Muscle tissue from 63 Synodontis zambezensis collected bimonthly in 2013 at Flag Boshielo Dam were analysed for metals and metalloids in a desktop human health risk assessment. The Hazard Quotient, based on a weekly meal of 67 g of fish muscle, exceeded the maximum acceptable level of one for lead, cobalt, cadmium, mercury, arsenic and selenium. The concentrations of these elements were higher in 2013 than those recorded in 2009 and 2012 in other fish species from Flag Boshielo Dam and these may pose a long-term health risk if consumed regularly by impoverished rural communities reliant on fish as a source of protein. 相似文献

In order to diversify the production of plants with pharmacological interest, it is important to understand the mechanisms
involved in their tolerance to environmental constraints, such as salinity. Basil (Ocimum basilicum), known for its therapeutic uses, has been claimed to be salt tolerant, but physiological aspects of this behavior remain
unknown. Since salt tolerance is known to be associated with several characteristics concerning Na+ transport to leaves, we studied this function in hydroponically grown basil. We analyzed the response of 30-day-old seedlings
to 25–50 mM NaCl applied for 15 days. Growth was poorly affected, indicating that these concentrations corresponded to the
tolerated salinity range. Leaves accumulated Na+ at relatively high concentration, without dehydrating. Potassium concentration in leaf tissues was maintained close to control
level, indicating that K+ was 15- to 25-fold preferred over Na+ for ion transport and deposition. Collection of xylem sap on detopped plants revealed that this preference was only 10-fold
for ion introduction into root xylem sap. Short-term (24 h) changes in Na+ distribution between organs after stem (steam) girdling suggested that Na+ downward recirculation by phloem occurred. Although modest, this transport might have augmented K+ selectivity of ion deposition in leaves. 相似文献

organisms, including Gram-positive, Gram-negative bacteria, fungi and microalgae using the disk diffusion bioassay method.
Additionally a cytotoxicity test was performed by means of the brine shrimp microwell cytotoxicity assay. In a chemical screening
each of the crude extracts was analysed by TLC using various staining reagents and by HPLC-MS/MS measurements. The results
depicted an impressive chemical diversity of crude extracts produced by these strains. The taxonomic status of the selected

Background
Laboratory conditions nullify the extrinsic factors that determine the wild expected lifespan and release the intrinsic or potential lifespan. Thus, wild animals reared in a laboratory often show an increased lifespan, and consequently an increased senescence phase. Senescence is associated with a broad suite of physiological changes, including a decreased responsiveness of the circadian system. The time-keeping hormone melatonin, an important chemical player in this system, is suspected to have an anti-aging role. The Greater White-toothed shrew Crocidura russula is an ideal study model to address questions related to aging and associated changes in biological functions: its lifespan is short and is substantially increased in captivity; daily and seasonal rhythms, while very marked the first year of life, are dramatically altered during the senescence process which starts during the second year. Here we report on an investigation of the effects of melatonin administration on locomotor activity of aging shrews.Methodology/Principal Findings
1) The diel fluctuations of melatonin levels in young, adult and aging shrews were quantified in the pineal gland and plasma. In both, a marked diel rhythm (low diurnal concentration; high nocturnal concentration) was present in young animals but then decreased in adults, and, as a result of a loss in the nocturnal production, was absent in old animals. 2) Daily locomotor activity rhythm was monitored in pre-senescent animals that had received either a subcutaneous melatonin implant, an empty implant or no implant at all. In non-implanted and sham-implanted shrews, the rhythm was well marked in adults. A marked degradation in both period and amplitude, however, started after the age of 14–16 months. This pattern was considerably delayed in melatonin-implanted shrews who maintained the daily rhythm for significantly longer.Conclusions
This is the first long term study (>500 days observation of the same individuals) that investigates the effects of continuous melatonin delivery. As such, it sheds new light on the putative anti-aging role of melatonin by demonstrating that continuous melatonin administration delays the onset of senescence. The present research was undertaken to evaluate some biological parameters in rats fed with a supplemented diet with Opuntia ficus indica powder seeds. Feed intake and body weight of rats were measured every two days during nine weeks of treatment. Digestibility, feed conversion efficiency and protein efficiency ratio were determined. No difference in digestibility was noticed between the different diets. The results indicated a significant decrease in body weight of rats receiving a diet partially substituted with O. ficus indica powder seeds, probably due to a significant decrease in serum-free thyroxin (FT(4)) compared to the control group. In the treated group, a decrease of glucose concentration in blood and an increase of glycogen in liver and skeletal muscle were noticed. A significant increase in HDL-cholesterol was noted in the group receiving the supplemented diet with O. ficus indica powder seeds. These results suggest that O. ficus indica seeds can be used as a healthy food. 相似文献

