Concentration of Bacteriophages from Natural Waters 总被引:3,自引:2,他引:1
The methods used for concentrating animal viruses from drinking water were found to be unsuitable for the concentration of bacteriophages from natural waters. The factors affecting recovery were investigated and a concentration procedure devised which is amenable to larger scale and field use. This procedure involves: (1) passage of the water through a sand filter; (2) removal of dissolved organic material with an anion exchange resin; (3) addition of MgCl2 to a final concentration of 5 times 10-4 m ; (4) adjustment of the pH value to 3°8; (5) adsorption of the bacteriophages on to fibre glass and cellulose nitrate filters; (6) elution of bound phage with 3% (w/v) beef extract, and (7) concentration by ultrafiltration of the resulting eluates. Using this procedure a wide range of test bacteriophages was concentrated from 41 to 5 ml with recoveries ranging from 18–80%—concentration factors of 200–900 fold. 相似文献

SUMMARY.
- 1 Research was performed in laboratory streams to evaluate periphytic biomass accrual, export, and community composition over a range of limiting nutrient (phosphorus) concentrations with variable velocity, and suspended sediment addition, in comparison to constant velocity and no suspended sediment. In fixed-velocity treatments, velocity increase to 60 cm s?1 significantly enhanced biomass accrual, but further increase resulted in substantial biomass reduction. Average biomass loss rates did not change significantly over a velocity range of 10–80 cm s?1. Diatoms were favoured at relatively high velocities and low phosphorus concentrations, whereas the blue-green Phormidium tended to dominate at higher SRP concentrations and the green Mougeotia seemed to prefer lower velocities.
- 2 Sudden increases in velocity raised instantaneous loss rates by an order of magnitude or more, but these high rates persisted only briefly. As a result, marked biomass reductions were not apparent a day after the velocity change. Dominance change from filamentous green or blue-green to diatoms immediately after the increase was reversed within 2 days. Loss rate increases due to solids addition were much smaller than those accompanying velocity increase, but simultaneous velocity elevation and solids addition produced instantaneous loss rates approximately double those with velocity increase alone.
- 3 The experiments demonstrated that an elevation in velocity, above that to which algae were accustomed, led to increased loss rates and temporarily reduced biomass. However, recolonization and growth after biomass reduction were apparently rapid. Substantial export of periphyton following solids addition required erosion of the protective boundary layer accompanied by a velocity increase. These results arc applicable to understanding the response of lotic periphytic algae to elevated, turbid storm discharges and similar runoff or high-flow events.
- 4 Areal uptake rates of P by algae growing in the laboratory streams increased with soluble reactive phosphorus (SRP) concentration, up to approximately 15 μg I?1 in overlying water. They also increased above 35 cm s ?1. Overall, uptake rate seemed to vary inversely with biomass. The ralio of areal uptake rate/biomass was significantly less where mean biomass was 411±6 mg chl a m?2 compared to 223±17 mg chl a m?2.
- 5 The results suggested that although nutrient uptake is primarily a surface phenomenon, diffusion to interior cells can also determine the responses of attached communities. Both diffusion and uptake rate were stimulated by increasing nutrient concentration and velocity up to certain levels, but became limited by biofilm thickness and scouring.

To determine if increased abscisic acid (ABA) levels are associated with ultraviolet-B (UV-B) radiation (288–320 nm) stress, Rumex patientia L. plants were exposed to visible light with high or low (control) UV-B irradiance and then assayed for ABA by gas chromatography. Though leaf growth was inhibited by the UV-B irradiation, no differences in levels of free ABA were found between leaves from the two treatments after 1, 3 or 5 days of exposure. Unlike the situation in most environmental stresses, increased ABA levels are not associated with UV-B radiation stress. Further, photolysis or isomerization are not likely explanations for the absence of ABA increase. Calculations indicate that if ABA in the leaf were fully exposed to solar UV-B radiation only about 6.7% would be photolyzed by existing daily radiation at 40°N latitude, and 11.3% by radiation resulting from a 40% decrease in atmospheric ozone. The quantum yield for isomerization of cistrans-ABA by radiation in the 288–312 nm waveband was determined. Existing daily solar UV-B irradiation is capable of isomerizing ABA in solution to an equilibrium mixture of 50% cisjrans and 50%irans, trans-ABA., However, because of radiation attenuation in the leaf, it is estimated that only a small fraction of the cistrans-A.BA. would be isomerized. 相似文献

A positive family history of alcohol use disorders (FH) is a robust predictor of personal alcohol abuse and dependence. Exposure to problem-drinking models is one mechanism through which family history influences alcohol-related cognitions and drinking patterns. Similarly, exposure to alcohol advertisements is associated with alcohol involvement and the relationship between affective response to alcohol cues and drinking behavior has not been well established. In addition, the collective contribution that FH, exposure to different types of problem-drinking models (e.g. parents, peers) and personal alcohol use have on appraisal of alcohol-related stimuli has not been evaluated with a large sample. We investigated the independent effects of FH, exposure to problem-drinking models and personal alcohol use on valence ratings of alcohol pictures in a college sample. College students (n = 227) completed measures of personal drinking and substance use, exposure to problem-drinking models, FH and ratings on affective valence of 60 alcohol pictures. Greater exposure to non-familial problem-drinkers predicted greater drinking among college students (beta = 0.17, P < 0.01). However, personal drinking was the only predictor of valence ratings of alcohol pictures (beta = -0.53, P < 0.001). Personal drinking level predicted valence ratings of alcohol cues over and above FH, exposure to problem-drinking models and demographic characteristics. This suggests that positive affective responses to alcohol pictures are more a function of personal experience (i.e. repeated heavy alcohol use) than vicarious learning. 相似文献
