In the 'F' horizons of acid mor-humus soils of heathland ecosystems, mycorrhizal roots of the dominant ericaceous species form a large fraction of the soil biomass. Rapid turnover of these roots provides the potential for recycling of substantial amounts of nitrogen contained in their fungal and plant components. Here, we first determine the amount of N in the biomass of ericoid roots growing in heathland and show it to constitute a large proportion of total soil N. In order to assess the accessibility of this N to ericaceous plants, experiments were then conducted using aseptically produced shoot and root necromass of Vaccinium macrocarpon Ait., the roots being grown with or without mycorrhizal colonization. These materials were provided as sole nitrogenous substrates in growth experiments using the ericoid mycorrhizal fungus Hymenoscyphus ericae (Read) Korf & Kernan in pure culture and V. macrocarpon in the mycorrhizal (M) or non-mycorrhizal (NM) condition as test organisms. The experiments were designed to test the hypothesis that the N contained in these substrates can be mobilized by the mycorrhizal endophyte. The ability of the endophyte to utilize the substrates was determined by measuring fungal yields and by assessing the presence of its extra-cellular protease and chitinase enzymes. Transfer of N to the host by the endophyte was determined through measurements of plant yield and tissue N contents. H. ericae produced a significantly greater yield on shoot and mycorrhizal root necromass than on non-mycorrhizal root necromass. The extra-cellular enzymes protease and chitinase were produced by the fungus when grown on the M root necromass. The fungus also transferred N to the host plant, up to 76% of N contained in the substrate being found in M plants whereas less than 5% was present in their NM counterparts.  相似文献

This work presents a cooperative effort to integrate new molecular (isozyme and SSU analyses) characters into the morphological taxonomy of the genus Gigaspora (Glomales). Previous analyses of published Gigaspora SSU sequences indicated the presence of a few polymorphic nucleotides in the region delimited by primers NS71-SSU 1492'. In our study, the SSU of 24 isolates of arbuscular mycorrhizal (AM) fungi from the Gigasporaceae were amplified and the NS71-SSU 1492' region was directly sequenced. The corresponding sequences of four more isolates of AM fungi from Gigasporaceae, already published, were also included in our analyses. Three Gigaspora groups were identified on the basis of a 6 nucleotide-long 'molecular signature': Gigaspora rosea group ( G. rosea + G. albida ), Gigaspora margarita group ( G. margarita + G. decipiens ) and Gigaspora gigantea , which constituted a group by itself. The isozyme profiles (malate dehydrogenase, MDH) of 12 of these 28 isolates, and seven other isolates not sequenced, were compared. The results obtained further supported the grouping of isolates provided by the SSU analysis. Both SSU and MDH analysis indicated that two out of the 35 isolates had been misidentified, which was confirmed when their morphology was reassessed. The use of the Gigaspora intrageneric molecular signature as a quick, unambiguous and objective method to recognize Gigaspora isolates under any (field or laboratory) experimental conditions is suggested.  相似文献

The composition and amount of carotenoid pigments were determined in etiolated seedling leaves of 6 barley (Hordeum vulgare L.) mutants, comprising 1 xantha and 5 tigrina mutants. All mutants had on a mole basis approximately the same content of carotenoids as the wild type. The mutants xan-u21, tig-n32, and tig-33 contained significantly higher amounts of carotenes than the wild type, ranging from 32 to 68% of the total carotenoid content as compared to the 4–8% found in the wild type. In the mutants tig-b23 and tig-o34, only a slight increase in the amount of carotenes was notable. The carotene content and composition in tig-d12 was indistinguishable from that of the wild type. The carotenes extracted from xan-u21, tig-b23, tig-n32, tig-33, and tig-o34 were characterized by adsorption chromatography and spectrophotometry. Mutant xan-u21 is in the dark blocked in β-carotene synthesis, and accumulates the aliphatic polyenes: phytofluene, proneurosporene, poly-cis-lycopenes, neo-lycopene and lycopene. The other four mutants synthesize β-carotene, but accumulate in addition various higher saturated carotenes, the main components being ζ-carotene in tig-b23, a lycopenic pigment in tig-n32 and tig-33, and lycopene in tig-o34. Accumulation of higher saturated carotenes appears correlated with specific aberrations of the membrane structure in plastids. The regulation of carotene and protochlorophyllide syntheses in etioplasts are closely linked as shown by the single gene mutants which affect both pathways. However, several mutants have been identified which cause defects in protochlorophyllide synthesis only.  相似文献

