Background and aims

Osteoporosis, which is a disease characterized by weakening of the bone, affects a large portion of the senior population. The current therapeutic options for osteoporosis have side effects, and there is no effective treatment for severe osteoporosis. Thus, we urgently need new treatment strategies, such as topical therapies and/or safe and effective stem cell therapies.

Methods

We investigated the therapeutic potential of directly injecting human tonsil-derived mesenchymal stem cells (TMSC) into the right proximal tibias of ovariectomized postmenopausal osteoporosis model mice. Injections were given once (1×) or twice (2×) during the 3-month experimental period. At the end of the experiment, micro-computed tomographic images revealed some improvement in the proximal tibias and more significant improvement in the femoral heads of treated mice.

Results

Osteogenic effect was qualitatively and quantitatively more pronounced in TMSC/2×-treated mice. Furthermore, TMSC/2×?mice exhibited significant recovery of the serum osteocalcin level, which is pathologically elevated in osteoporosis, and increased serum alkaline phosphatase, which indicates bone formation. TMSC therapy was generally well tolerated and caused no apparent toxicity in the experimental mice. Moreover, TMSC therapy reduced visceral fat.

Conclusion

Our results demonstrate that double injection of TMSC directly into the proximal tibia triggers recovery of osteoporosis, and thus could be a potential therapeutic approach for severe bone loss.  相似文献

1. Although potentially vulnerable to predators, the offspring of subsocial insects are effectively protected by their parent(s). The female giant water bug Kirkaldyia deyrolli lays its egg masses on the vegetation above the water surface in aquatic environments and the males supply the eggs with water and guard them against cannibalistic females until hatchling dispersal. Field observations showed that egg masses are attacked by ants if the attending males are not present. 2. Laboratory experiments were conducted to evaluate the effects of paternal care by K. deyrolli against the ant Tetramorium tsushimae by means of four treatments: attending male with ant approach (WM‐WA); no attending male with ant approach (NoM‐WA); attending male without ant approach (WM‐NoA); and no attending male without ant approach (NoM‐NoA). 3. The rate of offspring survival was lower in the NoM‐WA group (45.3%) than in any other group, which showed similar offspring survival (WM‐WA = 80.4%, WM‐NoA = 75.1%, NoM‐NoA = 80.3%). Moreover, there were a total of 44 interactions between the attending male and ants in WM‐WA, and of these, a chemical compound was released by the attending male four times; this probably deterred ants from attacking because the ants went back to their colony. 4. In conclusion, the attending male can protect its eggs from ant predators and its care has an important role. To the best of the authors' knowledge, this is the first report of Lethocerinae males protecting their egg masses from ants by means of physical and chemical defence.  相似文献

Cells dissociated from the neural retina of embryonic chick differentiate into lens and pigment cells, when cultured in vitro. Using 3.5-day-old and 8.5-day-old chick embryos, we examined whether neuronal specificities would be expressed in such transdifferentiating cultures of neural retinal cells. The synthesis of acetylcholine and γ-aminobutyric acid (GABA) and the activity of choline acetyl transferase (CAT) was searched for in these cultures. The synthesis of an appreciable amount of these two putative neurotransmitters was detected in cultures of 3.5-day-old embryonic retinas by about 15 days. The activity of CAT was maximum in 7-day cultures of the 3.5-day-old materials and in 2-day cultures of the 8.5-day-old materials, and then decreased. Concomitant with the decrease of CAT-activity, δ-crystallin became detectable and increased thereafter. CAT-activity changed in parallel with the increase in the number of small neuroblast-like cells in cultures. The results demonstrate that the neuronal specificity identified by the appearance of acetylcholine and GABA and of the enzyme for the synthesis of acetylcholine is expressed in the early period of transdifferentiating cultures, which would later differentiate into lens and pigment cells. The possible mechanisms of the transition from neuronal to non-neuroretinal specificities of the transdifferentiating cultures are discussed.  相似文献

The phylogenetic relationship of Eurasian species of the Drosophila obscura species group remains ambiguous in spite of intensive analyses based on morphology, allozymes and DNA sequences. The present analysis based on sequence data for cytochrome oxidase subunit I (COI) and a-glycerophosphate dehydrogenase (Gpdh) suggests that the phylogenetic position of D. alpina is also ambiguous. These ambiguities have been considered to be attributable to rapid phyletic radiation in this group at an early stage of its evolution. Overwintering strategies are diversified among these species: D. alpina and D. subsihestris pass the winter in pupal diapause, D. bifasciata and D. obscura in reproductive diapause, and D. subobscura and D. guanche without entering diapause. This diversity may also suggest rapid radiation at an early phase of adaptations to temperate climates. On the other hand, adult tolerance of cold was closely related to overwintering strategy and distribution: D. obscura and D. bifasciata with reproductive diapause were very tolerant; D. alpina and D. subsilvestris which pass the winter in pupal diapause were less tolerant; D. subobscura having no diapause was moderately tolerant and D. guanche occurring in the Canary Islands was rather susceptible. Tolerance of high temperature at the preimaginal stages seemed to be also associated with overwintering strategy; i.e. lower in the species with pupal diapause than in those with reproductive diapause or without diapause mechanism.  相似文献

