SYNOPSIS. Methods for the isolation and purification of DNA from the intraerythrocytic stages of the avian malaria parasite Plasmodium lophurae are described. The DNA of P. lophurae was found to have a 19 mole-percent guanine plus cytosine (G+C) composition as determined by melting temperature and density measurements in CsCl gradients, whereas that of the host cell nucleus was 35 molepercent G+C. Synthesis of DNA by P. lophurae was studied by following the incorporation of P32 during the in vitro growth of plasmodia in infected cell suspension cultures. DNA synthesis was linear during the multiple nuclear divisions of a single growth cycle in highly synchronous cultures. Separation of P32 labeled parasite DNA from the DNA of the duck erythrocyte on CsCl gradients showed that only parasite DNA was synthesized. 相似文献

In the context of an evolutionary study of the chemical communication in termites, sex pheromones and trail‐following pheromones were investigated in two Termopsidae, Zootermopsis nevadensis and Z. angusticollis. In these species, in which the presence of sex‐specific pheromones has been demonstrated previously, the chemical structure of the female sex pheromone has now been identified as (5E)‐2,6,10‐trimethylundeca‐5,9‐dienal and the male sex pheromone as (+)‐ or (?)‐syn‐4,6‐dimethyldodecanal. The amount of sex pheromone was estimated at 5–10 ng per individual in females and 2–5 ng in males. Because these two sympatric species do not differ in their pheromonal chemical composition, reproductive isolation is probably mediated chiefly by differences in dispersal flight chronology. The trail‐following pheromone was shown to be composed of the same compound as the male sex pheromone, that is syn‐4,6‐dimethyldodecanal. The compound syn‐4,6‐dimethyldodecanal was 10 times more active than the racemic (+/?)‐syn + (+/?)‐anti‐4,6‐dimethyldodecanal in eliciting trail‐following. The amount of syn‐4,6‐dimethyldodecanal was estimated at 0.1–0.5 ng per pseudergate. Regarding the phylogenetic aspects, the nature of the female sex pheromone of Zootermopsis is structurally akin to the trail‐following pheromone of Mastotermes darwiniensis of Mastotermitidae and Porotermes adamsoni and Stolotermes victoriensis of Termopsidae. When koa haole leaves were stored at elevated temperatures the mimosine content was decreased. The effect was most pronounced and rapid when the temperature was over 70 °C. in the presence of moisture. A similar effect occurred in seeds when sufficient moisture was present. The phenomenon did not take place in dry leaves. Steam was effective in extracting mimosine from koa haole leaves.Heated koa haole leaf meal has been demonstrated to be less toxic than unheated koa haole leaf meal when the meals are fed to albino rats as part of their rations. The development of alopecia in rats fed unheated koa haole meal has been described, and the growth curves of rats fed heated and unheated leaf meals and basal rations have been compared.Ferrous sulfate added to rations containing unheated koa haole leaf meal has been shown to be effective in reducing mimosine toxicity. 相似文献

Background
In rectal cancer, total mesorectal excision surgery combined with preoperative (chemo)radiotherapy reduces local recurrence rates but does not improve overall patient survival, a result that may be due to the harmful side effects and/or co-morbidity of preoperative treatment. New biomarkers are needed to facilitate identification of rectal cancer patients at high risk for local recurrent disease. This would allow for preoperative (chemo)radiotherapy to be restricted to high-risk patients, thereby reducing overtreatment and allowing personalized treatment protocols. We analyzed genome-wide DNA copy number (CN) and allelic alterations in 112 tumors from preoperatively untreated rectal cancer patients. Sixty-six patients with local and/or distant recurrent disease were compared to matched controls without recurrence. Results were validated in a second cohort of tumors from 95 matched rectal cancer patients. Additionally, we performed a meta-analysis that included 42 studies reporting on CN alterations in colorectal cancer and compared results to our own data.Results
The genomic profiles in our study were comparable to other rectal cancer studies. Results of the meta-analysis supported the hypothesis that colon cancer and rectal cancer may be distinct disease entities. In our discovery patient study cohort, allelic retention of chromosome 7 was significantly associated with local recurrent disease. Data from the validation cohort were supportive, albeit not statistically significant, of this finding.Conclusions
We showed that retention of heterozygosity on chromosome 7 may be associated with local recurrence in rectal cancer. Further research is warranted to elucidate the mechanisms and effect of retention of chromosome 7 on the development of local recurrent disease in rectal cancer.Electronic supplementary material
