Resistance to the potato cyst nematode (PCN) species Globodera pallida, derived from the wild diploid potato species Solanum vernei, has been investigated. This source of resistance, which is effective against all of the major pathotypes of G. pallida and Globodera rostochiensis, has been assumed to be due to several genetic factors, but it has proved difficult to deploy effectively in breeding strategies for potato cultivars. Diploid and tetraploid potato populations segregating for 'vernei' resistance were analysed. At the tetraploid level, a bulk segregant analysis (BSA) approach was employed and detected AFLP markers linked to a resistance QTL on potato linkage group V. Conventional linkage analysis of a diploid population identified QTL on linkage groups V and IX. A marker linked to a QTL on linkage group V has been converted to a single-locus PCR-based marker, which can be used to detect the presence of the QTL in diploid and tetraploid potato germplasm. Moreover, there is evidence that one of the AFLPs detected by BSA appears to be specific to an introgressed segment of DNA from S. vernei. These results are compared with those obtained from other studies on resistance to the PCN species G. pallida.  相似文献

Richards (1985) showed that veridical three-dimensional shape may be recovered from the integration of binocular disparity and retinal motion information, but proposed that this integration may only occur for horizontal retinal motion. Psychophysical evidence supporting the combination of stereo and motion information is limited to the case of horizontal motion (Johnston et al., 1994), and has been criticised on the grounds of potential object boundary cues to shape present in the stimuli. We investigated whether veridical shape can be recovered under more general conditions. Observers viewed cylinders that were defined by binocular disparity, two-frame motion or a combination of disparity and motion, presented at simulated distances of 30 cm, 90 cm or 150 cm. Horizontally and vertically oriented cylinders were rotated about vertical and horizontal axes. When rotation was about the cylinder's own axis, no boundary cues to shape were introduced. Settings were biased for the disparity and two-frame motion stimuli, while more veridical shape judgements were made under all conditions for combined cue stimuli. These results demonstrate that the improved perception of three-dimensional shape in these stimuli is not a consequence of the presence of object boundary cues, and that the combination of disparity and motion is not restricted to horizontal image motion.  相似文献

Recent evidence suggests that the visual control of prehension may be less dependent on binocular information than has previously been thought. Studies investigating this question, however, have generally only examined reaches to single objects presented in isolation, even though natural prehensile movements are typically directed at objects in cluttered scenes which contain many objects. The present study was designed, therefore, to assess the contribution of binocular information to the control of prehensile movements in multiple-object scenes. Subjects reached for and grasped objects presented either in isolation or in the presence of one, two or four additional 'flanking' objects, under binocular and monocular viewing conditions. So that the role of binocular information could be clearly determined, subjects made reaches both in the absence of a visible scene around the target objects (self-illuminated objects presented in the dark) and under normal ambient lighting conditions. Analysis of kinematic parameters indicated that the removal of binocular information did not significantly affect many of the major indices of the transport component, including peak wrist velocity. However, peak grip apertures increased and subjects spent more time in the final slow phase of movement, prior to grasping the object, during monocularly guided reaches. The dissociation between effects of binocular versus monocular viewing on transport and grasp parameters was observed irrespective of the presence of flanking objects. These results therefore further question the view that binocular vision is pre-eminent in the control of natural prehensile movements.  相似文献

Calcium/calmodulin dependent kinase II (CaMKII), PDZ-domain scaffolding protein Discs-large (DLG), immunoglobin superfamily cell adhesion molecule Fasciclin 2 (FAS2) and the position specific (PS) integrin receptors, including betaPS and its alpha partners (alphaPS1, alphaPS2, alphaPS3/alphaVolado), are all known to regulate the postembryonic development of synaptic terminal arborization at the Drosophila neuromuscular junction (NMJ). Recent work has shown that DLG and FAS2 function together to modulate activity-dependent synaptic development and that this role is regulated by activation of CaMKII. We show that PS integrins function upstream of CaMKII in the development of synaptic architecture at the NMJ. betaPS integrin physically associates with the synaptic complex anchored by the DLG scaffolding protein, which contains CaMKII and FAS2. We demonstrate an alteration of the FAS2 molecular cascade in integrin regulatory mutants, as a result of CaMKII/integrin interactions. Regulatory betaPS integrin mutations increase the expression and synaptic localization of FAS2. Synaptic structural defects in betaPS integrin mutants are rescued by transgenic overexpression of CaMKII (proximal in pathway) or genetic reduction of FAS2 (distal in pathway). These studies demonstrate that betaPS integrins act through CaMKII activation to control the localization of synaptic proteins involved in the development of NMJ synaptic morphology.  相似文献

This study uses 61 years of death certificates for Bexar County, Texas, uniformly coded under ICD 9, to describe the transition in heart disease mortality from 1935-1995. We find that life expectancy for persons dying with heart diseases increased throughout this period, with clear differences in rates of increase for males and females, associated with acute ischemic heart disease. Our data point to an epidemic of AIHD in the 1950s and 1960s, which is now abating. Findings are less clear for chronic ischemic heart disease, while other major heart diseases cannot be traced with any confidence owing to changes over time in the emphasis accorded particular causes. Our findings suggest caution with respect to the socioeconomic analysis of heart disease mortality data, particularly where the instability of the coding conventions has been most acute.  相似文献

