We asked whethercrystalloid administration improves tissue oxygen extraction inendotoxicosis. Four groups of anesthetized pigs(n = 8/group) received either normalsaline infusion or no saline and either endotoxin or no endotoxin. Wemeasured whole body (WB) and gut oxygen delivery and consumption duringhemorrhage to determine the critical oxygen extraction ratio(ERO2 crit). Just after onset of ischemia (critical oxygen delivery rate), gut was removed for determination of area fraction of interstitial edema and capillary hematocrit. Radiolabeled microspheres were used todetermine erythrocyte transit time for the gut. Endotoxin decreased WBERO2 crit(0.82 ± 0.06 to 0.55 ± 0.08, P < 0.05) and gutERO2 crit(0.77 ± 0.07 to 0.52 ± 0.06, P < 0.05). Unexpectedly, saline administration also decreased WBERO2 crit (0.82 ± 0.06 to 0.62 ± 0.08, P < 0.05) and gutERO2 crit (0.77 ± 0.07 to 0.67 ± 0.06, P < 0.05) in nonendotoxin pigs. Saline administration increased thearea fraction of interstitial space (P < 0.05) and resulted in arterial hemodilution(P < 0.05) but not capillaryhemodilution (P > 0.05). Salineincreased the relative dispersion of erythrocyte transit times from0.33 ± 0.08 to 0.72 ± 0.53 (P < 0.05). Thus saline administration impairs tissue oxygen extractionpossibly by increasing interstitial edema or increasing heterogeneityof microvascular erythrocyte transit times.

Checklists of the marine algae of Malaysia have been published. The last checklist included the marine algae of Singapore. The checklist recorded 212 taxa for the region. This paper lists an additional 7 taxa of Chlorophyta, 18 taxa of Rhodophyta and 5 taxa of Phaeophyta, which had been left out of the last checklist. These include marine algae from Pulau Pinang, Pulau Redang and Pulau Sibu. In addition, new records of Cyanophyta (2 species), Chlorophyta (4 species), Rhodophyta (10 species) and Phaeophyta (3 species) are reported.  相似文献

The medullary bone serves as a source of labile calcium mobilized during calcification of the egg shell in birds. Quantitative histological methods demonstrate that the numbers of medullary bone osteoclasts and nuclei per osteoclast remain unchanged during the egg cycle in the Japanese quail (Coturnix). Therefore, cyclic changes in bone resorption cannot be explained by modulations of osteoclasts from and into other bone cells, a mechanism previously suggested for certain species of birds. Rather, dramatic changes in osteoclast cell-surface features occur during the egg cycle, which might account for cyclic variations in resorptive activity. During egg shell calcification, osteoclasts with ruffled borders are closely apposed to bone surfaces; the cytoplasm is rich in vacuoles that contain mineral crystals and seem to derive from the ruffled border. At the completion of egg shell calcification, the ruffled borders and vacuoles move away from the bone surface, although the osteoclast remains attached to the bone along the filamentous or "clear" zone. Associated with the disappearance of the ruffled borders is the appearance of extensive interdigitated cell processes along the peripheral surface of the osteoclast away from the bone. These unusual structures, which may serve as a reservoir of membrane, largely disappear when ruffled borders and associated structures reappear. Therefore, in these hens, the osteoclasts modulate their cell surface rather than their population during the egg cycle.  相似文献

Profound changes in calcium metabolism occur during pregnancy. The mother has to make available extra calcium for fetal requirements while ensuring that her plasma and bone calcium concentrations are satisfactorily maintained. In a cross-sectional study plasma concentrations of the major calcium-regulating hormones--namely, calcitonin, parathyroid hormone, 25-hydroxyvitamin D (25-OHD), and 1,25-dihydroxyvitamin D (1,25-(OH)2D)--were measured to establish their interrelations during normal pregnancy. The major changes observed were increases in the circulating concentrations of 1,25-(OH)2D and calcitonin. Concentrations of parathyroid hormone and 25-OHD remained within the normal range. The increased concentrations of 1,25-(OH)2D enable the increased physiological need for calcium to be met by enhancing intestinal absorption of this element. The simultaneous rise in calcitonin opposes the bone-resorbing activities of 1,25-(OH)2D, thereby protecting the integrity of the maternal skeleton. Maternal calcium homeostasis is thus maintained yet the requirements of the fetus are fulfilled.  相似文献

We previously reported that pyrroline-5-carboxylate (PC), the intermediate in the interconversions of proline, ornithine and glutamate markedly stimulates hexosemonophosphate-pentose pathway activity in human erythrocytes. The stimulation is mediated by pyrroline-5-carboxylate reductase which generates NADP+ accompanying the conversion of pyrroline-5-carboxylate to proline. We now report that the previously demonstrated effect of pyrroline-5-carboxylate on glucose oxidation through the hexose-monophosphate-pentose pathway is accompanied by increased phosphoribosyl-pyrophosphate production and increased formation of nucleotides via the salvage pathway. The demonstrated effect of pyrroline-5-carboxylate on purine processing may provide a regulatory link between amino acid and nucleotide metabolism.  相似文献