It has previously been shown that the concentration of 3:5-dinitro-ocresolrequired in the external medium to halve (a) the growth rateof the mould Trichoderma viride or (b) the respiration rateof yeast increased same 75–100 times as the pH changedfrom 4 to 7. In the present investigation it has been foundthat when solutions or suspensions of 3:5-dinitro-o-cresol areapplied as sprays to seedlings of Brassica alba the concentrationneeded to kill half the plants changes by a factor of less than2 over the same pH range. On the other hand, when Lemma minor is grown in nutrient solutioncontaining the toxicant, the concentration required to halvethe rate of frond multiplication increases sharply as the pHis raised from 5.4 to 6.8. Moreover, when leaf disks of B. albaare vacuum infiltrated with buffered solutions of 3:5-dinitro-o-cresolthe concentration needed to halve the respiration rate rises50-fold between pH 4 and 8. It is considered that the essential condition which determinesthe similarity of the result for L. minor, the leaf disks, andmicro-organisms is that a relatively large volume of solutioncontaining the toxicant is in direct contact with the tissues.In contrast, under the conditions of spraying the volume ofspray droplets relative to that of plant tissue is small. Thusthe pH effect ie masked since the buffering capacity of thecells exceeds that of the droplets, with the result that 3:5-dinitro-o-cresollargely acts upon the cells at a pH determined by the tissues,a pH which may bear little relation to that of the originalsolution. Supporting evidence is produced that the pH factor operatesin the same way for other weak acids, such as the chlorophenoxyaceticacids.  相似文献

Survey‐gap analysis in expeditionary research: where do we go from here?   总被引:2,自引:0,他引:2  
Research expeditions into remote areas to collect biological specimens provide vital information for understanding biodiversity. However, major expeditions to little‐known areas are expensive and time consuming, time is short, and well‐trained people are difficult to find. In addition, processing the collections and obtaining accurate identifications takes time and money. In order to get the maximum return for the investment, we need to determine the location of the collecting expeditions carefully. In this study we used environmental variables and information on existing collecting localities to help determine the sites of future expeditions. Results from other studies were used to aid in the selection of the environmental variables, including variables relating to temperature, rainfall, lithology and distance between sites. A survey gap analysis tool based on ‘ED complementarity’ was employed to select the sites that would most likely contribute the most new taxa. The tool does not evaluate how well collected a previously visited site survey site might be; however, collecting effort was estimated based on species accumulation curves. We used the number of collections and/or number of species at each collecting site to eliminate those we deemed poorly collected. Plants, birds, and insects from Guyana were examined using the survey gap analysis tool, and sites for future collecting expeditions were determined. The south‐east section of Guyana had virtually no collecting information available. It has been inaccessible for many years for political reasons and as a result, eight of the first ten sites selected were in that area. In order to evaluate the remainder of the country, and because there are no immediate plans by the Government of Guyana to open that area to exploration, that section of the country was not included in the remainder of the study. The range of the ED complementarity values dropped sharply after the first ten sites were selected. For plants, the group for which we had the most records, areas selected included several localities in the Pakaraima Mountains, the border with the south‐east, and one site in the north‐west. For birds, a moderately collected group, the strongest need was in the north‐west followed by the east. Insects had the smallest data set and the largest range of ED complementarity values; the results gave strong emphasis to the southern parts of the country, but most of the locations appeared to be equidistant from one another, most likely because of insufficient data. Results demonstrate that the use of a survey gap analysis tool designed to solve a locational problem using continuous environmental data can help maximize our resources for gathering new information on biodiversity. © 2005 The Linnean Society of London, Biological Journal of the Linnean Society, 2005, 85 , 549–567.  相似文献

Abstract Biodiversity estimates are typically a function of sampling effort and in this regard it is important to develop an understanding of taxon‐specific sampling requirements. Northern hemisphere studies have shown that estimates of riverine fish diversity are related to sampling effort, but such studies are lacking in the southern hemisphere. We used a dataset obtained from boat electro‐fishing the fish community along an essentially continuous 13‐km reach of the Murrumbidgee River, Australia, to investigate sampling effort effects on fish diversity estimates. This represents the first attempt to investigate relationships between sampling effort and the detection of fish species in a large lowland river in Australia. Seven species were recorded. Species‐specific patterns in catch per unit effort were evident and are discussed in terms of solitary and gregarious species, recreational fishing and the monitoring of rare and threatened species. There was a requirement to sample substantial lengths of river to describe total species richness of the fish community in this river reach. To this end, randomly allocated sampling effort and use of species richness estimators produced accurate estimates of species richness without the requirement for excessive levels of effort. Twenty operations were required to estimate species richness at this site, highlighting the need for comparable studies of river fish communities in lowland rivers elsewhere in Australia and the southern hemisphere.  相似文献