Photoperiod and thermosensitive genetic male sterile (PTGMS) lines have become one of the main sources of global rice production increasing. This study was conducted to evaluate the fertility alteration and validate the male sterility genes using validation markers in novel Egyptian Indica and Japonica PTGMS lines under natural conditions. The study revealed that the new genetic male sterile lines belong to the type of photo–thermosensitive genetic male sterility (PTGMS). The fertility alteration of these lines has influenced by photoperiod and temperature interaction. The new PTGMS lines have three sensitive periods of fertility alteration; transformation, sterility, and fertility period. Furthermore, the sensitive stage of fertility transformation might be from secondary branch primordial to pollen mother cells (PMC) meiosis. Under the natural Sakha condition, the new PTGMS lines were stable sterile under the condition of day length upper 13,75 h and temperature over 25 °C, while its convert to fertile under day length under 13 h, and temperature lower than 24 °C. The co-dominant markers identified the pms3 and tms5 genes in the new PTGMS lines, indicated that the fertility alteration in these lines controlled by photoperiod and thermosensitive stages. 相似文献

MurAs (enolpyruvyl-UDP-GlcNAc synthases) from pathogenic bacteria such as Borrelia burgdorferi (Lyme disease) and tuberculosis are fosfomycin resistant because an Asp-for-Cys substitution prevents them from being alkylated by this epoxide antibiotic. Previous attempts to characterize naturally Asp-containing MurAs have resulted in no protein or no activity. We have expressed and characterized His-tagged Lyme disease MurA (Bb_MurA(H6)). The protein was most soluble at high salt concentrations but maximally active around physiological ionic strength. The steady-state kinetic parameters at pH 7 were k(cat) = 1.07 ± 0.03 s(-1), K(M,PEP) = 89 ± 12 μM, and K(M,UDP-GlcNAc) = 45 ± 7 μM. Mutating the active site Asp to Cys, D116C, caused a 21-fold decrease in k(cat) and rendered the enzyme fosfomycin sensitive. The pH profile of k(cat) was bell-shaped and centered around pH 5.3 for Bb_MurA(H6), with pK(a1) = 3.8 ± 0.2 and pK(a2) = 7.4 ± 0.2. There was little change in pK(a1) with the D116C mutant, 3.5 ± 0.3, but pK(a2) shifted to >11. This demonstrated that the pK(a2) of 7.4 was due to D116, almost 3 pH units above an unperturbed carboxylate, and that it must be protonated for activity. This supports D116's proposed role as a general acid/base catalyst. As fosfomycin does not react with simple thiols, nor most protein thiols, the reactivity of D116C with fosfomycin, combined with the strongly perturbed pK(a2) for D116, strongly implies an unusual active site environment and a chemical role in catalysis for Asp/Cys. There is also good evidence for C115 having a role in product release. Both roles may be operative for both Asp- and Cys-containing MurAs. 相似文献

Basil (Ocimum basilicum L.) seedlings were cultured on liquid medium in controlled conditions. Two varieties differing in leaf size were compared. When plants were 30?days old, the medium was supplemented with 50?mM NaCl. After 15?days of treatment, root, stem and leaf biomass, leaf number, and leaf surface area were measured. Ion accumulation was determined in roots, stems, and leaves. Photosynthetic parameters (CO2 fixation rate, internal CO2 concentration, stomatal conductance) as well as transpiration rate were determined on separate leaves. Electrolyte leakage and malondialdehyde content were used to estimate damage to membranes and lipid peroxidation, respectively. Several antioxidant enzymatic activities were used as proxies of oxidative stress. High Na+ concentration was reached in leaf tissues. Salt restricted whole plant biomass deposition rate by diminishing leaf number and leaf expansion, as well as photosynthetic activity were estimated from whole plant biomass production per unit leaf surface area. Diminished stomatal conductance restricted CO2 fixation rate, and decrease in chlorophyll content presumably limited photosynthetic activity. Lipid peroxidation revealed damages to membranes. The magnitude of these responses differed between the two varieties, indicating that an intraspecific variability in salt response exists in basil. 相似文献
