end of cortical morphogenesis, all old ciliary organelles, with the exception of the adoral zone of membranelles, are either reorganized or replaced. These results suggest an evolutionary affinity between the ventral and marginal cirral rows and raise questions about the control of the developmental competence of individual primordia.  相似文献

Abstract. 1. We have explored the question of whether breadth of diet in moths is associated with adaptations other than host selection behaviour, with particular reference to body size.
2. Generalist species in Britain and New York are larger than specialist species.
3. This relationship is probably not due to latitudinal clines, nor to higher fecundity in generalist species.
4. We suggest that the large bodies of generalists buffer them against environmentally-induced physiological stress.  相似文献