Molecular sexing of birds has been possible for over a decade, but for practical reasons many studies still use biometric data for sex discrimination. In some species, the sexes are easy to distinguish but sexual dimorphism is often more subtle, requiring the use of statistical analyses of biometric measurements to discriminate sexes. These models are usually parameterized and validated using data from a limited number of sites and years. However, the resulting discriminant functions are often applied to other populations and periods. A crucial, but usually untested, assumption is that sexual dimorphism does not vary in time and space. Here we illustrate the consequences of violation of this assumption in Eurasian Oystercatchers Haematopus ostralegus , a species for which most studies have relied on biometric sexing. Using biometric data from captures of known-sex birds, we show that sexual dimorphism varied substantially in time and even reversed in some months and years. Furthermore, some biometric traits used in sexing changed gradually over time, causing a reduction in sexual dimorphism. We show that the consequences of this variation on sex discrimination in Oystercatchers are subtle and easily overlooked, but can result in inaccurate and strongly male- or female-biased sex-ratio estimates. We recommend that biometric sexing should be avoided in Oystercatchers unless specific calibration for each month, year and area is carried out. This recommendation also applies to other species where biometric traits may depend on environmental conditions. We argue that this condition might apply to many bird species and therefore advise caution when interpreting results based on biometric sex discrimination.  相似文献

1. Algal growth in lotic systems is controlled either from the bottom‐up (e.g. nutrients and light, which determine growth rates) or from the top‐down (e.g. grazing pressure, which reduces accumulated biomass). Nutrient‐enriched streams that support large and diverse grazing macroinvertebrate populations and those with shaded riparian corridors rarely suffer from excessive algal growth. 2. In this study, the density of benthic algivorous macroinvertebrates was experimentally manipulated in shaded and open nutrient‐enriched stream habitats of the Owennagearagh River, south‐west Ireland. The ability of macroinvertebrate grazers and riparian shade to control benthic algal growth [particularly the nuisance alga, Cladophora glomerata (L. Kütz)] was investigated. Three sites with markedly different concentrations of plant nutrients (one site upstream and two sites downstream of the sewage outfall) were selected. The density of grazing invertebrates colonising ceramic tiles was reduced using high‐voltage localised electric pulses. Replicates of treatment (grazer‐excluded) and control (grazed) tiles were deployed in open and shaded (<25 and >80% canopy cover, respectively) patches of stream bed, in each site. 3. After 2‐week Cladophora cover, periphytic chlorophyll a and biofilm ash‐free dry mass (AFDM) were quantified for all experimental tiles. Values for all three parameters were highest on grazer‐excluded tiles from open patches. Grazed tiles from open patches accrued little Cladophora and had significantly lower levels of chlorophyll a and AFDM. Nutrient inputs were found to have an impact on the density of grazing invertebrates, with higher densities of Baetis nymphs at the most nutrient‐enriched site. 4. Our results demonstrate that in eutrophic, high‐light streams, filamentous algae can quickly accumulate to nuisance levels in the absence of invertebrate grazers. In future, greater attention should be paid to the role of grazing invertebrates in controlling nuisance algae in streams, in addition to algal–nutrient relationships.  相似文献

1. The functional feeding group approach has been widely used to describe the community structure of benthic invertebrates in relation to organic matter resources. Based on this functional framework, positive interactions between feeding groups (especially shredders and collector‐gatherers) were postulated in the River Continuum Concept. However, relationships with organic matter have been poorly documented for invertebrates living in the hyporheic zone. 2. We hypothesised that the common subterranean amphipod Niphargus rhenorhodanensis would feed on fine particulate organic matter (FPOM), which is more abundant than coarse particulate organic matter (CPOM) in hyporheic habitats, and should be favoured by the occurrence of shredders that produce FPOM from CPOM. 3. We used laboratory experiments to quantify leaf litter processing by N. rhenorhodanensis and a common shredder, the surface amphipod Gammarus roeselii. We estimated rates of feeding and assimilation (using nitrogen stable isotopes) of the two species separately and together to reveal any potential shredder–collector facilitation between them. 4. Measured leaf litter mass loss showed that N. rhenorhodanensis did not act as a shredder, unlike G. roeselii. Organic matter dynamics and 15N/14N ratios in tissues of niphargids indicated that N. rhenorhodanensis was a collector‐gatherer feeding preferentially on FPOM. We also found a positive influence of the gammarid shredders on the assimilation rate of N. rhenorhodanensis, which fed on FPOM produced by the shredders, supporting the hypothesis of a positive interaction between surface shredders and hyporheic collector‐gatherers.  相似文献
