The Coquerel's sifaka (Propithecus coquereli) is one of the species of Propithecus, living in the dry forest of northwest Madagascar. This species is endangered due to the loss and fragmentation of its natural habitat, consequential of deforestation. Twenty nuclear microsatellite loci were isolated from genomic DNA derived from a free ranging Coquerel's sifaka from the Mariarano Classified Forest, Madagascar. Population genetic parameters were estimated as baseline values from samples collected in that forest.  相似文献

ABSTRACT Use of non-invasive sources of DNA, such as hair or scat, to obtain a genetic mark for population estimates is becoming commonplace. Unfortunately, with such marks, potentials for genotyping errors and for the shadow effect have resulted in use of many loci and amplification of each specimen many times at each locus, drastically increasing time and cost of obtaining a population estimate. We proposed a method, the Genotyping Uncertainty Added Variance Adjustment (GUAVA), which statistically adjusts for genotyping errors and the shadow effect, thereby allowing use of fewer loci and one amplification of each specimen per locus. Using allele frequencies and estimates of genotyping error rates, we determined, for each pair of specimens, the probability that the pair was obtained from the same individual, whether or not their observed genotypes match. Using these probabilities, we reconstructed possible capture history matrices and used this distribution to obtain a population estimate. With simulated data, we consistently found our estimates had lower bias and smaller variance than estimates based on single amplifications in which genotyping error was ignored and that were comparable to estimates based on data free of genotyping errors. We also demonstrated the method on a fecal DNA data set from a population of red wolves (Canis rufus). The GUAVA estimate based on only one amplification genotypes compares favorably to the estimate based on consensus genotypes. A program to conduct the analysis is available from the first author for UNIX or Windows platforms. Application of GUAVA may allow for increased accuracy in population estimates at reduced cost.  相似文献

ABSTRACT A majority of North American breeding habitat for neotropical migrants exists on private lands, requiring monitoring strategies focused on habitat in these private holdings. We outline study designs and protocols using repeated Presence-Absence surveys across a gradient of patch sizes to develop a range-wide monitoring program for the endangered golden-cheeked warbler (Dendroica chrysoparia) in Texas, USA. We surveyed 200–400 point-count locations across approximately 30 private properties annually from 2005 to 2008. We used data from our surveyed patches (n = 147) and the Ψ (occupancy), p (detection), and γ = 1 - ɛ parameterization to estimate patch dynamics and associated detection probabilities for golden-cheeked warblers. Patch size had a strong association with patch occupancy, and all patches >160 ha were predicted to be occupied. We found no evidence that large golden-cheeked warbler populations located on public lands in the vicinity of our study area influenced occupancy dynamics. We conducted simulations across a range of detection probabilities to evaluate potential sample sizes for both standard- and removal-based occupancy modeling. Simulations using parameter estimates from our analysis indicated that removal-based sampling is superior to standard sampling. Based on our results, surveying golden-cheeked warbler presence in oak-juniper (Quercus-Juniperus) patches under a removal modeling framework should be considered as one alternative for range-wide monitoring programs because patch-level monitoring would be necessary to estimate proportion of range occupied. Large contiguous patches are rare across the species’ range; hence, conservation and management of the mosaic of smaller patches within a landscape context would be required for maintaining species viability. Thus, we recommend the identification of areas where smaller, contiguous patches represent a significant portion of the available habitat within the local landscape and targeting these areas for habitat maintenance and improvement.  相似文献

Twenty‐one polymorphic microsatellite loci were isolated and characterized from the genome of the prehensile‐tailed skink (Corucia zebrata). The markers were screened using a set of 16 individuals from the collection of the Philadelphia Zoo. The corresponding characterization indicated significant levels of heterozygosity and information potential for use in future studies. These markers will be a valuable tool in analysing the paternal contributions in twin birth questions and for population genetic studies in wild populations.  相似文献

The effects of four potential inhibitors of dictyosome activityon the root cap secretory system were monitored by visual estimationof slime droplet reformation rates and by quantitative microscopyof the secretory cells. Only monensin was found to affect bothdroplet reformation and cell structure. While some of our structuralobservations on the effects of this drug, such as swelling ofvesicles and dictyosome cisternae, agreed with those made previously,others did not. We are able to confirm a real increase in vesiclenumber, in addition to the numerical increase in vesicle profilesthat follows from an increase in vesicle size. Formation ofcup-shaped dictyosomes and separation of cisternae were foundto be just as prevalent in the normal and in the solvent controls,especially when fixed with permanganate. Scopoletin, tunicamycin and 2, 6-dichlorobenzonitrile all affecteddroplet formation but had no significant effect on cell structure.It is suggested that these chemicals were affecting water flowinto the slime droplet, rather than directly inhibiting Golgi-activityor release of carbohydrates by the secretory vesicles. The problems of using the root cap system for the identificationof specific Golgi inhibitors are discussed. Climate change may influence pelagic fish by altering advective processes or by changing where fish choose to spawn. Using a simulation model, the effects of altered advection and spatial distribution of spawning by anchovy on recruitment off South Africa were explored. Cape anchovy Engraulis capensis spawn on the region of the Agulhas Bank, south of South Africa. Currents transport eggs and larvae to nursery areas. Transport of eggs and larvae was modelled using a flow field based on averaged Acoustic Doppler Current Profile data, Feasible Scenarious of altered advection were modelled. For modelling purposes, the ocean surrounding South Africa was divided into blocks with dimensions of a quarter-degree latitude by a quarter-degree longitude. Acoustically measured distributions of spawner biomass for the years 1986–92 were used to calculate egg production per block. In the model, batches were released from each of these blocks each day of the spawning season. The modelling study indicates that passive transport of young anchovy may account for a substantial proportion of year-class variability. Model results show that distribution of spawners influences the distribution of young of the year, as well as the number and the location of advective losses across offshore boundaries.  相似文献

1. The bottom‐up factors that determine parasitoid host use are an important area of research in insect ecology. Host size is likely to be a primary cue for foraging parasitoids due to its potential influence on offspring development time, the risk of multiparasitism, and host immunocompetence. Host size is mediated in part by host‐plant traits that influence herbivore growth and potentially affect a herbivore's quality as a host for parasitoids. 2. Here, we tested how caterpillar host size and host plant species influence adult fly parasitoid size and whether host size influences wasp parasitoid sex allocation. We measured the hind tibia lengths and determined the sex of wasp and fly parasitoids reared from 11 common host species of polyphagous caterpillars (Limacodidae) that were in turn reared on foliage of seven different host plant species. 3. We also tested how host caterpillar species, host caterpillar size, and host and parasitoid phenology affect how the parasitoid community partitions host resources. We found evidence that parasitoids primarily partition their shared hosts based on size, but not by host species or phenology. One index of specialisation (d′) supports our observation that these parasitoids are quite generalised within the Limacodidae. In general, wasps were reared from caterpillars collected in early instars, while flies were reared from caterpillars collected in late instars. Furthermore, for at least one species of solitary wasp, host size influenced sex allocation of offspring by ovipositing females. 4. Host‐plant quality indirectly affected the size attained by a tachinid fly parasitoid through its direct effects on the size and performance of the caterpillar host. The host plants that resulted in the highest caterpillar host performance in the absence of enemies also yielded the largest parasitoid flies, which suggests that host plant quality can cascade up to influence the third trophic level.  相似文献
