Hyaluronic acid (HA) has great importance in biomedical applications. In this work, a novel nanoparticle-based method that stimulates the hyaluronic acid (HA) production by the bacteria Streptococcus equi subsp. Zooepidemicus has been reported. CNTs with diameters of 40–50 nm and lengths of about 20 mm were used at four different concentrations (0, 10, 25, 50, and 100 μg) to the bacteria and determined the mass of the produced HA in dependence on the exposure time under UV-irradiation. The results clearly showed that the exposure for one minute with low power UV light (254 nm) and 100 µg (CNTs) treatments steadily increased HA production from the control (0.062 g/L) to the highest value (0.992) g/L of HA. The incubation of the streptococci with CNTs led to an increase of the HA production by a factor of 4.23 after 300S exposure time under UV light, whereas the HA production was no significant enhancement under visible light. It is explained that the CNTs nanoparticle-stimulated increase of the HA production with the internalization of the nanoparticles by the bacteria since they “serve as co-enzymes” under induced mutation by UV-irradiation. Transformation process was carried out and showed that the major protein band of Streptococcus equi was observed in the Streptococcus DH5α. RAPD analysis indicates that the amplified DNA fragments and the percentage of polymorphism was similar between Streptococcus equi and Streptococcus DH50α. The chemical structure and molecular weight of the photoproduced HA from Streptococcus equi was similar to the chemical structure of the standard sample. 相似文献

Caper plant (Capparis spinosa) extracts have been associated with diverse biological activities including anti-oxidant properties. In this work, we characterized the hydro-ethanolic extract obtained from C. spinosa leaves [hydroethanolic extract of C. spinosa (HECS)] by analyzing the content in anti-oxidant compounds such as polyphenols, flavonoids and anthocyanins. Further, we evaluated HECS antioxidant activities in vitro using bleaching of 1,1-diphenyl-2-picrylhydrazyl radical and ABTS test as well as by pretreatment of HeLa cells exposed to Fe2+ or H2O2. Our findings indicate that HECS contains high amount of total phenolic compounds and high levels of flavonoids and anthocyanins. Furthermore, HECS exhibited antioxidant activity in both chemical and biological tests. Specially, pretreatment of HeLa cells with different concentrations of the extract conferred protection against lipid peroxidation and modulated activities of two antioxidant enzymes, SOD and catalase. These results revealed HECS antioxidant effects and suggest that C. spinosa leaves are a potential source of natural antioxidant molecules with possible applications in industry and medicine. 相似文献

The effect of organochlorine insecticides lindane (1,2,3,4,5,6-hexachlorocyclohexane) and DDT (1,1,1-trichloro-2,2-bis (p-chlorophenyl)ethane) were studied in terms of their effects on the rat pineal N-acetyltransferase (NAT) activity, hydroxyindole-O-methyltransferase (HIOMT) activity and pineal and serum melatonin levels during the day (2000h) and at night (2300 and 0100h). Additionally, pineal levels of 5-hydroxytryptophan (5-HTP), serotonin (5-HT), and 5-hydroxyindole acetic acid (5-HIAA) were estimated. Nocturnal NAT activity was increased after lindane administration; likewise, lindane augmented pineal and serum melatonin levels at 2300h. Conversely, DDT was without a statistically significant effect on either NAT activity or on pineal or serum melatonin levels. Neither lindane nor DDT significantly influenced pineal HIOMT values either during the day or at night. Likewise, neither insecticide consistently influenced pineal levels of either 5-HTP, 5-HT or 5-HIAA. The results indicate that the organochlorine insecticide, lindane, modifies pineal melatonin synthesis in vivo. 相似文献

RNA recognition motifs as important regulators of gene expression are highly conserved in animals and plants. The FCA floral promotion gene in Arabidopsis encodes a protein, containing two RNA recognition motifs (RRM) and a WW protein interaction domain. Here we isolated FCA cDNA from rice. FCA in rice (rFCA) was homologous to FCA-gamma of Arabidopsis and contained conserved domains. To investigate the function of RRM domain, fragment RRM1 and RRM2 of rFCA were introduced into rice subspecies Oryza sativa L. subsp. Indica var. 9311 and another rice subspecies Oryza sativa L. subsp. Japonica var. zhonghua11 transformation. volume of transgenic plants was increased. These results showed that constitutive expression of RRMs could regulate cellular

BackgroundPrevious epidemiological studies have examined the prevalence and risk factors for a variety of parasitic illnesses, including protozoan and soil-transmitted helminth (STH, e.g., hookworms and roundworms) infections. Despite advancements in machine learning for data analysis, the majority of these studies use traditional logistic regression to identify significant risk factors.MethodsIn this study, we used data from a survey of 54 risk factors for intestinal parasitosis in 954 Ethiopian school children. We investigated whether machine learning approaches can supplement traditional logistic regression in identifying intestinal parasite infection risk factors. We used feature selection methods such as InfoGain (IG), ReliefF (ReF), Joint Mutual Information (JMI), and Minimum Redundancy Maximum Relevance (MRMR). Additionally, we predicted children’s parasitic infection status using classifiers such as Logistic Regression (LR), Support Vector Machines (SVM), Random Forests (RF) and XGBoost (XGB), and compared their accuracy and area under the receiver operating characteristic curve (AUROC) scores. For optimal model training, we performed tenfold cross-validation and tuned the classifier hyperparameters. We balanced our dataset using the Synthetic Minority Oversampling (SMOTE) method. Additionally, we used association rule learning to establish a link between risk factors and parasitic infections.Key findingsOur study demonstrated that machine learning could be used in conjunction with logistic regression. Using machine learning, we developed models that accurately predicted four parasitic infections: any parasitic infection at 79.9% accuracy, helminth infection at 84.9%, any STH infection at 95.9%, and protozoan infection at 94.2%. The Random Forests (RF) and Support Vector Machines (SVM) classifiers achieved the highest accuracy when top 20 risk factors were considered using Joint Mutual Information (JMI) or all features were used. The best predictors of infection were socioeconomic, demographic, and hematological characteristics.ConclusionsWe demonstrated that feature selection and association rule learning are useful strategies for detecting risk factors for parasite infection. Additionally, we showed that advanced classifiers might be utilized to predict children’s parasitic infection status. When combined with standard logistic regression models, machine learning techniques can identify novel risk factors and predict infection risk. 相似文献