Prolidase (EC 3.4.13.9) is a ubiquitously distributed imidodipeptidase that catalyzes the hydrolysis of C-terminal proline or hydroxyproline containing dipeptides. The enzyme plays an important role in the recycling of proline for collagen synthesis and cell growth. An increase in enzyme activity is correlated with increased rates of collagen turnover indicative of extracellular matrix (ECM) remodeling, but the mechanism linking prolidase activity and ECM is poorly understood. Thus, the effect of ECM-cell interaction on intracellular prolidase activity is of special interest. In cultured human skin fibroblasts, the interaction with ECM and, more specifically, type I collagen mediated by the β1 integrin receptor regulates cellular prolidase activity. Supporting evidence comes from the following observations: 1) in sparse cells with a low amount of ECM collagen or in confluent cells in which ECM collagen was removed by collagenase (but not by trypsin or elastase) treatment, prolidase activity was decreased; 2) this effect was reversed by the addition of type I collagen or β1 integrin antibody (agonist for β1 integrin receptor); 3) sparse cells (with typically low prolidase activity) showed increased prolidase activity when grown on plates coated with type I collagen or on type IV collagen and laminin, constituents of basement membrane; 4) the relative differences in prolidase activity due to collagenase treatment and subsequent recovery of the activity by β1 integrin antibody or type I collagen treatment were accompanied by parallel differences in the amount of the enzyme protein recovered from these cells, as shown by Western immunoblot analysis. Hematopoietic cells can be highly enriched for repopulating ability based upon the efflux of the fluorescent Hoechst 33342 dye by sorting for SP (side population) cells, a phenotype attributed to expression of ABCG2, a member of the ABC transporter superfamily. Intriguingly, murine studies suggest that forced ABCG2 expression prevents hematopoietic differentiation. We cloned the full-length rhesus ABCG2 and introduced it into a retroviral vector. ABCG2-transduced human peripheral blood progenitor cells (PBPCs) acquired the SP phenotype but showed significantly reduced growth compared with control. Two rhesus macaques received autologous PBPCs split for transduction with the ABCG2 or control vectors. Marking levels were similar between fractions with no discrepancy between bone marrow and peripheral blood marking. Analysis for the SP phenotype among bone marrow and mature blood populations confirmed ABCG2 expression at levels predicted by vector copy number long term, demonstrating no block to differentiation in the large animal. In vitro studies showed selective protection against mitoxantrone among ABCG2-transduced rhesus PBPCs. Our results confirm the existence of rhesus ABCG2, establish its importance in conferring the SP phenotype, suggest no detrimental effect of its overexpression upon differentiation in vivo, and imply a potential role for its overexpression as an in vivo selection strategy for gene therapy applications.  相似文献

Increased expression of prostaglandin endoperoxide H synthase-2 (PGHS-2) has been implicated in pathological conditions such as inflammatory bowel diseases and colon cancer. Recently, it has been demonstrated that inducible nitric oxide synthase (NOS II) expression and nitric oxide (NO) production are up-regulated in these diseases as well. However, the apparent link between PGHS-2 and NOS II has not been thoroughly investigated in nontransformed and nontumorigenic colonic epithelial cells. In the present study, we examined the concomitant expression of PGHS-2 and NOS II as well as the production of prostaglandin E2 (PGE2) and NO in conditionally immortalized mouse colonic epithelial cells, namely YAMC (Apc(+/+)). We found that the induction of PGHS-2 and generation of PGE2 in these cells by IFN-gamma and lipopolysaccharide (LPS) were greatly reduced by two selective NOS II inhibitors, L-NIL and SMT. To ascertain the effect of NO on PGHS-2 overexpression, we tested NO-releasing compounds, NOR-1 and SNAP, and found that they caused PGHS-2 expression and PGE2 production. This effect was abolished by hemoglobin, a NO scavenger. Using electrophoretic mobility shift assays, we found that both NOR-1 and SNAP caused beta-catenin/LEF-1 DNA complex formation. Super-shift by anti-beta-catenin antibody confirmed the presence of beta-catenin in the complex. Cell fractionation studies indicated that NO donors caused an increase in free soluble cytoplasmic beta-catenin. This is further corroborated by the immunocytochemistry data showing the redistribution of beta-catenin from the predominantly membrane localization into the cytoplasm and nucleus after treatment with NO donors. To further explore the possible connection between PGHS-2 expression and beta-catenin/LEF-1 DNA complex formation, we studied IMCE (Apc(Min/+)) cells, a sister cell line of YAMC with similar genetic background but differing in Apc genotype and, consequently, their beta-catenin levels. We found that IMCE cells, in comparison with YAMC cells, had markedly higher beta-catenin/LEF-1 DNA complex formation under both resting conditions as well as after induction with NO. In parallel fashion, IMCE cells expressed significantly higher levels of PGHS-2 mRNA and protein, and generated more PGE2. Overall, this study suggests that NO may be involved in PGHS-2 overexpression in conditionally immortalized mouse colonic epithelial cells. Although the molecular mechanism of the link is still under investigation, this effect of NO appears directly or indirectly to be a result of the increase in free soluble beta-catenin and the formation of nuclear beta-catenin/LEF-1 DNA complex.  相似文献

Taxonomic and systematic studies of the woody bamboos are traditionallybased on floral morphology, which can cause problems in identificationdue to the lack of, or infrequent, flowering. Limited studieshave been conducted using molecular techniques to overcome thisproblem. In this study, we used amplified fragment length polymorphisms(AFLPs) to conduct a study of four genera of bamboos (Bambusa,Dendrocalamus, Gigantochloa andThyrsostachys ) in the subtribeBambusinae. AFLP analysis using eight primer combinations wascarried out on 15 species of bamboo. Results showed that AFLPsdistinguish the different species by their unique banding patterns.Unique AFLPs were detected in 13 of the 15 species examined.The six Bambusa species examined separated into two clusters.The sixGigantochloa species studied formed a discrete clusterdiverging from one of the Bambusa clusters, whileThyrsostachyswas less similar to the Bambusa clusters. The similarity indexbetween B. lako and G. atroviolacea was the highest, suggestingthat B. lako is more appropriately included within the genusGigantochloarather than the genus Bambusa. The two Dendrocalamus speciesexamined were very different with D. brandisii clustering withinone of the Bambusa clusters and D. giganteus appearing as avery distant species. These results support the contention thatcritical study of the genus Dendrocalamus is required.
