Specific interactions of guanine nucleotides with rat brain membrane preparations were investigated; a two-step scheme of this interaction was proposed. The effect of GTP on the agonist binding to rat brain opioid receptors was studied. Nucleotides were shown to inhibit the ligand interactions with mu- and sigma-receptors. It was found that the GTP-induced inhibition differs from that of the GTP stable analog, GppNp, i.e., GTP does not initiate any noticeable dissociation of the receptor system component and its liberation from the membrane into solution as is the case with GppNp. A kinetic model of the GTP interactions with the opioid receptor system stipulating that the affinity of high affinity centers for mu- and delta-ligands changes after nucleotide hydrolysis on the N-protein, was proposed. The differences in the mechanisms of GTP effect on mu- and sigma-receptors were revealed, i.e., the inhibition of the ligand binding to mu-receptors may occur just after GTP binding to the N-protein. The ligand-receptor interactions in the presence of GTP were modelled according to the proposed scheme.  相似文献

The complete nucleotide sequence of pNB2, a 1.9-kilobases cryptic plasmid from thermophilic Clostridium thermosaccharolyticum has been determined. The plasmid consists of 1882 base pairs and has a G+C composition of 27.2%. The sequence contains three open reading frames capable of coding for polypeptides two of which were identified in maxicell Escherichia coli extracts. Our future studies are directed toward a construction of pNB2-derivatives as vectors for Clostridia.  相似文献

Kinetic relationships of photoreduction of 4,4'-dimethyldipyridyl and 4,4'-dibenzyldipyridyl in the presence of chlorplast were studied. It was found that photoreduction leads to the establishment of photosteady state in which the rate of photoreduction is equal to the rate of dark oxidation of reduced form. Kinetics of dark oxidation of photoreduced form were investigated. The study of inhibition of photoreduction of methylviologen and potassium ferricyanide by diuron was performed. It was shown that the nature of inhibition of both reactions is completely identical. The effect of hydrogen ions concentration and gas phase composition was studied. Stabilization of low potential electron acceptors to the oxidation by molecular oxygen in the reaction of photooxidation of water is discussed.  相似文献

The toroidal inhomogeneity of the poloidal magnetic field—the so-called error fields that arise due to imperfections in manufacturing and assembling of the electromagnetic system-was measured in the Globus-M spherical tokamak. A substantial inhomogeneity corresponding to the n = 1 mode, which gave rise to a locked mode and led to discharge disruption, was revealed. After compensation of this inhomogeneity with the help of special correction coils, the discharge duration increased and the global plasma parameters improved substantially. A technique for determining and compensating the n = 1 mode inhomogeneity is described, the measured dependences of the penetration threshold of the m = 2/n = 1 mode on the plasma parameters are given, and results of experiments in which record parameters for the Globus-M tokamak were achieved after correction of the poloidal magnetic field are presented.  相似文献

Prior research has indicated that the two sperm cells of Nicotiana tabacum are dimorphic, suggesting that they may participate in preferential fertilization during in vivo fusion with the egg and central cells. To probe the mechanism of potential preferential fertilization in this plant, it will be necessary to use modern sensitive molecular techniques. For this purpose, two individual populations of two sperm cells, constituting the Svn (associated with the vegetative nucleus) and Sua (unassociated with the vegetative nucleus), were isolated in the thousands from tobacco pollen tubes with a micromanipulator as a preliminary step toward research on gametic recognition using molecular techniques. Microelectrophoresis of paired sperm cells from a single pollen tube was conducted at different developmental stages. Sperm cells isolated from 1-, 2-, 3- and 4-cm stylar lengths migrated to the negative pole, with the Sua displaying significantly greater electrophoretic mobility than the Svn, reflecting a more positively charged cell surface on the Sua. The sperm cells isolated from 1-cm style are very sensitive to electron potential in an electrophoretic field, presumably reflecting that they are still in a young state. In animal cells arachidonic acid is metabolized via the 5-, 12- and 15-lipoxygenase pathways. The kinetic mechanism of action of plant (soya) and animal (reticulocyte) 15-lipoxygenases is now well established. 5-Lipoxygenase possesses, in all probability, the most complex mechanism of activity regulation. At present several effectors of neutrophil 5-lipoxygenase, both cytosolic and membrane-bound ones, have been identified. The molecular and kinetic mechanisms of action of the enzyme are still open to question. A kinetic scheme of regulation of synthesis of arachidonic acid 5-lipoxygenase metabolites which does not exclude the presence of two binding sites on the enzyme molecule, is proposed. Within the framework of this kinetic scheme the enzyme activator complex may be the active form of the enzyme. There is evidence that the curve for the time dependence of 5-HETE accumulation in neutrophils stimulated by the Ca2+ ionophore A23187 has a maximum, while the corresponding curve for the LTB4 accumulation is a curve with saturation. It was shown that an increase in the concentration of exogenous arachidonate induces the synthesis of 5-HETE, whereas the concentration of LTB4 remains practically unchanged. The results of mathematical analysis of the above kinetic scheme and a comparison of experimental and calculated values suggest that the reaction effector, Ca2+, plays a crucial regulatory role in the observed kinetic dependencies reflecting the formation of two sequential products of 5-lipoxygenase oxidation of arachidonate. In this way Ca2+ strongly influences the first step of the reaction, i.e., 5-HETE formation; its effect on the second reaction step (5-HETE conversion into LTA4) is far less apparent.  相似文献