Trichinellosis is a serious disease with no satisfactory treatment. We aimed to
assess the effect of myrrh (Commiphora molmol) and, for the first
time, thyme (Thymus vulgaris L.) against enteral
and encysted (parenteral) phases of Trichinella spiralis in mice
compared with albendazole, and detect their effect on inducible nitric oxide synthase
(iNOS) expression. Oral administration of 500 mg/kg of myrrh and thyme led to adult
reduction (90.9%, 79.4%), while 1,000 mg/kg led to larvae reduction (79.6%, 71.3%),
respectively. Administration of 50 mg/kg of albendazole resulted in adult and larvae
reduction (94.2%, 90.9%). Positive immunostaining of inflammatory cells infiltrating
intestinal mucosa and submucosa of all treated groups was detected. Myrrh-treated
mice showed the highest iNOS expression followed by albendazole, then thyme. On the
other hand, both myrrh and thyme-treated groups showed stronger iNOS expression of
inflammatory cells infiltrating and surrounding encapsulated T.
spiralis larvae than albendazole treated group. In conclusion, myrrh and
thyme extracts are highly effective against both phases of T.
spiralis and showed strong iNOS expressions, especially myrrh which could
be a promising alternative drug. This experiment provides a basis for further
exploration of this plant by isolation and retesting the active principles of both

In this study, we hypothesized that colonization of olive trees (Olea europaea L.) with the arbuscular mycorrhizal fungus Rhizophagus irregularis could modify the profiles of rhizosphere microbial communities with subsequent effects on nutrient uptake that directly affects olive tree physiology and performance. In this context, a greenhouse experiment was carried out in order to study the effects of mycorrhizal colonization by R. irregularis on photosynthesis, pigment content, carbohydrate profile, and nutrient uptake in olive tree. After six months of growth, photosynthetic rate in mycorrhizal (M) plants was significantly higher than that of nonmycorrhizal plants. A sugar content analysis showed enhanced concentrations of mannitol, fructose, sucrose, raffinose, and trehalose in M roots. We also observed a significant increase in P, K, Ca, Mg, Zn, Fe, and Mn contents in leaves of the M plants. These results are important, since nutrient deficiency often occurs in Mediterranean semiarid ecosystems, where olive trees occupy a major place. 相似文献

Oreochromis aureus was imported from Israel into South Africa in 1959 but data on its current status in South Africa are lacking. Genomic DNA was extracted and the COI gene amplified at the South African Institute for Aquatic Biodiversity. The identity of the sequences and specimens was determined using the Barcode of Life Data Systems and GenBank. Morphological and genetic assessment demonstrated that 11 specimens collected from two farm dams in the Eerste River System, Western Cape province, were Oreochromis aureus. A MaxEnt model compiled using global distribution, rainfall and temperature data predicted that large areas of southern Africa were climatically suitable for this species, indicating considerable invasion debt in southern Africa. As a result, surveys to assess for the extent of the invasion in South Africa and eradication of existing populations, if feasible, are recommended management actions. 相似文献

The present study aimed, for the first time, to examine the biochemical effects of new phthalimide analog, 2-[2-(2-Bromo-1-ethyl-1H-indol-3-yl) ethyl]-1H-isoindole-1,3(2H)-dione, compared to thalidomide drug against liver injury induced in mice. Carbon tetrachloride was intraperitoneal injected in mice for 6 consecutive weeks at a dose of 0.4 mL/kg twice a week for liver injury induction. Histopathological examination, levels of malondialdehyde, nitric oxide, and antioxidant enzymes were determined. Additionally, the protein levels of vascular endothelial growth factor, proliferating cell nuclear protein, tumor necrosis factor-alfa, nuclear factor kappa B-p65, B-cell lymphoma-2, and cysteine-aspartic acid protease-3 were determined. Results revealed that the treatment with phthalimide analog improved the detected liver damage and presented an obvious antioxidant activity through decreasing malondialdehyde and nitric oxide levels accompanied by increasing the levels of the antioxidant enzymes. Furthermore, the analog exhibited an effective inhibitory activity towards the studied protein expressions in liver tissues. Moreover, the B-cell lymphoma-2 protein level was increased while the cysteine-aspartic acid protease-3 level was suppressed after the treatment with phthalimide analog. Together, these results propose that phthalimide analog can ameliorate carbon tetrachloride-induced liver injury in mice through its potent inhibition mediating effect in oxidative stress, inflammation, and apoptosis mechanisms. 相似文献
