Methylmethacrylate (MMA) is the most commonly used embedding medium for sectioning undecalcified bone; however, a number of problems exist with its use in a research laboratory. MMA requires a long infiltration time and temperature control, and it reacts with many polymers. We used Kleer Set resin™ as an alternative embedding medium for sectioning undecalcified bone specimens. Fluorochrome labeled bone specimens were sectioned transversely using a ground section technique and longitudinally on a sledge macrotome. The slides were viewed using both transmitted light and epifluorescence microscopy. High quality sections were obtained using Kleer Set resin™ for both sectioning techniques. We have shown that this new embedding medium is simpler, safer, quicker to use and does not interfere with visualization of fluorochromes.  相似文献

This review examines some of the models to account for time measurement in insect photoperiodism. It considers the supporting evidence for these models and the attempts to discriminate among them. Although hourglass timers may exist, it is suggested that most photoperiodic mechanisms, including many hourglass‐like timers, are circadian‐based, making Bünning's original hypothesis, that the circadian system somehow provides the essential “clockwork” for photoperiodic timing, the most persuasive unifying principle. The apparent diversity among modern species in their modes of time measurement is probably the result of differences between the underlying circadian systems that were adopted for seasonal night length measurement as the insects, or groups of insects, moved northwards into areas with a pronounced winter season. Photoperiodic time measurement, therefore, exhibits both unity (in their common circadian basis) and diversity in detail. Attention to this diversity may provide invaluable insights into the problem of photoperiodic time measurement at comparative, and molecular, levels.  相似文献

Waddington's epigenetic landscape illustrates such characteristic features of development as homeorhesis and the existence of alternative developmental pathways. Simply recongnizing that these are typical allows us to make inferences about evolution, for example that macroevolution is often a different process from microevolution. We can account for the origin of these properties by assuming that many processes in development can be modelled by non-linear differential equatiions. The assumption then leads to two further predictions: that multiple speciation may be relatively common and that phenocopying is likely to occur in one direction only.  相似文献

The tolerance of spring and winter cultivars of wheat, barley and oats to infection by barley yellow dwarf virus (BYDV) was examined in field tests. Sererely affected plants were stunted and grain yields were considerably reduced. These changes in infected spring barley cultivars were associated with decreases in both ear length and number and sizes of grain. Most barley and oat cultivars were more susceptible than wheat cultivars. Yield losses decreased with successively later dates of inocultaiton, the yield response being characteristic of the tolerance of individual cultivars. Differences between tolerant and susceptible cultivars were insignificant in most cases where infection occurred after growth stage 7. Individual seedling symptoms assessed in April and May were more closely related to each other and to yield losses than those recorded in March and June. The incorporation of seedling symptoms into indices of infection permits estimates of decrease in yield to be made by applying the formulae, DY = (LC+LA+SH)/3 to barley cultivars and DY = 1.7 × (LC+LA+SH)/3 - 7 to oat cultivars. Thus decreases in grain yield (DY) can be related to decreases in seedling height (SH) and increases in percentage of leaf area discoloured (LA) and intensity of discoloration (LC). Plot symptoms of infection, scored on a 0–7 scale, offered a more convenient method of assessment than individual seedling symptoms. Plot scores (PA) were closely correlated with other seedling symptoms and with decreases in grain yield (DY), and can thus be used to estimate yield losses by substituting into the formula, DY = 14PA (April)-8. This formula was found to be applicable to all cereal cultivars examined in N.I.A.B. trials, there being no distinction between wheat, barley and oats.  相似文献

ABSTRACT. The blowfly, Calliphora vicina Meigen (Diptera: Calliphoridae), displays a maternally induced larval diapause. Progeny of adults exposed to short days enter diapause if the larval temperature is 15C or below; exposing adults to long days, or larvae to temperatures above 15C, results in non-diapause. By keeping progeny from short-day parents at the parental temperature of 23.5C for various lengths of time before transfer to 11C, it could be shown that the diapause-averting effect of high temperature operates in the late wandering stage of larva, possibly at the moment of the decision for the brain-ring gland complex to release PTTH/ecdysone at the diapause/non-diapause stage.
Although photoperiodic induction in the blowfly is maternal, previous work had shown that the larvae were also slightly sensitive to photoperiod. From the present study it is evident that also the eggs are photo-sensitive.  相似文献

ABSTRACT. The sensitive period for the induction of pupal diapause in Sarcophaga argyrostoma (R.-D.) (Diptera, Sarcophagidae) extends from the intra-uterine embryos to puparium formation, although the degree of 'sensitivity' declines during that period. Embryos and larvae 'add up' both long and short nights to a threshold. The incidence of diapause may also be influenced by the photoperiodic manipulation of female flies containing ovarian eggs. High temperatures eliminate or override the diapause response, whilst lower temperatures enhance it. Still lower temperatures (10–12°C) bring about a high larval and pupal mortality and a failure to pupate which is more marked in long than in short nights. The results are discussed in terms of the external coincidence model and the concept of the 'photoperiodic counter'.  相似文献

ABSTRACT. The photoperiodic mechanism governing larval diapause in the blowfly Calliphora vicina R.-D. (Diptera: Calliphoridac) is largely maternal. The critical daylength for adult flies at 20C, and larvae derived from day 12 eggs and maintained in darkness at 11C, is about 14.5 h/24. At short daylength (LD 12:12 h) and a range of temperatures, eggs laid on or before day 9 gave rise to a low incidence of diapause, whereas those laid on day 10 and later gave rise to a high incidence; the photoperiodic 'counter' therefore shows a temperature-compensated mechanism for 'accumulating' successive short days. The rate of switching from non-diapause to diapause progeny is hardly affected by the timing of protein meals, although an early supply of meat accelerates egg development and leads to an increasing proportion of early, non-diapausing progeny. There is no evidence for a maternal effect preventing diapause induction in the first post-diapause ('spring') generation of flies. Diapausing larvae terminate diapause spontaneously in darkness at 11C after a variable period which is positively correlated with the incidence of diapause in the group. Diapause incidence and duration therefore are both determined maternally.  相似文献