Photosynthetic nitrite reductase (PNiR) was purified extensivelyfrom spinach leaves by the method including acetone precipitation,ammonium sulfate fractionations and DEAE-cellulose column chromatography.The purified PNiR had the highest specific activity thus farobtained. Absorption spectrum of the purified PNiR indicatedthat it had no flavin component. There was no difference inthe rate of photosynthetic nitrite reduction between aerobicand anaerobic conditions. In chloroplasts two reductase systems related with ferredoxinhad been recognized, which were ferredoxin-nitrite and ferredoxin-NADPreductase system. The relationship between the two systems wasinvestigated by using the reconstituted enzyme system. Whenthe reduction of NADP was taking place, the reduction of nitritewas strongly inhibited. 1. Natural selection favours females who can correctly assess the predation risk and hence avoid high‐risk oviposition sites and reduce the mortality rate of their offspring. In spite of the potential significance of such behaviour, relatively few studies have assessed the relationship between oviposition behaviour and predation risk. 2. The present study aimed to determine the sublethal effects of predators on oviposition site selection by gravid females, the foraging activity of larvae, and the life history traits of two mosquito species that breed in different habitats, Aedes albopictus Skuse (container breeder) and Culex tritaeniorhynchus Giles (wetland breeder). 3. Female C. tritaeniorhynchus avoided laying eggs at oviposition sites in the presence of a predator cue. In contrast, female A. albopictus laid eggs in both the absence and presence of the predator cue. 4. To examine the effects of predator cues on larval behaviour, experiments were conducted in the absence and presence of a predator cue. Although larval activity was lower in the presence of the predator cue than that in its absence in both species, C. tritaeniorhynchus responded to the predator cue more strongly than A. albopictus. Female A. albopictus that had been reared with caged predators exhibited an extended larval development period, whereas the adult C. tritaeniorhynchus reared in the presence of predators were smaller than those reared in their absence. 5. This finding might explain why C. tritaeniorhynchus avoid laying eggs in predator‐conditioned water, for example to increase the fitness of their offspring, but A. albopictus either cannot detect predator cues or are not sensitive to them.  相似文献

  1. Two lactate dehydrogenases, L(+)- and D(–)- lactate cytochromec reductase, were extracted from the baker's yeast after disintegrationof the cells by a FRENCH press. They are separated by electrophoresison polyacrylamide gel and their activities were compared bycolor density of formazan, the reduction product of nitrobluetetrazolium.
  2. The ratio of L-lactate cytochrome c reductaseactivity to D-lactatecytochrome c reductase activity variedto a great extent, dependingon culture conditions. L-Lactatecytochrome c reductase waspredominant in resting cells; thereverse was the case withcells in early exponential stage ofthe growth.
  3. When the cells in exponential stage of growthwere aerated withoutnitrogen source, there occurred an intensiveincrease of L-lactatecytochrome c reductase, accompanied bythe decrease of D-lactatecytochrome c reductase.
  4. Effectsof inhibitors on the activity ratio of these two enzymeswereinvestigated. o-Phenanthroline, dinitrophenol, sodium azide,chloramphenicol, British antilewisite and antimycin A favored,in this order, the formation of L-lactate cytochrome c reductase.

Abstract: Pachytraga Paquier, 1900, the stratigraphically oldest genus of caprinine caprinid rudist, was previously known from only two chronospecies from a single lineage, that is the Hauterivian Ptubiconcha Astre, 1961 and the early Aptian Pparadoxa (Pictet and Campiche, 1869). Here, a new species, Pachytraga? tanakahitoshii, is erected on the basis of isolated left valves recovered from the Osaka and Sanchu areas, south‐west Japan. This species has a moderate shell size (antero‐posterior commissural diameter c. 30 mm), and its left valve is characterized by at least one possibly autapomorphic character (narrow anterior myophore, inclined inwards), as well as a mosaic of primitive (single longitudinal carina developed on the anterior side) and derived (simple marginal canals in the antero‐dorsal valve margin) characters of Mediterranean and Middle East Tethyan Pachytraga. The Japanese Pachytraga? represents the first probable record of this genus outside the Mediterranean/Middle Eastern Tethyan province, and its early Barremian age partly fills the ‘gap’ in its previously known stratigraphical record, although the evolutionary relationship of the Japanese form with Mediterranean and Middle East Tethyan Pachytraga remains unsolved. However, the discovery of early Barremian Pachytraga? in Japan indicates that the evolutionary history of the genus is more complex than previously thought and should thus be discussed in a broader palaeogeographical context that must now include the Pacific.  相似文献

ABSTRACT. To identify the unknown liver cell type initially invaded by sporozoiles of mammalian malaria, young rats were inoculated intravenously with large numbers of Plasmodium berghei sporozoites obtained from infected Anopheles stephensi mosquitoes. Fine structural studies of liver specimens obtained from the rats within 2 min after inoculation demonstrated the presence of morphologically unaltered sporozoites in the cytoplasm of hepatocytes. Many sporozoites were also observed undergoing cytolysis within the lysophagosomes of Kupffer cells, as well as other phagocytic cells. These observations strongly suggest direct infection of the hepatocyte by the sporozoite.  相似文献