The numbat (Myrmecobius fasciatus) is a diurnal and exclusively termitivorous marsupial. This study examines interrelationships between diet, metabolic rate and water turnover for wild, free-living numbats. The numbats (488±20.8 g) remained in mass balance during the study. Their basal metabolic rate (BMR) was 3.6 l CO2 day–1, while their field metabolic rate (FMR) was 10.8±1.22 l CO2 day–1 (269±30.5 kJ day–1). The ratio FMR/BMR was 3±0.3 for numbats. We suggest that the most accurate way to predict the FMR of marsupials is from the regression log FMR=0.852 log BMR+0.767; (r2=0.97). The FMR of the numbat was lower than, but not significantly different from, that of a generalised marsupial, both before (76%) and after (62–69%) correction for the significant effect of phylogeny on FMR. However the numbat's FMR is more comparable with that of other arid-habitat Australia marsupials (98–135%), for which the regression relating mass and FMR is significantly lower than for nonarid-habitat marsupials, independent of phylogeny. The field water turnover rate (FWTR) of free-living numbats (84.1 ml H2O day–1) was highly correlated with FMR, and was typical (89–98%) of that for an arid-habitat marsupial after phylogenetic correction. The higher than expected water economy index for the numbat (FWTR/FMR=0.3±0.03) suggests that either the numbats were drinking during the study, the water content of their diet was high, or the digestibility of their termite diet was low. Temperature-scan X-ray scattering was used to study the effect of the fusion peptide of simian immunodeficiency virus (SIV) on the lipid polymorphism of N-methylated dioleoylphosphatidylethanolamine (DOPE-Me), in the presence and absence of one or both of the fusion inhibitors carbobenzoxy-D-phenylalanine-L-phenylalanine-glycine and 1-lauroyl-2-hydroxy-sn-glycero-3-phosphocholine (LPC). Using X-ray diffraction at stations 2.1 and 8.2 of the Synchrotron Radiation Source at Daresbury Laboratory, UK, the structure of multilamellar vesicles (MLVs) was probed as the temperature was raised from 20 to 90 degrees C. The results are compared to those of similar studies, reported earlier, that used the fusion peptide of feline leukaemia virus (FeLV) which, at 28 amino acid residues in length, is considerably longer than the SIV peptide (12 amino acid residues). We interpret the results within the framework of current understanding of membrane fusion, and demonstrate how observed lipid polymorphism might describe the fusion process.  相似文献

Therapeutic pegylated interferon-alphas (IFN-alpha) are mixtures of positional isomers that have been monopegylated at specific sites on the core IFN-alpha molecule. The pegylation results in lower in vitro specific activity associated with the core IFN-alpha molecule that is related to the site of pegylation and size of polyethylene glycol (PEG) attached. We prepared purified, homogeneous, positional pegylation isomers of IFN-alpha2b that were monopegylated using 5-30-kDa linear PEG molecules attached at 7 primary reactive amino acid residues: Cys(1), His(34), Lys(31), Lys(83), Lys(121), Lys(131), and Lys(134). The isomers were evaluated for STAT translocation and antiviral and antiproliferative activity. The site of pegylation strongly influenced activity relative to an IFN-alpha2b control. The highest residual activity was observed with the His(34) positional isomers, and the lowest was observed with the Cys(1) positional isomers. The Lys positional isomers demonstrated intermediate activity, with a general order of Lys(134) > Lys(83) approximately Lys(131) approximately Lys(121) > Lys(31). The progressive relationship between decreased activity and increased PEG size suggests that pegylation may interfere with interaction and binding of IFN-alpha to the IFNAR1-IFNAR2 heterodimeric receptor. The higher specific activity associated with the His(34) positional isomer suggests that this site may be favorable for pegylating IFN-alpha2b molecules.  相似文献

Potato Cyst Nematodes (PCN) currently represent a serious threat to potato cultivation. However, many sources of resistance are known amongst primitive and wild relatives of cultivated potato, Solanum tuberosum ssp. tuberosum. Currently, in the UK, the major threat is due to Globodera pallida, resistance to which has not yet been effectively deployed in potato cultivars. We have performed linkage and QTL analysis of a tetraploid potato population segregating for the H3 source of resistance to G. pallida that was introgressed into cultivated potato from the primitive species, Solanum tuberosum ssp. andigena. This source is highly effective against the most common UK pathotype of G. pallida (Pa2/3) and its deployment in breeding material is a major goal. We adopted an approach involving bulked segregant analysis (BSA) as well as genome wide linkage analysis using AFLP and SSR markers. BSA provided a concentration of markers linked in coupling to a QTL on linkage group IV, and improved the accuracy of the QTL localisation. By performing an analysis on residual scores after removal of the effects due to the major QTL, we detected a second QTL on linkage group XI.  相似文献