Pitfall traps were used to monitor predatory arthropod numbers along two types of field boundary, a post and wire fence line and a Cupressus macrocarpa hedge, along the same paddock margin in Canterbury, New Zealand, over 24 months. The seven most abundant predator groups recorded were: Araneae > Phalangiidae > Staphylinidae > Coccinellidae > Chilopoda > Hemerobiidae > Carabidae. Araneae, Phalangiidae, Staphylinidae, Chilopoda and Hemerobiidae were found in larger numbers at the wire fence than at the hedge site, whereas the numbers of Carabidae and Coccinellidae adults exhibited no field margin preference. However, more species of Araneae and Staphylinidae were caught at the hedge site, whereas species richness of carabid beetles was greatest at the wire fence. Principal component analysis clearly separated the samples collected from the two habitats based on the assemblages of Araneae, Staphylinidae and Carabidae, and certain species in each of these taxonomic groups appeared to be particularly associated with one boundary type or the other. All the main taxonomic groups exhibited clear seasonal patterns, with distinct peaks in abundance occurring at certain times of the year. The results of the study reinforce the idea that management of field boundaries can be used to manipulate the type and abundance of particular groups of predatory arthropods, and that seasonal patterns should be taken into account in schemes of integrated pest management so that any adverse effects of biocide application on these beneficial species may be minimised.  相似文献

The stereoselective affinity of small-molecule binding to proteins is typically broadly explained in terms of the thermodynamics of the final bound complex. Using Brownian dynamics simulations, we show that the preferential binding of the MDM2 protein to the geometrical isomers of Nutlin-3, an effective anticancer lead that works by inhibiting the interaction between the proteins p53 and MDM2, can be explained by kinetic arguments related to the formation of the MDM2:Nutlin-3 encounter complex. This is a diffusively bound state that forms prior to the final bound complex. We find that the MDM2 protein stereoselectivity for the Nutlin-3a enantiomer stems largely from the destabilization of the encounter complex of its mirror image enantiomer Nutlin-3b, by the K70 residue that is located away from the binding site. On the other hand, the trans-Nutlin-3a diastereoisomer exhibits a shorter residence time in the vicinity of MDM2 compared with Nutlin-3a due to destabilization of its encounter complex by the collective interaction of pairs of charged residues on either side of the binding site: Glu25 and Lys51 on one side, and Lys94 and Arg97 on the other side. This destabilization is largely due to the electrostatic potential of the trans-Nutlin-3a isomer being largely positive over extended continuous regions around its structure, which are otherwise well-identified into positive and negative regions in the case of the Nutlin-3a isomer. Such rich insight into the binding processes underlying biological selectivity complements the static view derived from the traditional thermodynamic analysis of the final bound complex. This approach, based on an explicit consideration of the dynamics of molecular association, suggests new avenues for kinetics-based anticancer drug development and discovery.  相似文献

By using high performance liquid chromatography with simultaneous detection of unlabeled and radiolabeled product of lipoxygenase oxidation of arachidonic acid, the mechanism of exogenous arachidonate involvement in leukotriene synthesis in human neutrophils induced by the Ca2+ ionophore A23187 was studied. It was found that after addition of labeled arachidonate the specific radioactivity of the reaction product (leukotriene B4) does not change on a time scale, i.e., the free arachidonic acid exchange between the cell and extracellular space is a very rapid process. Exogenous arachidonic acid was found to be the substrate of the lipoxygenase reaction which acts in parallel with the endogenous one. The dependence of specific radioactivity of leukotriene B4 in added arachidonic acid concentration is described by a hyperbolic curve with saturation. When exogenous arachidonate is used at a concentration of 10.8 +/- 3.9 microM, that of intracellular arachidonic acid increases twofold at the expense of the exogenously added acid.  相似文献
